2. Distribution and phylogenetic analysis of endophytic fungi in stems and leaves of Desmos chinensis based on histochemistry and rDNA ITS sequence

OBJECTIVE: To study the distribution and phylogenesis of endophytic fungi in the stems and leaves of Desmos chinensis. METHODS: Permanent paraffin-cut section, optical microscope photography, and histochemistry were used to observe the distribution of endophytic fungi in the stems and leaves of Desmos chinensis. Plate isolation method was used to separate and cultivate the endophytic fungi. The genomic DNA of endophytic fungi were extracted, then the rDNA ITS region was amplified using polymerase chain reaction (PCR). The PCR products were digested by Hae III and Hha I restriction enzymes to determine the species and genotypes, and sequenced according to the digestion genotyping results. Nucleotide sequences of the rDNA ITS of endophytic fungi were used for the identification and phylogenetic analysis. RESULTS: Endophytic fungi mainly existed in the parenchyma cells of the phloem in the stems, and in the parenchyma cells, palisade tissue or sponge tissue of the leaves. Fourteen strains of endophytic fungi were isolated from the stems and leaves. Four different genotypes and their sequences were obtained from RFLP analysis and sequencing. These strains were identified as Guignardia mangiferae, Colletotrichum gloeosporioides, and Phomopsis spp. The dominant group of those endophytic strains was Phomopsis, represented by 8 strains and accounting for 57.1% of all. CONCLUSION: The endophytic fungi distribute in both stems and leaves of Desmos chinensis without obvious tissue specificity. The result of the phylogenetic analyses with rDNA ITS sequences reveals the phylogenetic relationships of endophytic fungi in Desmos chinensis.
4.Clinical study on intermaxillary traction screws used in internal fixation in the treatment of jaw fracture

Objective:To analyse the methord and effects of intermaxillary traction(IMT)screws used in the treatment of jaw frac-ture.Methods:In the treatment of 1 68 cases of jaw fracture IMT screws were used for the restoration of normal and stable occlusion. The screw number,position,traction effect and postoperative complications of the treatment were analysed.Results:705 screws were used in 168 patients,4 screws were used in each of 147 cased (88%),6 in 12(7%),5 in 9(5%).336(47.7%)screws were fixed between the roots of first and second premolar,292(41 .4%)between the roots of canine and first premolar,50 (7.1%)be-tween the roots of second premolar and first molar,27(3.8%)between the other tooth roots.Normal postoperative occlusion relation was achived by the use of screws in 92 cases without traction.Occlusion disharmony or deviation was found in 76 cases by the used of screws and was restored to normlal by 1 4-day traction in 71 cases.The total efficiency of the treatment was 97%.Pain of the mucosa surrounding the screws was observed in 23 cases (1 3.7%).Root damage,traction screw loosening and adjacent tooth dislocation were observed in 1 3(1 .8%),1 1 (1 .6%)and 2(0.3%)cases respectively.Conclusion:For the fixation of IMT screws,the posi-tions between tooth root from canine to first molar were most common and safe.The fixation point should be in 5 ~8mm below the gin-gival margin.Use of 4-6 screws can meet treatment need.
5.Surgical treatment of trigeminal neuralgia of the third division by nerve avulsion inside and outside the mandibular cannel

Objective:To study the effects of the surgical treatment of nerve avulsion inside and outside the mandibular cannel.Methods:34 patients with trigeminal neuralgia of the third division,received neurotomy and avulsion of inferior alveolar nerve in the mandibular cannel above the lingual or through the mental foramen,concurrently with the avulsion of buccal nerve.Patients were fol-lowed up for 2-4 years.Results:All the operations were successfully completed,The wounds were well-healed.All patients felt that pain disappeared in the area innervated by inferior alveolar nerve.During the 2-4 years of follow-up,no recurrence was found.Conclusion:Inferior alveolar nerve neurotomy and avulsion in the mandibular cannel is simple,safe and effective in the treatment of trigeminal neuralgia of the third division.

