1.Value of MRS in the differential diagnosis of corpus callosum lesions

Objective To explore the value of multivoxel 1H-MRS in the differential diagnosis of corpus callosum lesions.Methods 17 patients with corpus callosum lesions confirmed by clinical or pathological method underwent MRS examination.These patients included 2 cases of lymphoma,4 cases of Marchiafava-Bignami disease,3 cases of glioma, 7 cases of infarction,and 1 case of multiple sclerosis.MRS features of lesions were analyzed.Results 2 cases with corpus callosum lymphoma revealed significantly increased Cho peak, decreased Cr and NAA peak,towering Lip peak.Among 4 cases with Marchiafava-Bignami diseases, 2 cases revealed increased Cho/Cr and decreased NAA/Cr,1 case showed normal,1 case revealed inverted Lac peak.For 3 cases with corpus callosum glioma,NAA peak decreased or disappeared,and Cho peak increased in varying degrees.7 cases with corpus callosum infarction revealed significant inverted Lac peak, and NAA peak decreased in varying degrees.1 case of multiple sclerosis revealed increased Cho peak,decreased NAA peak,and inverted Lac peak during acute period with enhancement,while these features returned normal during inactive period.Conclusion Multivoxel 1H-MRS plays an important role in the differential diagnosis of corpus callosum lesions.
2.1H-MRS research of cerebral metabolic changes in patients with type 2 diabetes mellitus

Objective To determine the efficacy of glucose-loweringtreatment by proton magnetic resonance spectroscopy (1H-MRS).Methods Bilateral frontal lobes of 45 male patients with type 2 diabetes mellitus (T2DM) underwent 1 H-MRS separately bythe time it was first diagnosed.Among them,30 were treated with insulin,15 were rejected.The bilateral frontal lobes of all patients were re-examined with 1 H-MRS after 6 months.The ratios of metabolites in the frontal lobe were recorded and compared,and observing whether the lactate(Lac) peak appeared or not.Results Significantly higher values of the N-acetylaspartate (NAA)/creatine (Cr)ratio and NAA/choline (Cho) ratio after 6 months (2.414 2± 0.206 6 and 2.244 2±0.341 5) were found in bilateral frontal lobes compared with pre-therapy (1.905 3±0.135 7 and 1.906 0±0.322 1)(P<0.05).Significantly lower values of the NAA/Cr ratio and NAA/Cho ratio in the refuse treatment group(1.504 2 ± 0.262 4 and 1.510 0± 0.220 8) were found in bilateral frontal lobes compared with pre therapy (1.960 0±0.175 0 and 1.880 8±0.297 0)(P<0.05).Lac peak was not detected in the all experiments.Conclusion 1 H-MRS provides valuable information in the assessment and evaluation of clinical efficacy and prognosis of T2DM by detecting the changes of brain metabolism.
3.Study on biological characteristics of adult adipose-derived mesenchymal stem cells and its differentiation into cardiomyocytes in vitro

Objective To induce adipose-derived mesenchymal stem cells (ADMSCs) differentiation into cardiomyocytes, so as to provide stem cells for myocardial regeneration.Method ADMSCs were isolated and cultured from adult adipose tissue in vitro.They were induced with 10μmol/L 5-azacytidine (5Aza) for 24h.At the 7th, 14th, 21st days after induction, the expression of α-sarcomeric actin, and myosin heavy chain were repeatedly detected by immunocytochemistry.The expression of ANP was detected by RT-PCR.Results At the 7th day after induction, there was no expression of α-sarcomeric actin and MHC.At the 14th day, there was a little expression of α-sarcomeric actin and MHC that could be seen in cells.At the 21st day, there was increased expression of α-sarcomeric and MHC, and the expression of ANP was positive results detected by RT-PCR.Conclusions 5-Aza can induced ADMSCs to differentiate into cardiomyocytes.ADMSCs might be a potential source of cell transplantation for myocardial.
4.Detection of human papillomavirus L1 capsid protein expression in liquid-based cytology samples with abnormal cytology

