1.Clinical Characteristics of Extrapulmonary Complication Induced by Mycoplasma Pneumoniae Infection Complicating with Infectious Mononucleosis Syndrome

Objective To approach the clinical characteristics of extrapulmonary complication induced by mycoplasma pneumoniae(MP) infection and infectious mononucleosis syndrome. Methods The MP-IgM(+), EBV- IgM(+) were determined by enzyme- linked immunosorbent assay.There were 18 cases infected with MP and infectous mononucleosis syndrome with abnormal fever,cervix lyphadenectasis,pharyngitis,liver and spleen intumescence,lung infection, and so on.They were analyzed by reviewing.Results There were 8 cases(44.4%) with extrapulmonary complication, among them 3 cases(16.6%) with liver injury,1 case (5.56%) with heart injury,2 cases(11.1%) with both heart and liver injuries,1 case(5.56%) with urinary system injury (nephritis), 1 case (5.56%) with nervous system injury (encephalitis).The complication usually arised 1 week after primary affection, but had no typical clinical symptoms.There was only 1 case with encephalitis who expressed the symptom of nervous system symptom firstly, and was clinically dia- gnosed by assistant examinations. All the cases recovered after 3 weeks′ treatment with macrolides, 5-7 days with ganciclovir, and also with regular complication and support treatments.Conclusions The incidence of extrapulmonary complication induced by MP infection and infectous mononucleosis syndrome is higher than that by the single infection, and its clinical symptom isn′t obvious and is easy to be covered by original illness. And a few showing the extrapulmonary as the first symptom is easily mistakenly diagnosed .As a result, the prognosis is favorable if we make subsidiary inspection and diagnosis as soon as possible and give it with regular treatment.

3.Preparation and release mechanism of gestodene reservoir-type intravaginal rings.

This study taking gestodene (GEST) as a model, investigated the factors affecting reservoir-type intravaginal ring (IVR)'s drug release. This paper reported a gestodene intravaginal ring of reservoir design, comprising a gestodene silicone elastomer core encased in a non-medicated silicone sheath, separately manufactured by reaction injection moulding at 80 degrees C and heating vulcanization at 130 degrees C is reported. The test investigated the factors affecting drug release through a single variable method, taking the drug release rates of 21 days as standards. When changing the thickness of the controlling sheath outside, the ratio of the first day of drug release and mean daily release (MDR), named the relatively burst effect, is closing to 1 with the thickness of controlling sheath increasing, while the 1.25 mm sheath corresponding to 1.04 controlled the burst release effectively; a positive correlation (r = 0.992 2) existed between the average drug release (Q/t) and drug loading (A) within a certain range. The C6-165 controlling sheath with high solubility of GEST is easier to achieve controlled release of the drug; GEST crystalline power is more effective to implement controlled release of drugs among difficent states of the drug. A 1/4 fractional segment core gives a relatively burst effect of 1.76, while the 1/1 and 1/2 are 1.93 and 1.87 separately, at the same drug loading, concluding that use of a fractional segment core would allow development of a suitable GEST reservoir IVR. In summary, GEST reservoir-type IVR could be adjusted by the thickness of controlling sheath, the loading of drug, the material properties of controlling sheath, the dispersion state of drug, the additive composition and structure of intravaginal ring, to control the drug release behavior and achieve the desired drug release rate.

5.Liver impairment in murine hepatitis virus 3 induced murine severe acute respiratory syndrome model

Objectives To investigate dynamic pathological features and virus distribution in the liver with a murine severe acute respiratory syndrome(SARS)model injected with murine hepati- tis virus 3(MHV-3)through trachea.As a representative of host genes,mouse fgl2(mfgl2)pro- thrombinase gene expression and its clinical significance were discussed in SARS associated liver dam- ages.Methods The Balb/cJ mice were infected with 100 PFU of MHV-3 through trachea and Balb/ cJ mice injected with saline were served as control.Survival rate,pathological features in organs and liver function were observed.Virus titers in different organs were determined on monolayer of L2 cells by a standard plaque assay.Virus distribution and cellular localization were studied by in situ hy- bridization.Both mfgl2 and fibrin expressions were examined in the liver by in situ hybridization and immunohistochemistry to investigate the role of mfgl2 in the liver impairment.Results Mice infected with MHV-3 through trachea developed multiple organs damages and died within 5 days,while all mice in control group survived with no histopathological changes.Infected liver tissues showed wide- spread cloudy swelling,prominent ballooning degeneration with mild lymphocytic infiltration in the portal area.Dot and zonal hepatocellular necrosis could be found occasionally.The lungs showed typi- cal interstitial pneumonia and hyaline membranes formation.Other histological changes also could be found in other organs examined.MHV-3 virus replication was identified in all organs observed.The liver function was injured,mfgl2 expression were evidenced mainly in the necrosis areas with fibrin deposition around the necrosis areas.Conclusions Pathological changes of the liver in this murine SARS model can mimic the liver impairment characteristics of SARS in human.In addition to the physical damage induced by the virus,the up-regulation of novel gene mfgl2 in the liver in association with fibrin deposition may play a vital role in the development of SARS associated liver damages.
6.Insulin receptor substrate expression and insulin resistance in intrauterine growth retarded rats

