1.Effect of ERK pathway inhibitor U0126 on cell cycle of leukemia K562 cell lines

Objective To study the mechanism of extracellular signal-regulated kinase (ERK) pathway inhibitor U0126 on cell cycle of K562 cell lines. Methods The mRNA and protein expressions of ERK, Cyclin D2, Cyclin E in and P27 K562 cell lines after 10 ?mol/L U0126 treatment for 12, 24 and 72 h were detected by RT-PCR and Western blot, respectively. The cell cycle was determined by flow cytometry. Results The mRNA and protein expressions of ERK, Cyclin D2 and Cyclin E in K562 cell lines after U0126 treatment were decreased, while those of P27 were increased. The percentage of cells in G0/G1 phase was increased and that in S phase was decreased. All data mentioned above showed significant difference between the K562 cell lines before and after U0126 treatment(P
2.Clinical Application of the Expression of LOXL2 mRNA and Tenascin-C mRNA in Tissues of the Bile Duct Cancer

Objective To explore the clinical application of the expression of LOXL2 mRNA and Tenascin-C mRNA in tissues for the disease with the bile duct cancer.Methods The serum and clinical data in 35 cases of patients with the bile duct cancer (cancer group) and 28 cases of patients with normal bile duct tissue (control group) were collected,used the real-time fluorescent quantitative PCR (real-time-PCR,RT-PCR) technology to detect the expression of LOXL2 mRNA and TenascinC mRNA in tissues toobserve the relationship between the changes and the bile duct cancer for the two markers.Results The expression of LOXL2 mRNA and Tenascin-C mRNA in tissues in the cancer group were 1.27±0.18 and 1.39±0.19,which of ones in the control group were 0.20±0.06 and 0.23±0.06.In the cancer group,the expression of LOXL2 mRNA and Tenascin-C mRNA in tissues respectively with comparision to those in the control group were significantly higher,the differences had statistical significance(t=52.18,56.87,P<0.01),which of ones in the cancer group was positively related (r=0.687,P<0.01).Conclution The expression of LOXL2 mRNA and Tenascin-C mRNA in tissues may be a molecular targets for the disease with the bile duct cancer in the early diagnosis and judgment of progression in the courses of this disease.
4.In vitro study of xylitol on the growth and acid production of Actinomyces viscosus.

OBJECTIVEThis research aimed to study the inhibitory effect of xylitol on the growth and acid production of Actinomyces viscosus (A. viscosus).
METHODSWe cultivated A. viscosus in anaerobic conditions with different concentrations (128, 64, 32, 16, 8, and 4 g x L(-1)) of xylitol brain heart infusion liquid medium and determined the minimum inhibitory concentration (MIC). Subsequently, we measured the pH value of the control group, as well as those of 1/2, 1/4, 1/8 MIC, and MIC concentration groups at 1.5, 3, 6, 12, 24, and 48 h. The Delta pH and OD550 at 2, 4, 6, 8, 10, and 12 h were calculated. We discovered that the minimum xylitol concentrations suppressed 50% and 90% A. viscosus biofilm formation (i.e., MBIC50 and MBIC90). SPSS 19.0 was used to analyze the collected data, and conclusions were drawn afterward.
RESULTSXylitol inhibited the growth ofA. viscosus at MIC of 64 g x L(-1). After 12 h, the differences of pH value among groups were all statistically significant (P < 0.05), and Delta pH increased when the MIC concentration decreased. Except for the 1/2 MIC and MIC groups, the differences of OD550 among groups had no statistical significance (P>0.05), and OD550 also increased when the MIC concentration decreased. These results imply that the ability ofA. viscosus to grow and produce acid in 1/2 MIC and MIC conditions will be reduced with the increase in xylitol concentration. The value of MIBC50 was 64 g x L(-1), whereas the value of MIBC90 was 128 g x L(-1). This finding indicates that the xylitol medium can restrict A. viscosus biofilm formation.
CONCLUSIONXylitolcan effectively inhibit the growth, adhesion, and acid production ofA. viscosus, protecting teeth from cariogenic bacteria and preventing caries to a certain extent.

