1.Suitable construction of tissue-engineered adipose with silk fibroin porous scaffolds: A choice of optimal pore

BACKGROUND: In previous studies on tissue-engineered adipose, the pore size of scaffolds is easily neglected. If the pore size was large, cells would flow away along the pore and could not maintain in the scaffold. If the pore size was small, cells would distribute on the surface of the scaffold, and could not enter the scaffold. Simultaneously, it would be not contribute to growth of new vessels. OBJECTIVE: To screen appropriate silk fibroin porous scaffolds to construct tissue-engineered adipose. METHODS: Under unchanged concentration of flbroin, six kinds of silk fibroin porous scaffolds in different aperture were prepared by changing freezing and drying temperature and time. Human umbilical cord mesenchymal stem cells (hUCMSCs) were harvested using attachment method. The potentiality of its osteogenic and adipogenic differentiation was detected by chemistry staining. The apertures of six kinds of silk fibroin porous scaffolds were measured by scanning electron microscope. The adhesion and proliferation of hUCMSCs on silk fibroin porous scaffolds in different aperture were observed. RESULTS AND CONCLUSION: The apertures of six silk fibroin porous scaffolds were (39.94±17.27), (53.51±16.18), (63.97±19.76), (71.08±18.07), (87.33±21.78), (121.97±44.10) μm, respectively. The strong ability of attachment of hUCMSCs appeared in the number two scaffold, and in other materials (number 1,3,4,5,6), there were not any cell in them except number one and three. Therefore, 50 μm might be a suitable aperture to construct tissue-engineered adipose with silk flbroin porous scaffolds using hUCMSCs.
2.Cervical carcinoma HLA gene susceptibility and NK cell receptor immune re-sponses

Objective:To investigate the effect of HLA genetic susceptibility and NK cell receptors and immune response on the occurrence and development of the Cervical cancer.Methods: Select the 200 patients confirmed by the pathological biopsy in our hospital from January 2013 to January 2014 as the observation group.At the same time,randomly select the 200 healthy women as the control group.Both of them blood 2 ml peripheral blood,sample the cervical cell from the observation group.Having the cytological ob-servation and the DNA′s probe of the HPV,observe two group′s HPV infection rates and HLA′s parting.Results: The HPV infection rates of the observation group is 91%,and the rates of the control group is 16%.The differences between them were all significant(P<0.05).The HLA-KIR*1003,HLA-KIR*14,HLA-KIR*17,HLA-KIR*02,HLA-KIR*12 distribution frequency of the observation group are 41%,39%,35%,15%,53%.The HLA-KIR*1003,HLA-KIR*14,HLA-KIR*17,HLA-KIR*02,HLA-KIR*12 distribution frequency of the control group are 18%,15%,14%,52%,89%.The differences between them were all significant ( P<0.05).Among them The HLA-KIR*1003, HLA-KIR*14, HLA-KIR*17 distribution frequency of the observation group are significantly higher than the control group, HLA-KIR*02 , HLA-KIR*12 distribution frequency of the observation group are significantly lower than the control group.Conclusion:During the occurrence and development of the Cervical cancer,the HLA-KIR*1003,HLA-KIR*14,HLA-KIR*17 may be the risk factors for the Cervical cancer;the HLA-KIR*02,HLA-KIR*12 may be the protective factors for the Cervical cancer.
3.Progress in clinical research of asteroid hyalosis

Asteroid Hyalosis (AH) is a common clinical disease,which has been considered a benign disorder as it rarely impairs visual acuity.It was often discovered when the patient was treated for other eye diseases.The mechanism was unclear.Its characteristic B-ultrasound property makes the B-ultrasound a very helpful diagnostic technique.In the case of the patients with other fundus diseases associated with AH,optical coherence tomography (OCT) and fluorescein angiography (FA) may be used to reduce the interference from asteroid bodies,therefore improve the fundus visibility.Recent studies have shown that AH can incorporate with many other eye diseases.For example,in patients with cataracts,asteroid hyalosis can cause surface calcification of silicone plate intraocular lenses,which in most cases may lead to the need for explantation of the calcified intraocular lenses.The efficacy of pars plana vitrectomy (PPV),the removal of some,or all,of the eye`s vitreous humor for AH remains controversial.In this paper,we provide a review of the recent literature on AH disease: the etiology,diagnosis and treatment.We hope to thus improve the awareness and outcomes of AH disease.
5.Advances in nanocrystal technology and its application to improve the pharmacological efficacy for poorly-water soluble drugs

In order to solve the problems of erratic drug absorption and low bioavailability after oral administration for poorly-water soluble drugs due to low solubility, a series of novel pharmaceutical dosage forms as solid dispersion, liposome, microemulsion, vesicle, cyclodextrin inclusion complexes and drug nanocrystal have been developed in recent years. Among which drug nanocrystal attracts more attentions for its simpler preparation method, higher drug loading and easier manufacturing technology in the design of dosage forms suitable for different administration routes. In this paper, the nanocrystals of the poorly-water soluble drugs prepared based on bottom-up and top-down technologies were introduced. The characteristics and applications of the nanocrystal-based dosage forms as suspension, tablet and capsule were also introduced and carefully evaluated with the focus on their pharmacokinetics, pharmacodynamics and tissue targeted drug distribution after delivery by oral administration, intravenous injection and pulmonary inhalation. The advantages of drug nanocrystals in their therapeutics effects over the bulk drugs were discussed together with the inherent mechanism. Finally, the problems existing in basic research and scaled-up manufacture of drug nanocrystal as well as the possible ways of solution were listed out so as to make the nanocrystal-based preparations exert their maximum therapeutic effect after clinical application.
6.Advances in novel anti-HIV-1 drugs and drug candidates: 2005-2008.

