Objective To evaluate catheter-directed thromlysis (CDT) through three different approaches in combination with intervention for acute deep venous thrombosis (DVT) of the lower extremities. Methods In this study 137 acute DVT cases were enrolled. CDT was performed through small saphenous veins in 107 cases, through the great saphenous veins in 21 and through popliteal veins in 9.Iliac vein balloon dilation was performed in 66 cases, and stents were placed in 60 cases. Results The limb edema reduction rates between small saphenous vein group and great saphenous vein group and popliteal vein group were of no significant difference (82.3% ±7.6% vs 81.6% ±6.0% vs 83.9% ±6.1%,P>0.05).The difference of rates of thrombolysis ( 63.5 % ± 7.7% vs 66.9% ± 8.4% vs 66.1% ± 2.7% )between the 3 groups was not statistically significant (P > 0.05 ). The mean time required for the cannulation was shorter in great saphenous vein group than the other two groups [ (7.3 ± 0.3 ) min vs (20.8 ± 1.1 ) min and (15.7 ±0.6) min,P < 0.05 ].There were 12 cases complicating incision bleeding during thrombolysis in small saphenous vein group,in this group there were 5 cases complicating phlebitis and 8 complicating saphenous nerve injury. The complication rates in great saphenous vein group was lower (P < 0.05).Follow-up made in 112 cases for 3 -25 months. There were 97 cases without limb edema. While test by venography or color Doppler,89 cases showed venous patency and rethrombosis developed in 8 cases. Conclusions Catheter-directed thrombolysis with iliac venous intervention was an effective method for the treatment of acute deep venous thrombosis.CDT through great saphenous vein is easier with less complications.

AIM: To investigate the differentiation potential of human bone marrow mesenchymal stem cells (hBMMSCs) into hematopoietic cells in vivo. METHODS: hBMMSCs prepared from the bone marrow-aspirate sample obtained from healthy human donors were culture-expanded in vitro with 5-8 passages. hBMMSCs(P5-8, 4.8?10 5 cells/mouse) were injected into the severe combined immuodeficiency (SCID) mice treated by cyclophosphamide(CPA) and various tissues were analyzed at 35 days post-transplant for the presence of differentiated human cells. RESULTS: hBMMSCs(P5-8) viability, which was determined by typan blue staining at the end of the harvest and before infusion, was greater than 95% in every infusate at both time points. Cells characterized by flow cytometry using human MSC-specific monoclonal antibodies were uniformly positive for CD29, CD44, CD90, CD105, CD106, CD166 and negative for CD11a, CD14, CD34, CD38, CD45, CD80, CD86 which are common on cells of the hematopoietic lineages. Analysis of PB demonstrated that 5 of 6 hBMMSCs transplanted SCID mice had low level of circulating human CD45 +/ H-2D d- cells(range from 0.17% to 0.36%)and CD34 +/ H-2D d- cells(range from 0.10% to 0.50%). Analysis of BM for the presence of hematopoietic chimerism demonstrated human CD45 +/ H-2D d- cells and CD34 +/ H-2D d- cells in the marrow of 4 out of 6 hBMMSCs transplanted SCID mice (0.10%-0.19% and 0.03%-0.52%, respectively). Human hematopoietic cells with these same phenotype were also detected in the spleen 4 of the hBMMSCs transplanted SCID mice (range from 0.19% to 1.65% ,from 0.20% to 0.26%, respectively). No human hematopoietic cell was seen either in the PB, BM or spleen of all control animals. CONCLUSION: hBMMSCs have the ability to differentiate into blood cells of multiple lineages, including CD34 + hematopoietic stem cells/progenitor cells (HSC/HPC).

This study was aimed to establish a high performance liquid chromatography (HPLC) method coupled with Evaporative Light-scattering Detector (ELSD) in order to develop the determination of fingerprint of terpene lactones in Ginkgo biloba tablets. An Agilent Extend-C18 (4.6 mm í 250 mm, 5 μm) was employed as the analysis column and the normal propyl alcohol-tetrahydrofuran-water (1:15:84) as mobile phase. The column temperature was 30℃. And the flow rate was 1.0 mL·min-1. HPLC coupled with electrospray ionization quadrupole time-of-flight tandem mass spectrometry (Q-TOF MS) was used to identify the common peaks. The fingerprint was further evaluated by chemometrics methods including principal component analysis (PCA), similarity analysis (SA) and hierarchical clus-tering analysis (HCA). The results showed that the precision, stability and repeatability of this method were favorable. Five common peaks were identified by LC/Q-TOF MS as ginkgolide J ( M ) , C , A , B and bilobalide , respectively . Fourteen batches of Ginkgo biloba tablets were determined. With the aid of PCA, SA and HCA, the common pattern of the fingerprint of terpene lactones was established. Samples were divided into 4 clusters by their quality differ-ence. It was concluded that the method established in this paper can be used for quality evaluation of terpene lac-tones in G ink go b ilob a tablets.
