Journal of Third Military Medical University 1988;0(05):-
Objective To study the mechanism of extracellular signal-regulated kinase (ERK) pathway inhibitor U0126 on cell cycle of K562 cell lines. Methods The mRNA and protein expressions of ERK, Cyclin D2, Cyclin E in and P27 K562 cell lines after 10 ?mol/L U0126 treatment for 12, 24 and 72 h were detected by RT-PCR and Western blot, respectively. The cell cycle was determined by flow cytometry. Results The mRNA and protein expressions of ERK, Cyclin D2 and Cyclin E in K562 cell lines after U0126 treatment were decreased, while those of P27 were increased. The percentage of cells in G0/G1 phase was increased and that in S phase was decreased. All data mentioned above showed significant difference between the K562 cell lines before and after U0126 treatment(P

Journal of Clinical Otorhinolaryngology Head and Neck Surgery 2014;28(22):1791-1794
OBJECTIVE:
To explore the expression levels of three T-type calcium channel receptors (α1G; α1H; α1I) in the cochlea and spiral ganglion neurons of C57BL/6J mice with different ages.
METHOD:
Thirty cases of C57BL/6J mice were divided into three groups (6-8 W, 24-26 W, 42-44 W) according to the age. The expressions of three T-type calcium channel receptors were quantified by RT-PCR after hearing thresholds measured by ABR.
RESULT:
Three receptors were detected in the cochlea and spiral ganglion neurons of 6-8 W C57BL/6J mice. The quantitative results showed that the expression levels of α1H and α1I were highest among three receptors in spiral ganglion neurons and in the cochlea respectively. The expression levels of three receptors significantly decreased with age, especially at the age of 4244 W.
CONCLUSION
The expression of T-type calcium channel receptors reduced with age in the inner ear of C57BL/6J mice.

Objective To study the effect of p38 mitogen activated protein kinase (MAPK) pathway inhibitors SB203580 on cell cycle of K562 cell lines and its mechanisms. Methods The expression of mRNA and protein of p38,Cyclin D2,Cyelin E and P27 in K562 cell lines treated with SB203580 were detected by retrotranscription polymerase chain reaction (RT-PCR) and Western blotting, respectively. Cell cycle was determined by flow eytometry (FCM). Results The expressions of mRNA and protein of p38, Cyclin D2 and Cyclin E in K562 cell lines treated with SB203580 were decreased and the expression of p27 was increased. The percentage of cells in G0/G1 phase was increased and was decreased in S phase. There was a significant difference as compared with K562 cell lines before treated with SB203580. Conclusion SB203580 can affect cell cycle regulatory proteins by p38 pathway and eventually inhibit proliferation of K562 cells.

Methods 84 patients suffered from frozen shoulder were randomly divided into two groups from February 2012 to July 2014 , Group one was treated with local analgesic influsion equipment, another group was not treated with it. both groups received arthroscopic arthrolysis The mean duration of follow-up was 11.2 months after operation. The range of shoulder motion , VAS and ASES scores were recorded before the operation and one week, one month, three months after the operation. Results All patients were involved , the range of shoulder motion , VAS and ASES scores improved significantly after operation(P < 0.05). The differences of the range shoulder motion, ASES score after operation, the patients with local analgesic equipment improved significantly (P < 0.05),but there were no significant difference of VAS scores between two groups at 3months after operation (P > 0.05). Conclusion Local analgesic equipment could alleviate the pain of patients with frozen shoulder earlier and more effectively , promote recovery quickly , achieve expected effects.

OBJECTIVETo observe the expression of Ginkgo biloba Tablet (GbT) on scavenger receptor A (SRA) of the aortic wall and changes of serum inflammatory factors in atherosclerotic rats, and to explore its new mechanism for fighting against atherosclerosis (AS).
METHODSTotally 45 male Wistar rats were randomly divided into the control group, the model group, the GbT group, 15 rats in each group. Levels of blood glucose, blood lipids, blood calcium, serum C-reactive protein (CRP), soluble intercellular adhesion molecule-1 (slCAM-1), and soluble vascular cell adhesion molecule-1 (sVCAM-1) were measured in all rats. The expression of SRA in the aortic wall of atherosclerotic rats was observed by immunohistochemical assay. The correlation between the expression of SRA and levels of in-flammatory factors was also observed.
RESULTSCompared with the control group, blood glucose and blood calcium obviously increased (P < 0.05); levels of TG, TC, and LDL-C were significantly elevated (P < 0.01); neointimal areas were significantly thickened, increased intima percentage was significantly enlarged, narrowed lumen index was significantly reduced; levels of CRP, sICAM-1, and sVCAM-1 were significantly elevated in the model group (all P < 0.01). Compared with the model group, blood glucose and blood calcium obviously decreased (P < 0.05); levels of TG, TC, and LDL-C significantly decreased (P < 0.01) in the GbT group. Aortic lumens were obviously narrower in the model group than in the GbT group (P < 0.05). SRA expressed at the aortic wall. The aforesaid 3 indices were significantly improved in the GbT group than in the model group (P < 0.01). Serum levels of CRP, sICAM-1, and sVCAM-1 were significantly decreased in the GbT group than in the model group (P < 0.01). Serum levels of CRP, sICAM-1, and sVCAM-1 were positively correlated with the percentage of SRA positive expression area (r = 0.701, 0.604, 0.581, all P < 0.01).
CONCLUSIONSSerum levels of inflammatory factors in atherosclerotic rats were elevated, and the expression of SRA in the aortic wall was enhanced. The expression of SRA was closely correlated with serum levels of inflammatory factors. GbT could decrease serum levels of inflammatory factors and inhibit the expression of SRA.
