1.Analysis of the characteristics of dyslipidaemia in women with polycystic ovary syndrome

A cross-sectional study of 92 women with polycystic ovary syndrome(PCOS) was performed.The patients were composed of 4 groups according to the Rotterdam criteria and were also divided into 2 groups according to body mass index and divided into 2 groups according to insulin-releasing test.Anthropometric measurements were recorded,and hormonal profile was assessed by measuring FSH,LH,prolactin,and testosterone.The metabolic profile was investigated by measuring glucose and insulin level during oral glucose tolerance test (OGTT),then calculating the homeostasis model assessment (HOMA).Serum lipid levels,including total cholesterol,triglycerides,high-density lipoprotein-cholesterol,and low-density lipoprotein-cholesterol were determirned.The result suggests that the classification according to the revised 2003 Rotterdam consensus on diagnosis does not reflect dyslipidemia in PCOS.Dyslipidemia in hyperinsulinemic group and obesity group were more severe than that in non-hyperinsulinemic group and non-obese group ; this finding needs our attention in early treatment and prevention of late complications of PCOS.
2.The Influence of Immune RNA on Endotoxin-Induced Uveitis

Objective: Our aim was to investigate whether the immune RNA (iRNA) can inhibit the endotoxin-induced uveitis (EIU). Methods: The EIU was induced by injecting endotoxin of E.Coli into rats hypodermically. iRNA was abstracted from the livers and spleens of the rabbits which had been injected by E.Coli six times. The iRNA was introduced intraperitoneally into the rats both at half an hour and 3 hours after the injection of endotoxin. The effect of iRNA was evaluated by protein determination and cell count in aqueous houmer(AH). Results: The appearance of EIU in the iRNA treated rats was delayed, about 3 hours later than the rats received only endotoxin injection. The AH protein decreased by 58.34% and cellular infiltration decreased by 27.62% in iRNA treated group at the 24 hours. Conclusion: The iRNA may partly prevent the development of EIU and significantly reduce the severity of EIU.
3.CT Manifestations of Diffuse Axonal Injury:A Report of 56 Cases

Objective To make a further recongnizing of the manifestations of diffuse axonal injury (DAI)on CT images for the early accurate clinical diagnosis.Methods The CT image data and its correlated clinic features of 56 cases with diffuse axonal injury(44 male,12 female)were analyzed retrospectively.In this series,43 cases were caused by traffic accident,13 by falling from high place.Results 1. 44 cases had haemorrhage lesions(less than 2 cm in diameter)in brain parenchyma which were at the corticomedullary junction,corpus callosum,brain stem,basal ganglia,internal capsule.2. 41 cases had subarachnoid and/or intraventricular hemorrhage 3. 9 cases had acute generalized brain swelling 4. 5 cases were associated with epidural hematoma and 16 with subdural hematoma.Conclusion CT manifestation of DAI have some featrues,and can provide reliable evidence for accurate clinic diagnosis of DAI.
4.Independent design experiments carried out in the preventive medicine experimental teaching

To improve the quality of experimental teaching,investigating the independent design experiments are carrying out in preventive medicine by public health experimental teaching center in our school.We take preventive medicine professional as an example,and besides the classical basis experiments,some experiments are verification project at present,in which students are not strongly interested.On the basis of the integration of the classic experiments,according to the national standard of the experimental method,we are trying to carry out part of the independent design experiments to meet the needs of students learning.The effect of those independent design experiments shows that it is appropriate carrying out independent design experiments on the basis of the existing classical experimental project in preventive medicine experimental teaching,to a certain extent,which will stimulate students' learning interest and enhance their sense of achievement.This study is conducive to the cultivation of medical students' innovation,independent thinking,analysis and problem-solving ability,practical ability and teamwork spirit.
5.The Time Distribution of Physical Activity per Working Day of Non-agriculture Employed Population in China

Objective To study the time distribution of non-agriculture employed population spent in a working day in China, and to provide basic information for intervention strategies.Methods The data of 2002 China Nutrition and Health Survey were used. The information on daily activities including occupation,transportation,exercise,housework,sedentary activity and sleep was described.Results Non-agriculture employed population spent 8.41 h,0.58 h,0.09 h,3.11 h,1.40 h and 7.89 h on occupation, transportation,exercise,sedentary activity,housework and sleep,respectively.Administrator,technologist and clerks spent less time on occupation activity than service workers,production and transportation workers and others did,and they spent more time on sedentary activity.Male spent more time on occupation activity and less time on housework and sleep than female did.People in rural area spent more time on occupation activity than those in urban area,but less time On transportation and sedentary activity.Conclusion Differences in time use among different employed groups,gender and area were found,which should be considered when intervention measure is developed.
6.Hemodynamic changes and oxygen metabolism during off-pump coronary artery bypass grafting

