1.IMPROVEMENT OF NIGERICIN PRODUCTION IN STREPTOMYCES HYGROSCOPICUS NND-52 BY COMBINATION MUTANT

Based on the lethal rate, positive and negative mutation rate, the optimal doses of UV, HNO2, UV + AO treating S. hygroscopicus NND-52-29 were determined. The mutant strain A19 that was obtained showed the derepres-sive property to glucose and the nigericin yield of A19 could be increased by 128%. When it was subcultured combined with natural screening, its stability was rather satisfied.
2.Effects of obstructive sleep apneas on endothelial function and autonomic modulation in adult man.

Objective To study the effects of obstructive sleep apneas on endothelial function and autonomic modulation. Methods From June 2009 to June 2011, male patients with obstructive sleep apnea hypopnea syndrome (OSAHS) were consecutively enrolled in this study. Patients with an apnea/hypopnea index (AHI) of greater than 15 and without previous treatment for OSAHS were included as Group OSAHS and obese subjects with an AHI of less than 5 were included as non-OSAHS controls (Group Control). Electrocardiography and beat-to-beat blood pressure were continuously recorded from the radial artery by applanation tonometry which was synchronized with polysomnography recording. Endothelial function was measured by arterial augmentation index (AAI). Spectral analysis of heart rate variability (HRV) and blood pressure variability (BPV) were computed for cardiac parasympathetic modulation (high frequency power, HF); sympathetic modulation (low frequency power, LF), sympathovagal balance (LF/HF power of R-R variability, LF/HF) and BPV sympathetic modulation (BPV LF) in normalized units [total power of the components/(total power-very LF power)×100]. Results Finally, 27 moderate-severe OSAHS patients and 22 non-OSAHS obese controls were recruited in the Group OSAHS and Group Control, respectively. In Group OSAHS, the age was 43.3±9.3 year-old, body mass index (BMI) was 36.8±8.7 kg/m2; in Group Control, the age was 42.9±8.6 year-old, BMI was 34.4±7.9 kg/m2; there were no significant differences in age and BMI between the Group OSAHS and Group Control (all P>0.05). The baseline AAI (12.5%±2.2% vs. 8.2%±2.1%) and BPV LF (68.3%±13.5% vs. 61.1%±11.7%) of the Group OSAHS were significantly higher than those of the Group Control (all P<0.05). And after overnight sleep, systolic BP (143.7±14.2 vs. 132.8±13.3 mm Hg), diastolic BP (87.7±7.7 vs. 78.6±5.5 mm Hg), HRV LF (69.7%±14.4% vs. 64.3%±12.1%), HRV LF/HF (3.7±2.0 vs. 2.3±1.3) and BPV LF (77.8%±15.6% vs. 68.3%±13.5%) of the Group OSAHS were significantly increased (all P<0.001), while HRV HF was significantly decreased (21.1%±9.3% vs. 27.5%±10.3%, P<0.05) from baseline.

4.Trans-obturator Tension Free Vaginal Tape for Stress Urinary Incontinence in Women

Objective To evaluate the efficacy of trans-obturator tension free vaginal tape(TVT-O)for the treatment of stress urinary incontinence(SUI)in women.Methods From January 2006 to September 2007,15 female patients with SUI received TVT-O in our hospital.A 1-cm longitudinal incision was made along the anterior wall of the vagina for the dissection of the urethra from the vaginal submucosal space to the obturator membrane.Then,a butteryfly-shape guide pole was inserted to guide the puncture of a spiral needle from the obturator membrane to the skin covering the root of the thigh.Afterwards,a trans-obturator tape was placed underneath the urethra without tension.Results The operation time was 15-40 min(mean,30 min),and blood loss was 10-40 ml(mean,20 ml).No intraoperative complication occurred in the series.One patient developed urinary retention,and was relieved by indwelling urethral catheter.Ten patients complained pain at the root of the thigh,and was cured simultaneously 3 to 10 days postoperation.Fifteen of the patients were followed up for 2 to 19 months with a mean of 8 months,during which no recurrence was found.Conclusion TVT-O is a simple,effective and reliable method for SUI.