5.Prenatal ultrasonic diagnosis of fetal interrupted aortic arch

Objective To evaluate the value of prenatal ultrasound in the interrupted aortic arch (IAA), and analyze the reasons of misdiagnosis and improve diagnostic ratio of this kind of defects. Methods Ultrasonic characteristics were analyzed in 16 fetuses with aortic arch anomalies, which were compared with ultrasound image and autopsy results of the normal fetus. The relativity of fetal IAA and abnormal chromosome was also analyzed. Results Among the 16 cases, 15 cases were confirmed by anatomy and the accuracy of ultrasound diagnosis was 94% (15/16), of which one case was serious constriction of aortic arch, one case was error type, three cases were obtained explicit type due to unsatisfactory results of prenatal ultrasound. The coincidence rate of ultrasonic type was 73% (11/15). All of the corrected diagnosed cases were detected with ventricular septal, of which 5 cases were detected with complete endocardial cushion defect, 9 cases were found extra cardiac malformations. Among the 7 karyotype check cases, 3 cases were diagnosed with Trisomy 18, 1 case Trisomy 13, while the rest 3 cases normal. The incidence rate of Trisomy 18 was 43 % (3/7). Conclusions Prenatal ultrasound shows a high accuracy in diagnosing fetal interrupted aortic arch (IAA ), but there’s a need to be improved in explicit typing. It is difficult to identify fetal interrupted aortic arch (IAA) and serious constriction of aortic arch (COA). The incidence rate of Trisomy 18 in the abnormal chromosome of fetal IAA is high.
6.Effect of p38 MAPK pathway inhibitors SB203580 on cell cycle of leukemia K562 cell lines and its mechanisms

Objective To study the effect of p38 mitogen activated protein kinase (MAPK) pathway inhibitors SB203580 on cell cycle of K562 cell lines and its mechanisms. Methods The expression of mRNA and protein of p38,Cyclin D2,Cyelin E and P27 in K562 cell lines treated with SB203580 were detected by retrotranscription polymerase chain reaction (RT-PCR) and Western blotting, respectively. Cell cycle was determined by flow eytometry (FCM). Results The expressions of mRNA and protein of p38, Cyclin D2 and Cyclin E in K562 cell lines treated with SB203580 were decreased and the expression of p27 was increased.The percentage of cells in G0/G1 phase was increased and was decreased in S phase. There was a significant difference as compared with K562 cell lines before treated with SB203580. Conclusion SB203580 can affect cell cycle regulatory proteins by p38 pathway and eventually inhibit proliferation of K562 cells.
7.The influence of ursolic acid on VEGF, COX-2 and MMP-2 expressed in the mouse retinal ischemic model

Objective To investigate the influence of ursolic acid on vascular endothelial growth factor ( VEGF) , cycloxygen-ase-2 (COX-2), and matrix metalloproteinases-2 (MMP-2) expressed in the mouse retinal ischemic model , and to explore the mecha-nisms of anti-angiogenesis.Methods Sixty 7-day clean-class C57BL/6J mice were divided randomly into 6 groups [ n =10 mice (20 eyes) per group]:blank control, model control (PBS), positive control (triamcinolone), and ursolic acid (UA) intervention (low-dose, medium-dose, and high-dose).Mice in the blank control group were raised in air , and mice in other groups in(75%±2%)O2 high-oxygen environment for 5 consecutive days .Mice in the model control group and breastfeeding mice were put back in air environ-ment (21%O2 ) on the 12th day after the new-born mice to induce the generation of retinal neovascularization .When models were suc-cessful, the drug treatments were applied immediately to the corresponding groups , with injection of 3μl of sterile PBS in model control group, 3 μl of 1.5, 3.00 and 6.0 μg UA in UA intervention group, and 3 μl of triamcinolone (1 ml∶40 mg) in positive control group, respectively.All mice were killed after overdose anesthesia on the 17th day.Their eyeballs were made into samples and retinal tissue pathological sections with H-E dying method.The positive expressions of VEGF , COX-2, and MMP-2 were detected with immu-nohistochemical method .The fresh retinal tissue homogenate was prepared to detect the protein expressions of VEGF , COX-2, and MMP-2 in retinal tissue with western blot method ,and mRNA expressions of VEGF , COX-2, and MMP-2 were detected with real-time fluorescent quantitative polymerase chain reaction ( RT-PCR) .Results According to protein and mRNA expressions of VEGF , COX-2,and MMP-2 in retinal tissue among six groups , protein expressions of VEGF , COX-2, and MMP-2 in model group were significantly higher than those in blank group ( P <0.05 ) .Each protein expression in the high UA intervention group was significantly lower than that in the model group ( P <0.05 ) .Each protein expression in the high UA intervention group was not significantly different from that in the positive group ( P >0.05 ) .Each protein expression in the high UA intervention group was significantly lower than that in the low UA intervention group( P <0.05).Conclusions UA inhibited expressions of VEGF, COX-2, and MMP-2 in retinal ischemia model .UA also played an inhibitory role in the formation of neovascularization , and this role was positively correlated with UA dose .
8.Diagnostic Value of Fluorescent Quantitative Polymerase Chain Reaction for Mycoplasma Pneumoniae Pneumoniae in Children with Mycoplasma Pneumoniae Pneumonia

