4.Progress on bone marrow mesenchymal stem cells transplantation for spinal cord injury.

Bone marrow mesenchymal cells (BMSCs) are regarded as donor cells in cell transplantation therapies for spinal cord injury (SCI) for they have the ability of favourable proliferation and multi-directional differentiation, and are easily isolated and culturd and have less immunological reaction. It has been confirmed that subarachnoid space injection is the most ideal delivery technique of BMSCs. Bone marrow mesenchymal stem cells transplantation is safe and its reconditioning role is certain for SCI in early clinical application. The mechanism of BMSCs promoting functional recovery after SCI is probably concerned with vicarious function, nerve trophism, immunosuppression and promoting axonal regeneration by BMSCs.

5.Effects of QDTMT on brain natriuretic peptide and ventricular remodeling of acute myocardial infarction in rabbit

Objective To investigate the effects of Qidantongmai Tablet (QDTMT, a Chinese patent medicine) on the dynamic changes in plasma brain natriuretic peptide (BNP) after acute myocardial infarction (AMI) in rabbit induced by ligation of coronary artery, and to explore its influences on ventricular remodeling. Methods Thirty-six healthy rabbits were randomly divided into six groups with six animals for each group: normal group, sham operation group, model group (AMI group, reproduced by ligation of left circumflex branch of the coronary artery), control group (diltiazem hydrochloride group), high dose QDTMT group and low dose QDTMT group. The preventive effect of QDTMT on AMI and its impact on BNP and ventricular remodeling were then investigated. ObjectiveTo investigate the correlation between the level of resistin and the insulin resistance(IR) and the incorporation of microamount albuminuria(MA) of type 2 diabetes mellitus (T2DM).MethodsTwo hundred and twenty T2DM patients and 40 normal subjects (control group) were enrolled in this study.Two hundred and twenty T2DM patients were divided into 4 groups,IR group (group A,79 cases),non-IR group (group B,48 cases),IR complicating MA group (group C,51 cases),non-IR complicating MA group(group D,42 cases).The fasting serum resistin was measured by enzyme-linked immunosorbant assay (ELISA),and the fasting plasma glucose (FPG),glycosylated hemoglobin (HbA1c),fasting insulin (FINS) was also determined.ResultsThe level of fasting serum resistin in group A,B,C and D was higher than that in control group [(33.45 ± 1.37),(23.36 ± 1.47),(44.45 ± 1.39),(37.45 ±1.57) μ g/L vs.( 17.44 ± 1.26 ) μ g/L],and there was significant difference among 5 groups (P < 0.01 ).The level of fasting serum resistin in group A was higher than that in group B (P < 0.01 ),and the level of fasting serum resistin in group C was higher than that in group D (P < 0.01 ).The level of fasting serum resistin was both higher in group C and D than that in goup A and B (P< 0.01 ).Correlation analysis showed the fasting serum resistin was positively correlated with body mass index(BMI),FPG,HbA1c and FINS in group A(r =0.35,0.46,0.37,0.49,P <0.05),and the fasting serum resistin was positively correlated with HbA1c,BMI,systolic blood pressure and diastolic blood pressure in group C(r =0.45,0.32,0.37,0.29,P < 0.05 ).ConclusionsSerum resistin may participate in the process of IR and the formation of MA.It may become one of the diagnostic standard of the IR and one of the important index to estimate the MA.
7.A preliminary study of the relationship between the (C-106T) single nucleotide polymorphism in the aldose reductase gene promoter region and diabetic nephropathy

Objective To study the relationship between the (C 106T) single nucleotide polymorphsim of promoter region of aldose reductase gene and type 2 diabetes mellitus with nephropathy in Chinese Han from Chongqing. Methods The (C 106T) allele and genotype of 192 patients of type 2 diabetes were examined by polymerase chain reaction agarose gel electrophoresis and restriction enzyme Bfa Ⅰ. Results The frequencies of both allele C and genotype CC were significantly higher in type 2 diabetes patients with albuminuria than in those without albuminuria. Logistic regression analysis showed that allele C and genotype CC were associated with diabetic nephropathy in type 2 diabetes mellitus patients( P =0.032 and 0.046, respectively). Conclusion This finding suggests that (C 106T) single nucleotide polymorphism in the aldose reductase gene promoter region may be associated with diabetic nephropathy of Chinese Han type 2 diabetes. It may be a functional polymorphism and can serve as an inherited marker for susceptibility of diabetic nephropathy.
8.Advances in photodynamic therapy based on tumor hypoxia

Photodynamic therapy (PDT) has attracted wide attention due to its unique advantages such as minimal invasiveness, high efficiency and high selectivity, and its ability to induce anti-tumor immune response. However, the treatment process is heavily dependent on the oxygen content of the treatment site, and the widespread oxygen deficiency in malignant tumors severely limits its efficacy. In addition, PDT-mediated oxygen depletion exacerbates tumor hypoxia, which further reduces its therapeutic effect. In recent years, many researches have been devoted to overcoming this problem. This paper summarized various strategies based on tumor hypoxic PDT in recent years, discussing the advantages and disadvantages of these strategies, and analyzing the main challenges and future directions of PDT in the treatment of tumors, so as to provide references for the in-depth study of photodynamic therapy of tumors.
9.Effects of ultrasound-mediated SonoVue microbubbles destruction on the integrity and expression of hAng-1 gene

Objective To test the efficiency of gene transfer and expression mediated by ultrasound and microbubble strategy in 293T cell. Methods SonoVue microbubbles were mixed with pla.smid DNA encoding hAng-1 and green fluorescent protein. The mixture of the DNA and microbubbles was administer to cultured 293T cells by ultrasound exposue. The ultrasound condition was 1. 5 W/cm~2 and 30 s. Forty eight hours later, transfer rate was assessed by fluorescence microscopy and flow cytometry. Cell viability was assayed by Trypan Blue staining. RT-PCR and Western blot analysis was used to examine the expression of hAng-1 mRNA and protein. Agarose gel electrophoresis was used to evaluate the integrity of the plasmid. Results The transfection expression rate of eGFP in 293T cells was markedly increased with the additon of 20% microbubbles and 15 mg/L DNA. Fetal calf serum had no influence on the gene transfer rate. Ultrasound irradiation combined with microbubbles couldn't destroy the integrity of plasmid. Conclusions Ultrasound-mediated microbubbles destruction can increase the transfection and expression of hAng-1 gene in 293T.
10.Study on accumulation law of peimine in different Fritillaria thunbergii cultivar.

In order to study the accumulation of Fritillaria thunbergii cultivar, peimine content in Xiaye, Kuanye, Duozi and Xiaosanzi bulbs of different sizes and parts was determined by HPLC-ELSE. The results indicated that the peimine content varied significantly with the cultivar type, the size and part of bulb. The distribution laws of peimine were as follow: Xiaosanzi > Duozi > Xiaye > Kuanye, small-size bulb > big-size bulb, core bud > scale. The peimine yield per plant in Duozi was the highest.