7.Anti-tumor Effect of Zhongjiefeng Injection and Its Combination with Adriamycin

Objective To study the anti-tumor effect of Zhongjiefeng Injection in vitro and its combination with adriamycin.Methods MTT assay was adopted to determine the in-vitro anti-tumor effect,IC50 was used to measure the direct anti-tumor effect,and Jing' s formula was applied to analyze the combination of the drugs.Results Zhongjiefeng Injection can inhibit the proliferation of Bel 7402(human hepatoma cells),HCT-8(human colon cancer cells),and the IC50 was 33.13 mg/mL for Bel 7402 and 52.39 mg/mL for HCT-8 respectively.Zhongjiefeng Injection at the concentrations of 3.125,6.25,12.5 mg/mL showed an efficancy potentiation action with doxorubicin on the inhibition of HCT-8 cells in vitro,and 25,50 mg/mL showed a synergic effect on HCT-8 .Conclusion Zhongjiefeng Injection has a certain in-vitro inhibitory effect on the growth of Bel 7402 and HCT-8,its combination with doxorubicin in vitro can produce synergic effects(a simple combined or enhanced effects)to HCT-8 cells,especially high concentrations of Zhongjiefeng Injection with doxorubicin.It is suggested that Zhongjiefeng Injection in the doubled dosage may have a better synergistic effect with doxorubicin in clinical treatment of colon cancer.
8.The Imaging Diagnosis of Medullary Sponge Kidney

Objective To explore the clinical value of imaging examinations of medullary sponge kidney with the literature reviewed.Methods The features of X-ray plain film (1 case),IVP (2 cases),B-mode US(3 cases) and CT images (9 cases) of medullary sponge kidney (MSK) in 11 cases were analysed retrospectively.Results X-ray plain film showed a cluster of round and oval high density images in renal papillae and renal medulla in one case one kidney.Collecting tubes were showed as like line,brush and sac like dilatation on IVP in 2 cases 3 kidneys.B-mode US revealed a cluster of high echo spots scattered in renal medulla in 3 cases 5 kidneys and sac like dilatation of collecting tubes in 2 cases 3 kidneys.CT images showed the stones of spongiform kidney scattered alone renal medulla in 9 cases 16 kidneys,while the stones covered by contrast media on enhanced CT images in 3 cases 5 kidneys,and enhanced stripes of high density projected from renal papillae to renal medulla.Conclusion X-ray plain film,IVP,B-made US and CT all can be used in diagnosing spongiform kidney.Plain film and IVP are yet the first selected methods and IVP is specific and direct in showing collecting tubes dilatation.B-mode US especially CT have a high showing rate of stone in spongiform kidney and can be helpful to evaluate the collecting tube dilatation.
9.Effects of ganoderma lucidum triterpenoids on learning and memory function of aging mice induced by D-galactose

Objective To observe effects of ganoderma lucidum triterpenoids(GLT) on learning and memory function and anti-oxidative ability of aging model mice induced by D-galactose. Methods Senile model mice were established by D-galactose hypodermic injection for 8 weeks.GLT had been administered to two therapy groups.All the mice of different groups were tested with Morris water maze.Then the mice were killed and biochemically assayed of total anti-oxidative capacity(T-AOC),superoxide dismutase(SOD) and malondialdehyde(MDA) in the brain. Results The model mice showed worse ability in learning and memory in comparison with control mice.The T-AOC activity and SOD activity in the brain decreased and the MDA content increased in model rats in comparison with control.GLT significantly improved the changes mentioned above. Conclusion GLT improved the learning and memory dysfunction in aging model mice by modulation of the anti-oxidative ability.
10.Sequential therapy of CD19 and CD22 chimeric antigen receptor T cells for relapsed/refractory mediastinal B lymphoblastic lymphoma: report of one case and review of literature

Objective:To investigate the efficacy of sequential therapy of CD19 and CD22 chimeric antigen receptor T cells (CAR-T) for relapsed/refractory mediastinal B lymphoblastic lymphoma (B-LBL).Methods:One patient with relapsed/refractory mediastinal B-LBL treated with sequential therapy of CD19 and CD22 CAR-T who was admitted to Tongji Hospital of Huazhong University of Science and Technology Tongji Medical College in March 2017 was reported. At 1, 3, 6, 12 and 18 months after CAR-T therapy, the indicators of primary disease remission were monitored and the relevant literature was reviewed.Results:The patient relapsed and progressed after third-line chemotherapy, and then received sequential therapy of CD19 and CD22 CAR-T. In the course of cellular immunotherapy, the patient presented grade 1 cytokine release syndrome. After active treatment, the patient got stable condition and was discharged. The patient came to the hospital for regular review, and the mediastinal mass of the patient was dynamically followed up. After CAR-T therapy, the mediastinal mass of the patient was significantly reduced, and the patient was in continuous remission for 18 months.Conclusions:Sequential therapy of CD19 and CD22 CAR-T provides a new therapeutic approach for relapsed/refractory B-LBL. For patients with poor curative effect of conventional chemotherapy, CAR-T therapy should be actively performed as soon as possible to improve the remission rate and the long-term prognosis of patients.