AIM: To study the effects of IL-6 and IL-1? on the blood polymorphonuclear-neutrophils(PMN) apoptosis postburn. METHODS: Wistar rats inflicted by 30% total body surface area (TBSA) Ⅲ degree scalding were employed as the model. PMN were isolated by density gradient centrifugation using Percoll-hypaque and labeled with TdT-mediated and dUTP nick end labeling (TUNEL) and analyzed by flow cytometric analysis. The intracellular caspase-3 activation and the serum levels of IL-6 and IL-1? were analyzed by fluorometric immunosorbent enzyme assay and enzyme-linked immunosorbent assay, respectively. The present study aims to predict the action targets of antidepressant active ingredients of Xiaoyaosan to understand the "multi-components, multi-targets and multi-pathways" mechanism. Using network pharmacology, the reported antidepressant active ingredients in Xiaoyaosan (saikosaponin A, saikosaponin C, saikosaponin D, ferulic acid, Z-ligustilide, atractylenolide I, atractylenolide II, atractylenolide III, paeoniflorin, albiflorin, liquiritin, glycyrrhizic acid and pachymic acid), were used to predict the targets of main active ingredients of Xiaoyaosan according to reversed pharmacophore matching method. The prediction was made via screening of the antidepressive drug targets approved by FDA in the DrugBank database and annotating the information of targets with the aid of MAS 3.0 biological molecular function software. The Cytoscape software was used to construct the Xiaoyaosan ingredients-targets-pathways network. The network analysis indicates that the active ingredients in Xiaoyaosan involve 25 targets in the energy metabolism-immune-signal transmutation relevant biological processes. The antidepressant effect of Xiaoyaosan reflects the features of traditional Chinese medicine in multi-components, multi-targets and multi-pathways. This research provides a scientific basis for elucidation of the antidepressant pharmacological mechanism of Xiaoyaosan.

The tail suspension test (TST), forced swimming test (FST) and chronic unpredictable mild stress (CUMS) model were used to evaluate the anti-depressant effect of supercritical CO2 extract from Compound Chaigui Fang (FFCGF). A nuclear magnetic resonance (NMR)-based metabonomics combined with multivariate statistical analysis was performed to explore the mechanism of FFCGF. Rats were conducted by CUMS procedure for 28 days and drugs were administrated at the same time. The body weight, sucrose preference, crossings and rearings in open-field tests were evaluated and the urine was collected simultaneously. The metabonomic profiles of rats' urine were analyzed by NMR and potential biomarkers were searched by multivariate statistical analysis. The results showed that administration of FFCGF significantly decreasing the immobility time in FST and TST and improving rats' body weight, sucrose preference, crossings and rearings in CUMS, which were indication that the anti-depressant effect of FFCGF was abvious. Significant differences in the metabolic profile of the CUMS treated group and the control group were observed, which were consistent with the results of behavioral tests. Decreased levels of acetic acid, succinic acid, 2-oxidation glutaric acid and citric acid and increased glycine and pyruvic acid in urine were significantly affected by the CUMS procedure and the 6 biomarkers were reversed evidently after administration of FFCGF. These changes were suggestion that the anti-depressant mechanism of FFCGF was associated with energy metabolism, lipid metabolism and amino acid metabolism.

10.Analysis on biotransformation of Epimedium brevicornu flavonoids.

This study aims to investigate the biotransformation of Epimedium brevicornu flavonoids under the effect of hydrolytic enzymes in vitro. Snailase was mainly used to hydrolyze E. brevicornu flavonoids, and HPLC was used to determine the content of the main flavonoids in E. brevicornu flavonoids. The data results showed that the main known flavonoids included icariin, epimedin A, epi-mendin B and epimendin C, which were completely transformed into baohuoside I, sagittatoside A, sagittatoside B and 2"-O-rhamnosyl-icariside II in 1-2 h, respectively. Their transformed products were continuously hydrolyzed over time. In conclusion, snailase could transform E. brevicornu flavonoids into secondary glycoside or aglycone under 37 degrees C in pH 6.0 HBSS balanced salt solution in 2 h. Moreover, its enzymatic hydrolysates were consistent with intestinal metabolites.