Objective To investigate the possibility of detection of the human papillomavirus (HPV)L1 capsid protein to predict the coarse of mild or moderate cervical intraepithelial neoplasia(CIN).Methods Immunocytochemical analysis using antibody against HPV L1 capsid protein was carried out on 274 samples obtained from women performed Tri Path Pap tests.positive for high-risk HPV DNA detected by hybrid capture Ⅱ (HC-Ⅱ)or cytologic diagnosed atypical squamous cells of undetermined significance (ASCUS)or more severe.For cytological diagnosed,there were ASCUS 105 cases,low-grade squamous intraepithelial lesions (LSIL) 119 cases,atypical squamous cells cannot exclude high-grade squamous intraepithelial lesion (ASC-H)9 cases,high-grade squamous intraepithelial lesions(HSIL)36 cases,and squamous cell carcinoma(SCC)5 cases.But for the pathologic diagnosed,there were chronic cervicitis 96 cases.CIN Ⅰ 55 cases,CIN Ⅱ 55 cases,CIN Ⅲ 32 cases,and SCC 6 cases.Results Of the 274 cases,HPV L1 capsid protein was positive in 69.8%(67/96) of cervicitis,83.5%(71/85)of CIN Ⅰ,41.8% (23/55)of CIN Ⅱ,3.1%(1/32)of CIN Ⅲ and 0(0/6)of SCC.Cytologic diagnosis revealed a higher expression rate in LSIL(75.6%.90/119) than that in ASCUS(63.8%,67/105)or in HSIL + SCC (9.8%,4/41;all P<0.01).Of 71 cases with ASCUS and ISIL without treated,none of HPV L1 positive cases(0/55)progressed in cytology,while 19%(3/16)of HPV L1 negative cases progressed to ASC-H,HSIL(P<0.01).Conclusion The expression rates of HPV L1 protein in liquid-based cell specimen is decreased as the cytopathology diagnosis severe degree.which may imply the histopathology diagnosis of cervix,predict the progression of cervical lesion,and help to treat the cases with ASCUS and LSIL.
5.Adiponectin reduces apoptosis by improving myocardial anti-oxidative activity after myocardial ischemia/reperfusion injury in diabetic rats

Aim To study the effect of adiponectin ( APN) on myocardial ischemia reperfusion( IR) injury in diabetic rats and to explore the role of oxidation-an-tioxidation system. Methods Male Sprague Dawley rats were randomly divided into control group( NS) , IR group ( NIR ) , diabetes group ( DS ) , DS + IR group (DIR), DS +APN +IR group(APN). Experimental diabetes was induced in the animals by a single intrap-eritoneal injection of streptozotocin at a dose of 55 mg ·kg-1 . The IR and NIR group were subjected to myo-cardial I/R injury. APN group was administered APN through intravenous injection 10 min before the reper-fusion, the others were administered normal saline. The rats were considered diabetic and used for the study only if their glucose levels were higher than 15 mmol·L-1. Results All the diabetic rats exhibited increased levels of blood glucose and reduction of body weight ( P <0. 05 ) . Compared with those of NS and DS groups, the myocardial infarct size, cardiomyocyte apoptosis, MDA concentration and ROS in NIR and DIR groups were remarkably increased, activities of SOD and NO were decreased(P<0. 05 or P<0. 01). APN decreased oxidative stress product generation and myocardial apoptosis induced by diabetic myocardial I/R injury ( P <0. 05 ) . Conclusion APN exerts pro-tective effect on myocardial I/R injury in diabetic rats through anti-oxidative mechanism.

To study the mechanisms of total flavonoid from Glycyrrhizae Radix et Rhizoma (TFGR) and its ingredient isoliquiritigenin (ISL) on their regulation of M2 phenotype polarization of macrophages. IL-4 (60 μg x L(-1)) induced RAW264.7 cells for 6 h to establish the M2 macrophage model. TFGR and ISL restrained breast cancer cells migration with the aid of M2 macrophages in vitro. TFGR and ISL inhibited gene and protein expression of Arg-1, up-regulated gene of HO-1 and protein expression of iNOS, enhanced the expression of microRNA 155 and its target gene SHIP1, meanwhile down-regulated.the phosphorylation of STAT3 and STAT6. So TFGR and ISL were the bioactive fraction and ingredient in Glycyrrhizae Radix et Rhizoma to reverse M2 phenotype macrophages polarization. TFGR and ISL inhibited the promotion of M2 macrophages to breast cancer cells migration in vitro, STAT signal pathways and miR155 were partly involved.

