Bulletin of The Academy of Military Medical Sciences 2010;34(1):37-39
Objective To establish a calcification mode in vitro of human vascular smooth muscle cells (HVSMCs) induced by β-GP. Methods Primary HVSMCs were obtained from human embryo by plant method and confirmed by stain with α-sm-actin antibody. The cells after 4-6 passages were divided into two groups. The control group was incubated with normal DMEM medium while the calcification group was incubated with the medium containing 10 mmol/L β-glycerophosphate for 10 days.Calcification was confirmed by alizarin red S stain and alkaline phosphatase(ALP) assays. Results The primary cells observed by S-P stain were positive and the cells after being stained were pale yellow. After being induced with β-GP, the cells of calcification group began concentric growth and formed vesicles. Alizarin red S staining showed that the reaction of calcifying nodules was red,ALP activity was higher than that of controls at various time points(4 d,6 d,8 d and 10 d,P<0.01 ).Conclusion The HVSMCs could be induced into calcification in vitro by β-GP, and this model contribates to further studies of vascular diseases.

Anelectrochemicallyreducedgrapheneoxide/goldnanoparticle-chitosan(ERGO/AuNP-CS) composite film modified glassy carbon electrode ( GCE) was constructed by directly electrochemical reduction of GO, and then assembly of AuNP-CS polycation on the surface. The surface morphologies of different modified electrodes including bare GCE, GCE/GO, GCE/ERGO and GCE/ERGO/AuNP-CS were characterized by scanning electron microscopy ( SEM ) . The differential pulse voltammetric behaviors of the electrodes were investigated, and the results indicated that the composite of ERGO/AuNP-CS exhibited excellent electrocatalytic oxidation activity to uric acid ( UA) molecule. In 0. 10 mol/L of phosphate buffer solution (pH=6. 5) with a scanning rate of 100 mV/s, the proposed composite film modified electrode showed a linear electrochemical response to UA in the range of 0 . 05-110 μmol/L with a detection limit of 12. 4 nmol/L ( S/N = 3 ). The electrode displayed good selectivity, reproducibility and stability in the determination of UA in human serum and urine samples with a recovery of 93 . 8%-104 . 1%. The detection results were agreed with those of conventional spectrophotometry and uricase Kit methods.

Objective To probe the definition of cicateicial cryptotia and its plastic and reconstructive surgery. Methods Local skin flaps combined with full-thickness skin graft by zonation grafting technique were used to treat twenty-six patients with thirty-three cicateicial cryptotia.A cicateicial adhesion between auricle and cranial wall and mastoidea was removed, cranioaural angle was reformed, and helix and auricular lobule were reconstructed. Results All skin flaps survived, and the survival rate of skin grafts reached over 95 %.The appearance of auricle was very good, its major constructions might be noted, and cranioaural angle nearly returned to normal. The follow-up results from three months to one year post-operation showed that the appearance of auricle kept well, and the patients felt satisfactory. Conclusions In view of deformity characteristics of cicateicial cryptotia,it might be an effective approach to treat cicateicial cryptotia to take a seris of appropriate surgical procedures.
7.Effects of allicin on changes of hemorheology in focal cerebral ischemia-reperfusion injury.
China Journal of Chinese Materia Medica 2007;32(13):1314-1317
OBJECTIVETo explore the effects of allicin on the changes of hemorheology in focal cerebral ischemia-reperfusion injury in rats.
METHODMiddle cerebral artery occlusion (MCAO) was used to make focal cerebral ischemia-reperfusion model by intravascular nylon filament occlusion. The protective effects of allicin at different doses were evaluated by investigating neurological function score, infarction volume and water content of brain. The changes of blood rheology were detected.
RESULTCompared with model group, allicin (15, 25 mg x kg(-1)) increased the neurological function score and decreased the water content and infarction volume of brain in rats. Allicin (15, 25 mg x kg(-1)) inhibited the increasing of the blood viscosity, high shear rate reduced viscosity, high shear relative reduced viscosity and low shear relative reduced viscosity.
CONCLUSIONAllicin has protective effects on cerebral ischemia-reperfusion injuries. The mechanism may be related to inhibit the increasing of hemorheology.
