2.Expressions of CD133 and p53 in colorectal carcinoma and the clinical relvance

Objective To investigate the expressions of CD133 and p53 in colorectal cancer and their clin-ical significances. Methods The expressions of CD133 and p53 in 74 colorectal cancer patients were detected by the immunohistochemistry method. The relationships of CD133 and p53 with the clinicopathological parameters and prognosis were analyzed. Results The positive expression rates of CD133 and p53 in colorectal cancer tissues were 33.8%and 55.4%,respectively. The expression levels of CD133 and p53 were not related to age,sex,tumor location and histological type ,and but were significantly related to the histological differentiation ,TNM stage and distant metastasis(P<0.05,respectively). The Spearman correlation analysis showed that there was no correlation between the levels of CD133 and p53. Conclusions The high expressions of CD133 and p53 in colorectal cancer tissues were closely related to the histological differentiation and TNM stage. CD133 and p53 could be used as important biomarkers for the evaluation of malignant biological behavior,and the diagnosis and prognosis of colorectal cancer.
3.Diagnosis and treatment of primary colonic malignant lymphoma

Objective To investigate the diagnosis and treatment procedures of primary colonic malignant lymphoma(PCML). Methods Data of clinical and pathological findings, surgical treatment and prognosis of 12 cases with PCML verified by pathology were retrospectively analyzed, workups of barium clysma,colon-scopic examination and multislice spiral computed tomography(MSCT) were compared to evaluate their role in diagnosis. Results The classic presentation of PCML included abdominal pain, abdominal bulge, bowel obstruction, gastrointestinal bleeding, and serum levels of tumor markers were within the normal ran-ges. Six cases underwent procedure of barium clysma, and anomaly appeared in 2 cases, however, there were no indications of lymphoma, 6 cases underwent colon-scopic examination, anomaly in 3 cases, of which 1 revealed possibility of lymphoma, 10 cases underwent MSCT; 9 cases demonstrated tumor origin by CT scan of which diagnosis was established in 5 cases. All the patients underwent surgery. No incidence of peri-opreative death happened. Non-Hodgkin' s lymphoma was confirmed in all of cases. For all of the cases a T-cell origin was in 1 case and a B-cell origin in other cases. Ten cases were administrated postoperative chem-otherapy. 11 cases were followed up, and the overall 1-year and 3-year survival rates were 81.9% (9/11) and 54.5% (6/11), respectively. Conclusions Typical presentation is still lacking for PCML, workups of barium clysma and colon-seopic examination are little sensitive and specific to make the diagnosis. However, MSCT is useful in diagnosing PCML with its characteristic information as well as clinical findings. Surgical management and postoperative adjuvant-chemotherapy would be likely the optimal therapeutic maneuver of this disease in early period.
4.Analysis of Microstructure of Plant Fibers with Different Hemicellulose Content by Solid State Nuclear Magnetic Resonance and Atomic Force Microscope

Fibers with different hemicellulose contents were produced using various degree removal of hemicellulose to obtain large differences in cellulose and hemicellulose proportions at a similar lignin content. Solid state cross polarisation magic angle spinning carbon-13 nuclear magnetic resonance(CP/MAS ~(13)C-NMR) and atomic force microscope(AFM) had been employed to investigate the microstructure of fibers. The results showed that there was an increase in relative content of para-crystalline cellulose with the decreas of hemicellulose content obtained by the spectral fitting for the cellulose C1-region(δ 102-108). The elementary fibril size was relatively constant between 4.0 and 4.3 nm for the three samples obtained by the spectral fitting for the cellulose C4-region(δ 80-92). The difference in elementary fibril size between the samples was not significant. However, the elementary fibril aggregate size increased from 17.9 to 22.2 nm with the decreas of hemicellulose content, which was a significant change. The results of AFM analysis showed that the fiber with a high hemicellulose content had a porous surface structure. In fibres with a low hemicellulose content, the elementary fibril aggregates formed a much more compact surface structure. Lower hemicellulose content can promote the partially irreversible microfibril aggregation, which caused tensions in the microfibrils due to the finite dimensions of the cell wall, amounting to stress in the microfibrils. However, the porous structure can be improved as hemicellulose content decreased to a certain extent.

8.Development and clinical application of an quantitative head-band formed ocular compressor.

In order to investigate the efficiency of a new quantitative head-band formed ocular compressor to reduce intraocular pressure (IOP), ocular compression by this new reducer with 40 mmHg for 10 min was performed on 87 cataractous eyes of 78 cases. The changes of IOP (87 eyes) and anterior chamber depth (ACD) were observed. There was a significant decrease of IOP and increase of ACD within 30 min after decompression (P < 0.001). The mean decrease of IOP was 5.62 +/- 2.41 mmHg and the mean increase of ACD was 0.18 +/- 0.09 mm within 5 min after decompression. The IOP 5 min after decompression had no significant difference with that 10 min after decompression (P > 0.05). IOP below 10 mmHg could last for about 15 min. This apparatus had been successfully applied to 80 eyes for extracapsular cataract extraction. It was suggested that this device had the advantages of safety, accurate quantification, reliable effect, casually adjusting pressure according to various demands and time-saving.

9.Research advances of photosensitizer in photodynamic therapy of glioblastoma

Glioblastoma is a malignant tumor in central nervous system, which has strong invasion, poor prognosis and short survival time. At present, the main treatment strategy of glioblastoma is surgical excision, supplemented by radiotherapy and chemotherapy. However, due to incomplete resection and high recurrence rate, it is urgent to find novel therapeutic method for glioblastoma. Photodynamic therapy, as a promising non-surgical treatment, provides a new strategy for postoperative adjuvant therapy of glioblastoma. This review summarizes the mechanism and clinical application of photodynamic therapy mediated by various photosensitizers in glioblastoma, in order to provide help for the treatment of glioblastoma.
10.Effects of ultrasound-mediated SonoVue microbubbles destruction on the integrity and expression of hAng-1 gene

Objective To test the efficiency of gene transfer and expression mediated by ultrasound and microbubble strategy in 293T cell. Methods SonoVue microbubbles were mixed with pla.smid DNA encoding hAng-1 and green fluorescent protein. The mixture of the DNA and microbubbles was administer to cultured 293T cells by ultrasound exposue. The ultrasound condition was 1. 5 W/cm~2 and 30 s. Forty eight hours later, transfer rate was assessed by fluorescence microscopy and flow cytometry. Cell viability was assayed by Trypan Blue staining. RT-PCR and Western blot analysis was used to examine the expression of hAng-1 mRNA and protein. Agarose gel electrophoresis was used to evaluate the integrity of the plasmid. Results The transfection expression rate of eGFP in 293T cells was markedly increased with the additon of 20% microbubbles and 15 mg/L DNA. Fetal calf serum had no influence on the gene transfer rate. Ultrasound irradiation combined with microbubbles couldn't destroy the integrity of plasmid. Conclusions Ultrasound-mediated microbubbles destruction can increase the transfection and expression of hAng-1 gene in 293T.
