1.Effect of Progesterone on Neuronal Apoptosis and Nitric Oxide Level in Cortex and Hippocampus of Newborn Rats with Hypoxic-Ischemic Encephalopathy

Objective To explore the effect of progesterone on the rate of neuronal apoptosis and nitric oxide(NO) level in the cortex and hippocampus tissue of newborn rats with hypoxic-ischemic encephalopathy(HIE).Methods Thirty 7-day-old neonatal rats were randomly divided into 3 groups:sham-operated group,hypoxic-ischemic(HI) group and pretreatment group.Rats in HI group and pretreatment group were subjected to left common carotid artery ligation,then were exposed to 80 mL/L oxygen and 920 mL/L nitrogen gas in 37 ℃ closed container for up to 2.5 h to establish HIE model.Progesterone was injected intraperitoneally into rats in the pretreatment group respectively for 30 minutes before hypoxia,and solution was injected into the sham-operated group and HI group.All rats were killed at 24 h after operation.The neuron apoptosis was identified and analyzed by flow cytometry.Nitrate/nitrite was assayed to represent nitric oxide content of brain tissues.Results The ratio of neuronal apoptosis and NO contents in cortex and hippocampus tissue in HI group [(10.09?0.36)%,(12.32?0.28)%,(51.36?9.71) ?mol/L,(52.34?4.26) ?mol/L] were significantly higher than those in sham-operated group [(2.49?0.23)%,(2.58?0.26)%,(18.16?6.24) ?mol/L,(19.28?3.58) ?mol/L)](P_a
2.The Effects of a Joint Treatment on Menopausal Patients with Anxiety and Depression

Objective: To compare the effects of HRT joint Fluoxetine with HRT only in treating menopausal patients with anxiety/depressive symptom. Method: 66 Menopausal Patients who met the criteria of depression of ICD-10 co-morbid with anxiety were evaluated with HAMD, HAMA and TESS in terms of effects and side effects. All subjects were divided into three groups. The first group included 23 patients treated with HRT only, the second group included 23 patients treated with HRT joint Fluoxetine, and the third group included 20 patients treated with HRT joint Fluoxetine and psychotherapy. Results:All of the three groups showed significant improvement after 6 weeks treatment in terms of HAMD scores. 4.Investigation of Agars Culture Mediums Sterilized by Ethylene Epoxide

Objective To develop a new method for sterilization in agars culture mediums,i.e.Agars culture mediums is sealed to be sterilized and preserved after resolution.Methods The effect of rudimental Ethylene Epoxide was investigated,and the germiculture results by different preserving methods were analyzed.Air sedimentation bacterium was sampled by different methods to analyze the bacterium capture rate.Results Ethylene Epoxide sterilized agars culture mediums bacterium capture rate was similar with regular agars.Conclusion Ethylene Epoxide sterilized agars culture mediums can be used in sampling of clinical treatment and other outburst incident,reducing the preparation time.

6.Advances in photodynamic therapy based on tumor hypoxia

Photodynamic therapy (PDT) has attracted wide attention due to its unique advantages such as minimal invasiveness, high efficiency and high selectivity, and its ability to induce anti-tumor immune response. However, the treatment process is heavily dependent on the oxygen content of the treatment site, and the widespread oxygen deficiency in malignant tumors severely limits its efficacy. In addition, PDT-mediated oxygen depletion exacerbates tumor hypoxia, which further reduces its therapeutic effect. In recent years, many researches have been devoted to overcoming this problem. This paper summarized various strategies based on tumor hypoxic PDT in recent years, discussing the advantages and disadvantages of these strategies, and analyzing the main challenges and future directions of PDT in the treatment of tumors, so as to provide references for the in-depth study of photodynamic therapy of tumors.
7.Study of phonological encoding of Chinese disyllabic compound words in patients with mild cognitive impairment

ObjectiveTo explore the characteristics of phonological encoding of Chinese disyllabic compound words in patients with mild cognition impairment(MCI). Methods10 patients with MCI and 10 normal controls who matched with the age, gender, level of education and handedness performed homophone judgment task. In this task, a picture with a disyllabic compound name was presented, after 100 ms of the presentation there would be a Chinese character below the picture. Subjects were asked to judge whether one of the morpheme in the picture name was homophonic to the character. The reaction time and error ratio were analyzed statistically. ResultsIn the normal controls, the reaction time for the second constituents was shorter than that for the first constituents, while there were no difference between the first and second constituents reaction time in MCI patients. The error ratio of the first and second constituents showed no difference in normal controls, but MCI patients made more error rate for the first constituents. ConclusionAs the heathy elderly,the MCI patients have same pattern in phonological coding of Chinese disyllabic compound words, but much slower in activation speed, indicating the impairment of semantic memory and phonological activation in patients with MCI.
8.Expression of caspase-3 in the rat brain following pentylenetetrazole-induced epilepsy