7.Distinguishing benign from malignant main duct intraductal papillary mucinous tumors of the pancreas by CT and MRI

Objective To evaluate the value of imaging studies in diagnosis and differentiating malignant from benign main duct intraductal papillary mucinous neoplasms (MD-IPMNs).Methods The imaging studies of a total of 31 patients with MD-IPMNs confirmed by pathology after surgery was retrospectively reviewed.All patients underwent either CT,MR or MRCP.Two radiologists observed the lesions' imaging feature,and then the lesions was scored,and the differentiation between malignant and benign was made.The Bland Altman method was used for evaluation of inter-observer agreement.The score of the lesions was compared with the pathological results.Finally,a ROC curve was used to calculate AUC,and to evaluate the role of the maximum diameter of the main pancreatic duct (MPD) obtained by imaging studies in differentiation of malignant and benign IPMNs,and to determine the best cut-off point,and sensitivity,specificity.Results Histological analysis revealed low grade dysplasia in 13 patients,middle grade dysplasia in 6 and high grade dysplasia in 5,and adenocarcinoma in 7.Imaging studies suggested benign lesions in 16,malignant lesions in 10 patients and disdiagnosis in 5.The inter-observer agreement on major imaging features was good.The maximum diameter of the MPD was clinically meaningful for distinguishing malignant from benign lesions,and the AUC was 83.8%,and the best cut-off value was 14.8 mm,the sensitivity and specificity was 66.7% and 100%.The presence of wall nodules could be an imaging feature for distinguishing malignant from benign lesions,but the size of nodules,location of nodules within pancreatic duct,the atrophy of pancreatic parenchyma,and dilated bile duct was not useful for differentiation.Conclusions The imaging studies are sensitive for diagnosis and differentiation between malignant and benign MD-IPMNs,and it is of clinical value for preoperative diagnosis and follow up.
8.Comparison on MSCT and pathological findings of metastatic lymph nodes in pancreatic cancer patients

Objective To discuss the MSCT imaging characteristics of metastatic lymph nodes in pancreatic cancer.Methods The MSCT images of 30 pancreatic cancer patients with metastatic lymph node confirmed by pathology were selected and all the patients underwent enhanced MSCT before surgery.The location, maximum short axis diameter ( MSAD) , sizes, density, enhancement, necrosis and fusion of the metastatic lymph nodes were analyzed using the 5th edition of TNM stage of Japan Pancreatic Society in 2002 as the standard.Results Sixty-three metastatic lymph nodes was pathologically diagnosed , while 53were diagnosed by MSCT.The metastatic lymph nodes most commonly occurred in group 13 and 17in pancreatic head cancer, while group 18 lymph node metastasis was more common in pancreatic body and tail cancer.The MSAD of metastatic lymph nodes was 2~17 mm with the average of (7.2 ±4.0)mm.All of metastatic lymph nodes were divided into MSAD <5 mm group, 5 mm~<10 mm group and ≥10 mm group based on their size.The number of metastatic lymph nodes of the three groups were 10, 18 and 25, respectively.The number of enhanced metastatic lymph nodes was decreased as the size increased, while the number of necrotic and fused lymph nodes was increased, which were statistically different (all P<0.05).Comparison between the two groups showed that the number of enhancement metastatic lymph nodes in MSAD <5 mm group and 5 mm~<10 mm group were more than that in ≥10 mm group ( 7/10 and 11/18 vs 2/25 ) .The number of fused lymph nodes in MSAD <5 mm group and 5 mm~<10 mm group were less than that in≥10 mm group (0 and 2/18 vs 22/25 ) .Clear edge metastatic lymph nodes were more in MSAD <5 mm group than that in≥10 mm group (6/10 vs 5/25).The number of necrotic metastatic lymph nodes in MSAD <5 mm group was less than that in ≥10 mm group(2 /10 vs 18/25).All the differences above were statistically significant ( all P<0.05), but no other significant differences were found between two groups.Conclusions The main imaging findings of metastatic lymph nodes in pancreatic cancer were unobvious enhancement, intratumoral necrosis and fusion.Heterogeneous density and unclear edge could benefit the diagnosis of metastatic lymph node.
9.Protoplast Regeneration and Mutagenesis Breeding of Streptomyces qinlingensis sp. nov.

To improve the antibiotics production of Streptomyces qinlingensis sp. nov.,protoplast regeneration combined with physical and chemical mutagenesis was used to selected high-yielding strains. The results showed that the antibacterial activities of strain R-72 from protoplast regeneration and NTG-1,H30-7 from protoplast mutagenesis against Bacillus subtilis were more than 20% higher than that of the original strain,and the heredity characters of those strains were stable in successive ten generations. The further bioassay experiments exhibited that the fungicidal and antibacterial activities of the fermentation broth from R-72,NTG-1 and H30-7 were remarkable increased comparing with that of the starting strain.