Objective To examine the hemodynamic changes and oxygen metabolism during off-pump coronary artery bypass grafting (OPCABG) .Methods Twenty patients (18 male, 2 female) aged (60?8)yr, weighing (71?9)kg undergoing OPCABG were studied. The patients were premedicated with diazepam 10 mg PO 2h before operation and morphine 10 mg and scopolamine 0.3 mg im 30 min before operation. Anesthesia was induced with midazolam 0.1-0.2 mg?kg-1, fentanyl 5-10 ug?kg-1 and vecuronium 0.12-0.15 mg?kg-1 and maintained with 0.5%-2% isoflurane inhalation, fentanyl 10-20 ug?kg-1 and intermittent iv boluses of vecuronium and in some patients propofol infusion. Radial artery was cannulated. Swan-Ganz catheter was placed via internal jugular vein into pulmonary artery. Hemodynamic parameters including MAP, HR, cardiac index (CI), stroke volume (SV), MPAP, PCWP, SVR, left ventricle stroke work index (LVSWI) and RVSWI; oxygen metabolism including oxygen delivery (DO2) , oxygen consumption (VO2) and oxygen extraction ratio (ERO2); regional cerebral oxygen saturation (rSCO2) and arterial blood lactate level (ABL) were measured and recorded before skin incision (A), when pericardium was incised before operation on the heart (B), during operation on the heart (C), 10 min after operation on the heart (D) and at the end of surgery (E) . Results During surgical manipulation on the heart (C) MAP, SV and CI decreased while CVP and HR increased, but the decrease in CI and SV was not significant. During manipulation on the heart(C) DO2 decreased whereas ERO2 increased, but there was no significant change in VO2. After manipulation on the heart (D) DO2 and ERO2 returned to the pre-manipulation level. Arterial blood lactate concentration increased at E. Conclusion OPCABG can be performed uneventfully under our general anesthetic regimen. During operation while the beating heart is being manipulated MAP tends to decrease while CVP and HR increase. The changes are of short duration and do not result in any serious consequeaces.

Orjective:To obtain recombinant CHO-K1 with expressing sPDGFR? and to identify the biological activities of sPDGFR? secreted in non-serum medium.Methods:Recombinant human sPDGFR? expression vector pIRES-Neo3-sPDGFR?-Fc was constructed and then transfected into CHO-K1 cells by using LipofectamineTM 2000.After screened with G418 in 8 weeks,some monoclone cells were selected randomly to amplify in 96-well-plate to 24-well-plates,and then to identify positive cell clones by RT-PCR.Furthermore,the candidate cell clones were test by Real-Time PCR and Western blot assays.Finally,anti-proliferation activities of the expressed sPDGFR? were analyzed by MTT.Results:sPDGFR?-Fc was cloned into pIRES-Neo3 correctly.The sPDGFR?-Fc expression level in recombinant CHO-K1 cell clones were concordant in between Realtime PCR and Western blot assay.sPDGFR?-Fc obtained from cultured non-serum medium of positive CHO-K1 could significantly inhibit proliferation of vascular endothelial cell.Conclusion:Successed to select recombinant CHO-K1 cell lines with high expressed sPDGFR?-Fc.The sPDGFR?-Fc can inhibit the cell proliferation significantly and it means sPDGFR?-Fc might be a new anti-cancer drug in the future.

9.Effects of jinlongshe granules on apoptosis of MKN-45 human gastric cancer cells orthotopically transplanted in nude mice

OBJECTIVE: To assess the effects of Jinlongshe Granules (JLSG) on tumor growth of gastric carcinoma. METHODS: Fifty nude mice orthotopically transplanted with MKN-45 human gastric cancer cells were divided into five groups: untreated group, 5-fluorouracil (5-FU)-treated group and high-, medium-, and low-dose JLSG-treated groups. Corresponding antitumor drugs were administered in each group except the untreated group. The antitumor effects in vivo were evaluated. Cell cycle distribution and apoptosis of MKN-45 human gastric cancer cells were determined by using flow cytometry (FCM) and Annexin V-fluorescein isothiocyanate/propidium iodide (Annexin V-FITC/PI) staining assay. The ultrastructure of MKN-45 gastric cancer cells was observed by transmission electron microscope. RESULTS: In the mice treated with high-, medium-, and low-dose JLSG, the growth inhibition rates of gastric cancer were 68.13%, 55.94% and 50.31% respectively, and this antitumor effect was dose-dependent. In the mice treated with intraperitoneal injection of 5-FU, the growth inhibition rate of gastric cancer was 53.43% and not much different from those treated with JLSG. The apoptotic rates in the high-, medium-, and low-dose JLSG-treated groups were 22.81%, 28.27% and 38.54% respectively, in a dose-dependent manner, with the cell cycle arrested at G(0)/G(1) phase. An Annexin V-FITC/PI staining assay revealed that the percentages of early apoptotic cells in the three dose JLSG-treated groups were all significantly higher than that in the 5-FU-treated group, whereas the late apoptotic and necrotic cells were much more in the 5-FU-treated group than those in the three dose JLSG-treated groups. CONCLUSION: Jinlongshe Granules exert an inhibiting effect on MKN-45 human gastric cancer cell.
10.Inhibitory effect of polydatin on expression of toll-like receptor 4 in ischemia-reperfusion injured NRK-52E cells.

Polydatin is a monocrystaline compound isolated from Polygonum cuspidatum Sieb. et Zucc. (Polygonaceae) with biological properties, such as anti-inflammation, anti-oxidative and nephroprotective effects. Increasing number of studies have demonstrated the protective effect of polydatin on renal ischemia reperfusion injury. However, the possible mechanisms of this protection are not fully elucidated. This study aimed to investigate the effect of polydatin on ischemia-reperfusion induced expression of toll-like receptor4 (TLR4) in rat renal tubular epithelia cells (NRK-52E), and analyze the mechanism of polydatin on TLR4 signal pathway. The cultured NRK-52E cells were incubated in three gas incubators for a period of 6 h at hypoxia and 24h at reoxygenation to simulate the ischemia-reperfusion injury in vitro. TLR4 mRNA level was analyzed by real-time-PCR, and the protein expression of TLR4 and NF-κB by Western blotting, while TNF-α and IL-1β proteins expressions were detected by ELISA. Polydatin downregulated I/R induced mRNA and protein expressions of TLR4, and decreased the protein expression of NF-κB, TNF-α and IL-1β. The TLR4 blocker partially antagonized the effect of I/R on NF-κB signaling, and such inhibitory effect was markedly enhanced by polydatin. In the present study, polydatin protects NRK-52E cells from I/R injury possibly by relieving the inflammatory response through regulation of TLR4/NF-κB signaling pathway.