ObjectiveTo investigate the expression of caspase-3 in the brain of acutely pentylenetetrazole(PTZ)-kindled rats.MethodsThe caspase-3 positive cells were revealed using immunohistochemical SP method. CMIAS image analysis system was used to analyse the expression of caspase-3 hemi-quantitatively. ResultsFollowing PTZ induced epilepsy, the expression of caspase-3 increased both in the hippocampus and in the cortex, and that was more remarkable in the hippocampus than in the cortex.ConclusionCaspase-3 may be activated during neuronal apoptosis after epilepsy. Hippocampus is more sensitive to the neuronal damage due to epilepsy than the cortex is.
9.Establishment of real time PCR for detecting plasma cell free DNA of rats and its significance.

OBJECTIVEOvertraining is a serious problem in sports, assessed by comprehensive multi-index evaluation, but so far there is still no sensitive, specific monitoring indicator or simple evaluation method to evaluate it. This research established a method for detecting plasma cell free DNA (cfDNA) of rats by real time PCR and discuss edits significance: a new molecular marker of overtraining?
METHODSTwelve male SD rats were randomly divided into control group and overtraining group. The overtraining group rats were undertaken overtraining on a motor-driven treadmill for 5 weeks, while the control group rats kept quiescent. All the rats were drawn blood at pre-and after-5 weeks to detect plasma levels of cfDNA, testosterone (T) and corticosterone (Cort) as well as peroxidation/antioxidation parameters (T-AOC, MDA, SOD, GSH-Px) and creatin kinase (CK).
RESULTS(1) Plasma cfDNA of rat was detected specifically by our real time PCR. (2) Compared with control group rats, the plasma cfDNA of overtraining rats increased obviously (about 5.43 fold). (3) Plasma cfDNA was related to plasma T, Cort, T/C ratio and MDA (correlation coefficent were -0.729, 0.854, -0.655 and 0.720, respectively) rather than plasma T-AOC, GSH-Px, SOD and CK.
CONCLUSION(1) A real time PCR method was established successfully to determine plasma cfDNA of rat. (2) A remarkable raises of plasma levels of cfDNA were found in overtraining rats which were associated with T, Cort and T/C, suggested that plasma cfDNA might be a new molecular marker of overtraining. (3) The increase of plasma cfDNA of overtraining rat might correlate with enhanced oxidative stress induced by overtraining instead of muscle damage.

10.Preliminary application and discussion of independent 3D dose calculation in intensity-modulated radiotherapy for cervical cancer

Objective To assess the feasibility for the automated treatment planning verification system Mobius3D (M3D) to perform an independent 3D dose calculation in intensity-modulated radiotherapy (IMRT) for cervical cancer.Methods Twenty patients with cervical cancer were randomly selected.With treatment planning systems (Pinnacle,Version 9.2;Eclipse,Version 13.5),all IMRT plans were divided into 7 fields to meet the dosimetric goals.The optimized plans were exported to the M3D server.The percentage differences in the volume of region of interest (ROI) and the dose calculation of target volume and organ at risk (OAR) were evaluated for the two treatment planning systems,and theγ passing rate was used to assess the accuracy of M3D calculation.Results The difference in the volume of ROI for Pinnacle 9.2 to M3D was less than that for Eclipse 13.5 to M3D,with maximum differences of 0.22%±0.69% and 3.5%±1.89% for Pinnacle 9.2 and Eclipse 13.5,respectively.The differences in the dose calculation of target volume and OAR for the two treatment planning systems to M3D were within ± 1%.After recalculating by M3D,the dose difference between Pinnacle 9.2 and M3D was smaller than that between Eclipse 13.5 and M3D,but the mean differences were all within ±3%.The γ passing rates for target volume and OAR were more than 95% on average.Conclusions The method of utilizing the automated treatment planning verification system to validate the accuracy of plans is convenient.It can be used as a secondary check tool to improve accuracy in IMRT dose calculation.