Objective To evaluate the diagnostic value of fluorescent quantitative polymerase chain reaction(PCR) for Mycoplasma pneumoniae (MP) in children with MP pneumonia(MPP).Methods From Jun.2008 to Jan.2009,153 cases hospitalized with pneumonia were enrolled,and 30 cases without respiratory infection were enrolled as control group.Their respiratory secretion (including nasopharyngeal secretion,sputum,bronchialalveolar lavage fluid or pharyngeal swab) samples were collected for fluorescent quantitative PCR for MP.And their single or paired serums were collected for specific MP antibody detection.Results There were 123 cases confirmed with MPP by serology,among whom 114 cases were MP PCR positive.The quantitation of MP DNA was among 1.20?106-3.66?1010 gene copys/L. There were 30 cases with pneumonia negative with MP by the paired serum serology,among whom 2 cases were MP PCR positive,and the quantitation of MP DNA was (1.08-3.02)?107gene copys/L.All cases of control group were MP PCR negative.During the first and second weeks of the MPP onset,the sensitivity of MP-IgM from the first single blood samples were 66.7% and 83.9%,respectively.While the sensitivity and specificity of MP PCR were 92.7% and 93.3%,respectively.From the third week of the disease onset,the sensitivity of MP-IgM from the first single blood samples increased to 90.9%-100%.The clinical manifestations of MPP were nonspecific.Conclusions PCR is superior to serology for early diagnosis on MP infection.Combination of the 2 methods may be helpful to early and accurate diagnosis on MP infection.
9.Observation of therapeutic effects of point application at Shenque (CV 8) plus moxa-salt hot compress for prevention of gastrointestinal adverse reactions after chemotherapy for non-Hodgkin lymphoma

Objective:To observe the clinical efficacy of point application with Da Huang (Rhizoma Rhei Cruda) powder at Shenque (CV 8) plus moxa-salt hot compress on the umbilicus for preventing gastrointestinal adverse reactions after chemotherapy for non-Hodgkin lymphoma (NHL).Methods:A total of 60 cases with NHL under chemotherapy were divided into two groups by hospitalization order,with 30 cases in each group.The control group was treated with routine nursing and the observation group was additionally given point application with Da Huang (Rhizoma Rhei Cruda) powder plus moxa-salt hot compress on the umbilicus,to compare the effect in preventing gastrointestinal adverse reactions after chemotherapy between the two groups.Results:The occurrence rates of constipation,nausea,vomiting and poor appetite on the second day and fifth day after chemotherapy were obviously lower in the observation group than those in the control group,with statistically significant differences between the two groups (all P<0.05).Conclusion:The point application with Da Huang (Rhizoma Rhei Cruda) powder at Shenque (CV 8) plus maxa-salt hot compress on the umbilicus can produce more significant efficacy in preventing the gastrointestinal adverse reactions after chemotherapy for NHL than routine nursing.Moreover,it is simple and easy to be used and popularized.
10.Effect of whole-body hypothermia on cardiac hemodynamics neonatal piglet after hypoxia-ischemia

Objective To determine the effect of mild hypothermia on neonatal piglet cardiac hemodynamic function after hypoxia-ischemia (HI).Method Twenty five 7-day-old piglets were used for hypoxic ischemic brain damage (HIBD) model by the method of temporary occlusion of the bilateral carotid arteries and followed by mechanical ventilation with low concentration of oxygen (FiO_2=6%) for 30 minutes.The piglets were randomly divided into three groups:group A (normothermia with body temperature to 39℃,n=9),group B (body temperature to 36℃for 72 hours,n=8),and group C (body temperature to 34℃for 72 hours,n=8).Mild hypothermia was initiated at 4 hours after HI,the systolic and diastole function were evaluated by Doppler echocardiography at pre-HI,post-Hi 4 hours and post-HI 72 hours.Results There were no significant differences in left ventrieular ejection time/left ventrieular ejection time (LPEP/LVEF),right ventricular ejection acceleration time/right ventricular ejection time (RACT/RVET) and CO at post-HI with hypothermia 72 hours in three groups,but the heart rate decreased in B and group C group.Compared with nonnothermia,mild hypothermia treatment showed no significant differences in MAP,LPEP/LVET,RACT/RVET,CO,SV at post-HI with hypothermia 72 hours.Conclusions Body temperature decreased by 3~5℃for 72 hours will not aggravate hemodynamic abnormity.