Insulin receptor substrate(IRS)-1 and IRS-2 expression levels of liver tissues and skeletal muscle in intrauterine growth retarded(IUGR)rats were investigated by RT-PCR and immunohistochemistry.An IUGR animal model was established by maternal nutrition restriction during pregnancy.IRS-2 expression level of liver tissue and IRS-1 expression level of skeletal muscle in IUGR rats at 0 and 3 weeks old were significantly lower than those in normal rats at the same age respectively,and insulin resistance was induced in IUGR,and these findings might be the molecular mechanisms susceptible to metabolic syndrome in IUGR rats.
7.Surveillance analysis of drinking water-born endemic fluorosis in 2009 in Xuchang city, Henan province

ObjectiveTo investigate the status of an illness of endemic fluorosis in Xuchang city,and to evaluate the effect of implementation of control measures.MethodsIn 2009,Changge city and Xuchang county were chosen as survey counties(city).We investigated the surveillance counties (city) about the running state of water-inproving projects,and fluoride content of drinking water in the villages,and ten villages were chosen as survey villages,dental fluorosis and fluoride content of children aged 8 to 12,clinical skeletal fluorosis and urinary fluoride content of 16-year-old adult were investigated.ResultsIn all the villages with projects of defluoridation for drinking water of the two monitoring counties,that the projects was being used accounted for 61.39%(97/158),and the projects discarded accounted for 38.61%(61/158).In the 9 non-water-improving villages,the average water fluoride ranged 1.32 - 2.25 mg/L; in the 1 water-improving village,the mean water fluoride was 0.44 mg/L.The total rate of dental fluorosis of children aged 8 to 12 was 38.65% (80/207),and the geometric mean of urinary fluoride was 1.65 mg/L.The rate of adult skeletal fluorosis by clinical detection was 0.30% (9/2982),and the geometric mean of urinary fluoride was 2.09 mg/L.ConclusionsXuchang city is a mild to moderate fluorosis epidemic areas of endemic fluorosis,and condition of the disease has not yet been effectively controlled,and there is need to further intensify the prevention and control efforts.
8.Influencing Factors and Optimizing Cultivating Conditions of Primary Cortical Neurons of Rat in Serum-Free Culture

Objective To explore the influencing factors for the purity and viability of primary cultured cortical neurons of rat,and optimize the separating and cultivating conditions of the cortical neurons.Methods The primary cotical neurons were cultured in a serum-free culture system of B27-supplemented neurobasal medium.The differences in purity and viability of primary cultured neurons between embr-yonic rat group and newborn rat [postnatal 24 h and 5 d] group were evaluated by morphology,immunocytochemistry of neuron-specific enolase(NSE) and trypan blue staining.The changes of neurons purity and viability in different trypsin digestion time(0 min,5 min and 15 min) at different environment temperatures(20 ℃ and 30 ℃) were assessed by immunocytochemistry and trypan blue staining.Results The primary cultured neurons from fetal and newborn rats grew well.There was no significant difference in embryonic rat and postnatal 1 d newborn rat group[(91.30?1.03)%,(89.50?1.78)% respectively in purity;and(98.20?0.58)%,(97.10?0.98)% respectively in viability].The neurons from 5-days newborn rat were inferior to that from fetal and 1-day newborn rat in purity and viability[(82.00?1.25)% and(92.87?1.56)% respectively].Shortening operation time and adjusting digestion time according to the environment temperatures could improve neuronal viability:A digestion for 15min at the environment temperature of 20 ℃ or for 5 min at 30 ℃ could acquired cells with higher viability [(98.20?0.58)% and(96.70?0.64)% respectively].Conclusions It is an easy,practical choice to culture priamry cortical neurons from postnatal 1 d newborn rats.Optimizing the separating and cultivating condition(environment temperatures,digest time,et al.) will improve the neurons purity and viability.
9.Influence Factors and Absorbing Mechanics of Copper Ion by Yeast Strain Y17

Using the high efficient copper-adsorbing yeast strain Y17 as absorbing material, the major affect factors including pH, original concentration of Cu2+, cell biomass, adsorption time and temperature were examined, and then the absorbing sites of the Y17 was determined. The results showed that the solution pH was the most dominate factor which affected the biosorption of Cu2+, the other affecting factors were the ini- tial concentration of Cu2+, the cell biomass added, and adsorption time, respectively; the temperature had lit- tle effect on the rate of biosorption. The orthogonal experiment showed that the optimal absorption condition was as follow: the solution pH was 5.0, the absorption time was 40 min, the cell biomass of Y17 added was 5.0 g/L, and the concentration of Cu2+ was 8 mmol/L; the highest adsorbing rate was up to 82.7% at this condition. Based on the results of different pretreatments and the desorption of Cu2+, the cell wall of Y17 was identified as the main place occurring boisorption process, and the -NH2 group, -COOH group on the surface of the yeast cells played an important role on the boisorption process.
10.Neonatal behavioral neurological assessment and developmental screening test for newborns with hypoxic ischemic encephalopathy

ObjectiveTo discuss the effect of neonatal behavioral neurological assessment (NBNA) and developmental screening test (DST) on newborns with hypoxic ischemic encephalopathy (HIE). MethodsNBNA was performed for 60 newborns with HIE within 12-14 days and DST was also performed at 6th month after birth.ResultsMild and severe degree HIE newborns with low NBNA scores had unfavorable prognosis. Conclusions NBNA and DST can provide evidence for early interference for newborn's HIE.