HIV and AIDS remain as the crucial global health concern, therefore, research and development of novel anti-HIV-1 chemical therapeutics is still of paramount significance, which may be illuminated by cases of successful marketed drugs. Herein, we document the discovery and biological profile of new anti-HIV-1 drugs approved by FDA between 2005 and 2008 and some drug candidates are also discussed.
7.Comparison of therapeutic effects between different blood purification therapies for treatment of patients with multiple organ dysfunction syndrome

Objective To compare the clinical therapeutic effects of continuous veno-venous hemofiltration (CVVH) and continuous veno-venous hemodiafiltration (CVVHDF) for treatment of patients with multiple organ dysfunction syndrome (MODS).Methods A prospective observation was conducted, seventy patients with MODS admitted to the Department of Critical Care Medicine of the Affiliated Hospital of Zunyi Medical College from September 2013 to December 2016 were enrolled, and they were divided into a CVVH group and a CVVHDF group according to different treatment, 35 cases in each group. In the CVVH group, the ultrafiltration fluid flow rate was set at 1800 mL/h, while in the CVVHDF group, the flow rate was set at 2000 mL/h for both substitution fluid and dialysate, and the blood flow of the two groups was 150-180 mL/min. The changes of creatinine (SCr), urea nitrogen (BUN), pH value, HCO3-, helper T cell (Th1, Th2) and Th1/Th2 ratio were compared between the two groups before and after treatment for 24, 48 and 72 hours.Results With the extension of time, SCr, BUN were reduced in both groups after treatment compared to those before treatment, 72 hoursafter treatment they reached the lowest value, and the degree of change in CVVHDF group was more significant than that in CVVH group [SCr (μmol/L): 150.62±32.09 vs. 180.41±30.52, BUN (mmol/L): 7.31±2.19 vs. 9.06±2.36. allP < 0.05], after treatment; the pH value, HCO3- level in the two groups had a tendency at first elevated and then lowered, 48 hours after treatment they reached the peak values, compared with those before treatment, the differences were statistically significant [CVVH group: the pH value 7.42±0.08 vs. 7.25±0.10, HCO3- (mmol/L) was 22.49±5.11 vs. 15.65±4.16; CVVHDF group: pH value 7.41±0.04 vs. 7.24±0.11, HCO3- (mmol/L) 23.24±4.78 vs. 15.65±4.16, allP < 0.05], 72 hours after treatment, they began reduced (CVVH group: pH value, HCO3- were 7.39±0.09, 22.35±4.91 respectively, CVVHDF group: pH value, HCO3- were 7.38±0.06, 23.13±4.61 respectively), but they were still significantly higher than those before treatment, and at each time point compared between the two groups, no statistical significant difference was seen (allP > 0.05). With the extension of therapeutic time, Th1, Th1/Th2 in two groups were gradually elevated after treatment, and 72 hours after treatment they reached the peak values; Th2 showed a trend of gradual decrease and after 72 hours of treatment it reached the lowest value, and the changes in CVVHDF group were more significant than those in CVVH group [Th1 (×10-2): 1.51±0.12 vs. 1.21±0.11, Th2 (×10-2): 1.64±0.65 vs. 1.70±0.18, Th1/Th2: 0.91±0.14 vs. 0.71±0.15, allP < 0.05].Conclusion Both CVVH and CVVHDF can effectively remove inflammatory mediators and metabolic products, adjust the balance of acid and base, regulate immune system in the body of patients with MODS, and the CVVHDF is more effective than CVVH.
8.EFFECTS OF VITAMIN C AND GLUTATHIONE ON ULTRAWEAK LUMINESCENCE OF PULMONARY ALVEOLUS MACROPHAGES IN VITRO

Objective:[WT5BZ] The effects of exogenous vitamin C (VC) and glutathione (GSH) on ultraweak spontaneous luminescence of culturing pulmonary alveolus macrophages from rabbits were studiesd. [WT5FZ]Methods:[WT5BZ] In a special thermostat, which was passed through by airs with various concentrations of oxygen, the alveolar macrophages (AMs) were cultured in DMEM medium with VC or GSH, and the spontaneous luminescence of culturing AMs was examined by a luminometer. [WT5FZ]Results:[WT5BZ] when VC in medium was over 0.3 mmol/L, it could significantly enhance the oxidative luminescence of cells exposed to O 2 and cell death was resulted. However, when the cells were exposed to air without O 2 there was no significant effect. On the contrary,the lower concentration of VC (0.03 mmol/L) as well as GSH could reduce the spontaneous luminescence of cells exposed to a high concentration (99.1%) of O 2 in air. [WT5FZ]Conclusion:[WT5BZ] The results show that the spontaneous oxidation of culturing cells is an important reason for the ultraweak luminescence. High concentration of VC can promote cellular oxidative damage in vitro, but the exogenous GSH has a protective effect against oxidization in culturing cells.
10.Comparison of Humoral Immunity Indexes of Acute Glomerulonephritis and Nephritic Syndrome in Children

Objective To compare the humoral immunity features of acute glomerulonephritis and nephritic syndrome(NS) in children for earlier differential diagnosis and rational administration.Methods Nephelometry was used to determinate the serum level of immunoglobulin(Ig)G,IgA,IgM,C3,C4 in children by England MININEPH machine and correlated reagent. The serum level of IgE was determined by IgE enzyme linked immunosorbent assay(ELISA),assay ELISA kit of America E&ELABSINC company.
