Objective To investigate the effects of dibutylphthalate (DBP) on the proliferation and apoptosis of HL-60 leukemic cells and to study its mechanism to purge leukemic cells. Methods The effects of DBP on proliferation of HL-60 leukemic cells were measured by cell culture method. The effects on apoptosis were measured by the percentage of the apoptotic cells in morphology and of the DNA fragmentation and by DNA gel electrophoresis. The intracellular free Ca~(2+) concentration ([Ca~(2+)]i) of leukemic cells were measured by Fura-2AM method. The protein expressions of c-myc and bcl-2 genes of leukemic cells were measured by immunohistochemical assay. Results DBP could suppress the proliferation of HL-60 leukemic cells in a dose-dependent and time-dependent manner and induce them to die via apoptosis. It could elicit a intracellular Ca~(2+) redistribution and a potent extracellular calcium influx. It also down-regulated the protein expressions of c-myc and bcl-2 genes of HL-60 leukemic cells. Conclusion DBP can purge (HL-60) leukemic cells in vitro by increasing [Ca~(2+)]i of cells to initiate apoptosis and down-regulating bcl-2 and c-myc proto-gene expressions to promote cell apoptosis.

Objective By analyzing the graph features of fundamental frequency at different rates of/i/with continuously variable pitch and loundness made by normal adults,we could explore that the rate and gender have some impact on the graph to lay the foundation of regulating the clinical maximum counting ability. Methods Nor-mal adults of 51 (25 males,26 females were included). After each deep breathing, the subjects respectively took sustained speech/i/ with continuously variable pitch and lounolness twice at different rates of fast, medium and slow speed. Some parameters were measured byDoctor Speechat different rates of the tone/i/:Total time(T), Mean time standard deviation (MTSD),Mean wave crest (MWC),Mean wave trough (MWT),Mean frequency range (MFR),Mean frequency range standard deviation (MFRSD),comparing the speed having some effect on those above parameters with gender.ResuIts ①With the increasing rate of speech, the tone/i/of T (P<0.001), MFR (P<0.01)and MTSD (P<0.001)were significantly decreased,MWT significantly increased (P<0.001), While the MWC and MFRSD did not change significantly (P>0.05 ).②The tone /i/ of T (P>0.05 )were not effected by gender, female's MWT (P<0.001),MWC (P<0.001),MFR (P<0.001),MFRSD (P<0.01)were obviously higher than male's,while female's MTSD were significantly larger than male's (P<0.01).ConcIusion We should consider the impact of rate of speech and gender in clinical tests and values of MCA.The maximum counting capacity can be respectively tested at three different rates of speech for each subject, but only are the tone/i/of fun-damental frequency graphs with MWT,MWC,MFR,and MFRSD in normal range with corresponding speed and gender, can we get the value or can the value be valid.

Journal of Applied Clinical Pediatrics 1992;0(05):-
Objective To search for the reasons of high positive rate of amotile bacteria and the diagnosis of septicemia in new-born Methods The blood was drawn from the different site of the new-born with septicemia and carricd out blood culture. The drug sensitivity test had been done by the method of paper stripdiffusion. The plasmids of bacteria were extracted rapidly by medified Birnboim method and the plasmid analyss was carried out. The plasmids's DNA of 35 epidemic strain was cut off by both restriction enzyme of Hind Ⅲ and EcoR Ⅰ. The outer membrane protein (OMP) was determined by SDS-polyacrylamide gel electrophoresis. Results There are 51 patients with positive blood culture amotile bacterium,of them, pollution; 35 cases (68.6%), septicemia: only 16 cases (31.4%),54.8% (57/104) strains bacteria have drug resistance to more of 12 drugs. 87.3% (165/189) strains bacteria have plasmids. They are cut off as 6 DNA fragments (1.9,2,4,5, 8.5 and 18Kb) by Hind Ⅲ restrietion enzyme. and as 5 DNA fragments (2,2.6,3.2, 6.3 and 22 Kb) by EcoR Ⅰrestrietion enzyme, it is showed that they come from a same clone. The epidemic strain include 10 slips OMP, but non-epidemic strain have 11 slip OMP, increase a 25Kd belt. The amotile bacteria with above-mentioned plasmid spectrum, restriction enzyme spectrum and OMP spectrum are only seen in the air, therapeutic dish and syringe needle. Conclusion The pollution is an important reason of amotile bactorium high positiye rate in new-born.Diagnosing septicemia should depend on bacteria culture, plasmid analysis restriction enzyme analysis of plasmid DNA, oMP determination and combining medical history and clinical manifestation.

