1.Effect of Metal Ions on Cell Growth and Phenol Degradation of a Thermophilic Strain BF80

The effect on the growth and phenol degradation of the thermophilic strain BF80 by seventeen different metal ions were studied. The results showed that the metal ions certainly affected the growth and phenol degradation of the strain BF80. At the concentration of 0.01%,contrasting to the comparison,the growth and phenol degradation of the strain BF80 were restrained intensively by the metal ions of Cu2+,Zn2+,Co2+,Ba2+,Hg2+,Ni2+,Ag+,Al3+. The metal ions of Cr2+ restrained the phenol degradation of BF80 strongly while Cr2+ restrained the growth of BF80 faintly. The metal ions of Sn2+,Fe2+,Fe3+,Pn2+ restrained the growth and phenol degradation of the strain BF80 at a certain concentration,and the effect of inhibition in-creased with the increase of the concentrations of the metal ions. At the low concentration of Mn2+ or Mo2+,the growth and phenol degradation of the strain BF80 were increased,but if the concentration beyond the 0.1%,the growth and phenol degradation of the strain BF80 were inhibited. At the different concentration of Ca2+ or Mg2+,the growth of the strain BF80 were increased and the phenol degradation were accelerated,but the max rate of phenol degradation by BF80 was not influenced obviously. When the medium was added themixture of metal ions of Mo2+ and Mn2+,the strain BF80 grew better,but the rate of the phenol degradation was lower than that of single Mo2+ and Mn2+.
2.Volatile constituents in the flowers of Elsholtzia argyi and their variation: a possible utilization of plant resources after phytoremediation.

Phytoremediation effectiveness and remediation costs are driving factors of this project. Full utilization of plant resources after their being used for phytoremediation is an unsolved problem. GC/MS technique was used to investigate the volatiles of the flowers from Elsholtzia argyi (PFE1: Purple Flower Elsholtzia) and their variation (WFE: White Flower Elsholtzia), naturally growing in Pb/Zn mined area, and Elsholtzia argyi (PFE2: Purple Flower Elsholtzia), naturally growing in Jiuxi uncontaminated agriculture soil. Seventeen compounds constituting 86.88% of total essential oils were identified in PFE1, with 2,6-octadienoic acid,3,7-dimethyl-methyl ester being the main constituent (63.30%). Sixteen compounds accounting for 95.32% of total essential oils were identified in WFE, with caryophyllene being the main component (55.02%). Compared to PFE1, PFE2 contains lower level of 2,6-octadienoic acid,3,7-dimethyl-methyl ester (31.76%), which is the main constituent in the total essential oils of PFE2. Caryophyllene is the main ingredient of flavor. Elsholtzia ketone was identified in all the three Elsholtzia plants. It can be concluded that the selected Elsholtzia argyi plants can be exploited on their versatile uses as fragrances and antiseptics due to the perfume ingredient and antibacterial components existing in their essential oils.

3.Disease Prevention Effect of Improvement of Latrines in Rural Areas in Hunan Province

Objective To investigate the effect of non-hazard treatment of feces on control of intestinal infectious diseases and parasitosis. Methods During Nov, 2003-May, 2004, we selected 6 counties as observational points by the character of geographical position and the types of non-hazard treatment in Hunan province. The number of fecal coliform and the sedimentation rate of parasite ova were measured, the methods of questionnaire and retrospective study were used and the prevalance rate of intestinal infectious diseases, the incident rate of infectious diarrhea and infectious rate of parasite were analyzed. Results Among three types of non-hazard treatment, the eligibility rates of fecal coliform were 95.0%, 93.5%and 91.7% respectively,and the sedimentation rates of parasite ova were 90.0%, 90.3% and 87.5% respectively. Compared with the villages where the latrines had not been improved, the infectious rates of parasite decreased by 67.0%,the prevalance rate of intestinal infectious diseases decreased by 58.3%, and the incidence rates of infectious diarrhea decreased by 56.3% in villages where the latrines had been improved. Conclusion Non-hazard treatment of feces will play an important role in preventing of intestinal infectious diseases, infectious diarrhea and parasitosis.
4.Cultural Conditions for Production of Glutathione by Mutant Saccharomyces J-X25

The production conditions of glutathione with shaking flask fermentation by the mutant Saccharomyces J-X25, a methionine-defected strain were studied, and the optimum culture conditions are as follows: initial pH6. 0, temperature 30℃, 100ml in 500ml flask, the inoculum size 10% and agitation rate 220r/min. The emphasis was on the stimulating effect on the cells by dioxogen and the sodium lactate as surfactant. Both of which were added at the logarithmic phase of fermentation, and the GSH production was up to 0. 253g/L , 52% higher than the control without the additions. Compared with the production of GSH initial strain that by the mutant in optimum conditions was increased by 79%.
7.Gama-aminobutyric acid accumulation in Elsholtzia splendens in response to copper toxicity.

