Objective: To establish the standard for quality control of Zhiganqing Granules (Radix et Rhizoma Rhei, Fructus Ligustri Lucidi, Rhizoma Alismatis, Pericarpium Citri Reticulatae, Polyporus Umbellatus, etc.) Methods: Fructus Ligustri Lucidi, Rhizoma Alismatis and Pericarpium Citri Reticulatae in the preparation were identified by TLC. The emodin in the preparation was determined by HPLC. A good linear range was shown at the concentration from 0.224?g to 0.732?g. The average recovery was 96.8% and the RSD was 1.14%.
3.Application of lectin microarray technology for the diagnosis of common gynecological tumors

Objective To probe the clinical values of the human serum glycoprotein profiles for the diagnosis of common gynecological tumors. Methods A total of 123 clinical serum samples which included 31 breast cancer, 24 cervical cancer, 19 ovarian cancer and 49 healthy individuals were collected. A lectin microarray consisting of 15 lectins with different glycan binding specificities was used to determine the glycoprotein profiles of serum sam-ples. Stepwise discrimination analysis method was adopted to establish function model of clinical serum samples classification with SPSS 15. 0 software. Results Two grades of diagnostic discrimination function models were es-tablished. The first grade discrimination function could differentiate gynecological tumors from healthy individuals, the diagnostic accuracy rates of retrospective inspection were 85. 7% and 83. 8% respectively, and the total diag-nostic accuracy rate was 84.6%. The second grade discrimination function was used to differentiate breast tumor, cervical tumor and ovarian tumor, the diagnostic accuracy rates of retrospective inspection were 96.8%,75.0%and 78.9% respectively, and the total diagnostic accuracy rate was 85.1%. Conclusion The human serum gly-coprotein profiles are associated with gynecological tumors, and the established discrimination function models based on lectin microarray data have a helpful reference value for the clinical diagnosis of gynecological tumors.
4.Effect of Total Glucosides of Paeony on Bone Density, Protein Level of Skeletal Muscle and Relative Cytokine in Rat Model of Osteoporosis

Objective To determine the influence of total glucosides of paeony (TGP) on bone density, protein level of skeletal muscle and relative cytokine of osteoporotic rats. Methods Six-month-old SD rats were shamed as control (8 rats) or were ovariectomized (24 rats). The ovariectomized (16 rats) +TGP low dose and high dose group were treated with 50 mg/(kg?d) TGP and 150 mg/(kg?d) TGP respectively for 12 weeks. After the animals were sacrificed, IL-6, TNF-?, IGF-1, TGF-? in serum, protein level of skeletal muscle in right quadriceps femoris, and bone density of right femur and lumbar vertebra were determined. Objective To observe effects for exercise and diet on learning and memory ability in mice with encephalon aging induced by D-galactose(D-gal).Methods The model of mice with encephalon aging was made by D-gal.The learning and memory ability of mice was determined by Morris water maze.Results There was significant difference between high fat feed encephalon aging group and restrict food on normal feed encephalon aging group,normal feed and exercise encephalon aging group,high fat feed and exercise encephalon aging group,high fat feed normal group,normal feed and exercise group,normal feed group(all P<0.05).In spatial probe test,there was significant difference between restrict food on normal feed encephalon aging group and normal feed encephalon aging group(all P<0.05).Conclusion Exercise and restrict food can improve the learning and memory function in the mice;feed with high fat can promote encephalon aging.
7.Determination of Entrapment Efficiency of Matrine Microspheres by HPLC

Objective:To establish an HPLC method for the determination of entrapment efficiency of matrine microspheres .Meth-ods:A Fortis XiC18 (250 mm ×4.6 mm, 5μm) column was used , the mobile phase consisted of methanol-acetonitrile-3%phosphoric acid solution (11∶80∶9).The flow rate was 0.6 ml· min-1.The detection wavelength was 210 nm, the column temperature was at 25℃and the sample size was 20 μl.Results: The linear range of matrine was 3-150 μg · ml-1 ( r =0.999 7).The recovery was 99.92%(RSD=0.97%, n=9).The entrapment efficiency of matrine microspheres was 81.85%±3.22%(n=3).Conclusion: The method is suitable for the determination of entrapment efficiency of matrine microspheres .

Natural killer T (NKT) cells,a subset of lymphocytes that bridge innate and adaptive immune systems,involve in processes of infection immunity,tumor immunity,transplantation immunity and autoimmunity.A significant progress has been made in the mechanisms of origin,selection,differentiation and maturation of NKT cells.However,some viewpoints are still controversial,and need to be further intensively investigated.The potential therapeutic applications of functional NKT cells have been suggested in the prevention and the treatment of various diseases.
9.Study on Optimization of Extraction Technology of Jiuwei Yiwei Mixture by Orthogonal Design

OBJECTIVE:To optimize water reflux extraction technology of Jiuwei yiwei mixture. METHODS:With the over-all score of the content of salvianoli acid B,a water-soluble component of the principal drug Salvia miltiorrhiza,the content of pae-oniflorin in Paeonia Radix Rubra and dry ointment yield rate as the observed index,and taking soaking time,extraction time, amount of water added,and extraction times as the factors,L9(34)orthogonal test was designed to optimize the extraction technolo-gy of Jiuwei yiwei mixture. And the verification test was conducted. RESULTS:The optimal technology was to add water with an amount 8 times as much as that of the drugs to soak them for 1 h and to conduct extraction twice,1 h for each. The overall scores of 3 batches of samples in the verification test were respectively 98.793,99.078 and 99.220(RSD<2%,n=3). CONCLUSIONS:The optimal technology for Jiuwei yiwei mixture is stable and feasible.
10.Reverse hybridization applied in detection on human papillomavirus infection of twenty-three subfamilies

Objective To establish a method of reverse hybridization to detect five subfamilies of low risk Human Papillomaviruses(HPV6,11,42,43 and 44)and eighteen subfamilies of high risk HPV (HPV16,18,31,33,35,39,45,51,52,53,56,58,59,66,68,73,83 and MM4)in one reaction.Methods Special probes for twenty-three HPV subfamilies were fixed on nylon membrane bars,biotin labeled general primers mediated polymerase chain reaction(GP-PCR)were applied in HPV DNA amplification.PCR amplified DNA fragments were reversely hybridized with special probes that were fixed on the membranes. All samples(136)detected by reverse hybridization method were paralleled with the methods of Hybridization Capture Ⅱ(HC-Ⅱ)and sequencing.Results Positive rate of the 136 samples detected by reverse hybridization was 41.9%,while HC-Ⅱ 42.6% and sequencing 40.4%.Reverse hybridization detection indicated coherence with the other two methods(Kappa 0.8644 and 0.9089,respectively).While sequencing was lab standard for DNA test,the sensitivity was 96.36%,specificity was 95.06%,accuracy was 95.59%.Conclusions Method of reverse hybridization is adaptable to 23 kinds of HPV subfamilies, which can confirm the exactly subfamilies of HPV infection.This method is adaptable in clinical detection of HPV,with high sensitivity,high specificity,simply and convenient operation and the results are easily to be read.