Dingzhi Xiaowan is a widely used traditional Chinese medicine in treating depression, which is a similar formula of Kaixinsan. In this research, a rapid ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF/MS(E)) method was established to analyze the metabolites of Dingzhi Xiaowan in depressive model rat plasma, bile, urine and feces. After we established Chronic unpredictable mild stress (CUMS) model rats and orally administrated Dingzhi Xiaowan, rat plasma, bile, urine and feces samples were collected and prepared. Using Waters Cortects UPLC C18 column (2.1 mm x 50 mm, 1.6 μm), acetonitrile-0.1% formic acid mobile phase gradient, these samples were analyzed and 33 metabolites of nine bioactive compounds were detected and tentatively identified by Metabolynx. Among the 33 metabolites, three metabolites were identified from plasma sample, three came from bile sample, and 27 metabolites were identified from urine and feces samples. This approach provided a rapid method for characterizing the metabolites of Dingzhi Xiaowan and gave the truly active structures and the action mechanism of their antidepressant effects.

To observe the effects of total flavones of Metasequoia glyptostroboides Hu et Cheng (TFM) on volume-overload cardiac hypertrophy and the expression of c-Fos protein in rat. Methods Volume-overload cardiac hypertrophy of rat was induced by aortocaval shunts. The rats were given ig TFM (400, 40 and 4 mg/kg/d). c-Fos protein in the ventricles were measured by immunocytochemical study. Results TFM at the above dosage decreased heart weight and contents of RNA and protein in the myocardium, inhibited the expression of c-Fos protein in the ventricles. Conclusion TFM can prevent volume-overload cardiac hypertrophy in rats. The inhibitory effects on the expression of c-Fos protein may be its mechanism in the molecular level.

The efficacy of Chinese herbal formula in treating depression has been proved in many studies. In this study, six different Kaixin San formulas were compared to investigate their effects on central monoamine neurotransmitters of chronic stress rats and against depression based on their different components in plasma, in order to discuss the efficacy-comparability relationship and the possible efficacy mechanism. The classic isolation method and the chronic unpredictable mild stress (CUMS) depression model were combined to investigate the changes in contents in hippocampus and monoamine neurotransmitters (NE, DA, 5-HT) and the components of some formulas in plasma with HPLC and UPLC-Q-TOF-MSE methods. As a result, Dingzhi Xiaowan recorded in Essential Recipes for Emergent Use Worth A Thousand significantly increased the behavioral scores, NE and 5-HT contents in hippocampus and NE, DA and 5-HT contents in cortex, with the best anti-depressant effect. Dingzhi Xiaowan recorded in Complete Records of Ancient and Modern Medical Works showed a notable increase in sucrose preference and open field score in model rats, NE content in hippocampus and NE, DA and 5-HT contents in cortex, with a certain anti anti-depressant effect. Kaixin San recorded in Ishinpo showed remarkable rise in weight of model rats. NE content in hippocampus and DA content in cortex. Puxin Decoction recorded in A Supplement to Recipes Worth A Thousand Gold showed 5-HT content in hippocampus and DA content in cortex. Kaixin San recorded in Yimenfang only showed DA content in cortex. Kaixin Wan recorded in Essential Recipes for Emergent Use Worth A Thousand did not mention the antidepressant effect. According to the results, the formulas' different anti-depressant effects may be related to the different plasma components.