A solution with different Cu supply levels was cultured to investigate gama-aminobutyric acid (GABA) accumulation in Elsholtzia splendens, a native Chinese Cu-tolerant and accumulating plant species. Increasing Cu from 0.25 to 500 micromol/L significantly enhanced levels of GABA and histidine (His), but considerably decreased levels of aspartate (Asp) and glutamate (Glu) in the leaves. The leaf Asp level negatively correlated with leaf Cu level, while leaf GABA level positively correlated with leaf Cu level. The leaf Glu level negatively correlated with leaf GABA level in Elsholtzia splendens. The depletion of leaf Glu may be related to the enhanced synthesis of leaf GABA under Cu stress.

8.Clinical research of protein-free calf blood extract eye gel for corneal epithelial defect

AlM: To evaluate the clinical effect of 200g/L protein-free calf blood extract eye gel for corneal epithelial defect.METHODS: One hundred and sixty - eight cases of corneal epithelial defect ( 58 cases with herpes simplex keratitis; 24 cases with chemical injury; 85 cases with pterygium operation injury ) were randomly divided into two groups: 84 eyes were treated with protein-free calf blood extract eye gel; 84 cases were treated with basic fibroblast growth factor eye gel ( bFGF ) . The bFGF and protein-free calf blood extract eye gels were used 4 times a day. The treatment course was 7d. Epithelial defect restoration, local symptom and sign were observed.
RESULTS: The difference between pre-treatment and post-treatment was significant ( P<0. 01 ) in the two groups. The effect of protein-free calf blood extract eye gel (83. 3%) group was prior to that of bFGF (69%) for corneal epithelial defect. The effective rate of protein-free calf blood extract eye gel in the herpes simplex keratitis, chemical injury and pterygium operation injury was 72. 4%,69. 2% and 95. 2%. Localized stimulus and adverse reaction of all over the body were not been observed.
CONCLUSlON: Protein-free calf blood extract eye gels is valuable and safe for corneal epithelial defect.
9.Regulation mechanism of autophagy-related protein LC3 by c-Jun in methotrexate resistant human choriocarcinoma JEG-3 cells

Objective To explore the regulation mechanism of autophagy-related protein, microtubule-associated protein 1 light chain 3 (LC3), via c-Jun in methotrexate resistant human choriocarcinoma JEG-3 cell lines. Methods Human choriocarcinoma JEG-3 cell lines, and methotrexate resistant choriocarcinoma JEG-3 (JEG-3/MTXR) cell lines were used in our present study. Phosphorylation c-Jun (p-c-Jun) was evaluated after exposure to 0.02 ng/ml methotrexate for 72 hours in both cells by western blot. c-Jun gene was knockdown by small interference RNA (siRNA) in JEG-3/MTXR cells, and LC3 was evaluated by western blot and reverse transcription-PCR. The binding of LC3 promoter with c-Jun protein was detected via chromatin immunoprecipitation assay (ChIP) with or without 0.02 ng/ml methotrexate exposure. Results The results showed that p-c-Jun was up-regulated after methotrexate treatment for 72 hours (1.99±0.20, versus 0.20±0.06 at 0 hour;P<0.05) by western blot analysis in JEG-3/MTXR cell lines. Further investigation demonstrated that c-Jun-siRNA could inhibit the up-regulation of LC3 formation and after methotrexate exposure (LC3 mRNA:1.24±0.17 versus 3.03±0.43;LC3 protein:0.52±0.07 verus 1.20± 0.15; all P<0.05). The binding of LC3 promoter by c-Jun protein was up-regulated after methotrexate treatment by the method of ChIP in methotrexate resistant JEG-3/MTXR cells [(2.95 ± 0.35) times]. Conclusion Autophagy-related gene LC3 expression regulated by c-Jun protein may be involved in the effect mechanism of the development of methotrexate resistance in choriocarcinoma JEG-3 cells.
10.Clinical Study on Acupuncture in Treating Diarrhea-predominant Irritable Bowel Syndrome

Objective: To observe the changes of 5-hydroxytryptamine (5-HT) in colonic membrane of diarrhea-predominant irritable bowel syndrome (IBS-D) and the clinical efficacy of herbal cake-partitioned moxibustion in treating it, and to explore the action mechanisms. Methods: Seventy-three subjects were randomized into two groups: moxibustion group in which 37 cases were treated by herbal cake-partitioned moxibustion and needling group in which 36 cases were treated by needling. The clinical efficacy and 5-HT expression in colonic membrane were observed. Results: The cure rate was 43.2% in the moxibustion group and 36.1% in the needling group, with no significant difference between the two groups (P>0.05). Herbal cake-partitioned moxibustion could reduce 5-HT expression in colonic membrane. Conclusion: Herbal cake-partitioned moxibustion might improve the clinical symptoms of IBS-D by reducing 5-HT expression.
