2.The Establishment of ELISA Method for Detecting Babesia gibsoni by Recombinant Antigen BgAMA-1

In order to improve diagnostic method to Babesia gibsoni infection,gene recombination technique was used in our study.By this technique,we constructed and expressed recombinant apical membrane antigen-1(BgAMA-1),and recombinant BgAMA-1 was used in ELISA experiment as diagnostic antigen.The results showed the recombinant expressed plasmid was triumphantly constructed and the recombinant antigen BgAMA-1 had a high specificity in ELISA test.These results demonstrate that the recombinant BgAMA-1 might be a useful diagnostic reagent for detection of antibodies to B.gibsoni in dogs.
3.RECONSTRUCTION OF A COMPOSITE SKIN SUBSTITUTE WITH COLLAGEN SPONGE AS DERMAL SCAFFOLD

To reconstruct a composite skin substitute composed of keratinocytes and collagen sponge as dermal scaffold. Collagen was extracted from fresh porcine skin. After being mixed with chondroitin sulfate, it was frozen and dried under vacuum to form a spongy membrane. Keratinocytes were separated from the foreskin with the routine method, and they seeded on the collagen membrane. The keratocytes which were carried by the collagen sponge were cultured, and the growth and proliferation were observed. The result showed that keratinocytes could grow and proliferate to form a confluent layer after 2~3 weeks. It indicated the the collagen sponge membrane showed no toxicity to human epidermal cells, and it could be used as a dermal scaffold in the construction of a composite skin substitute.
4.The use and effect of subcutaneous flap and tunica vaginalis flap technique in hypospadias repair and hypospadias fistulas repair

Objective To investigate the use and effect of subcutaneous flap and tunica vaginalis flap in hypospadias repair and hypospadias fistulas repair. Methods A total of 73 cases of hypospadias and hypospadias fistulas following surgery were included.The new urethras or closed fistulas were wrapped with various types of subcutaneous flaps or tunica vaginalis flaps.In the 45 cases of hypospadias,dorsal dartos flap technique was used in 27,tunica vaginalis flap technique in 3,and adjoining pedicle advance dartos flap technique in 15.In the 28 cases of urethral fistulas,dorsal dartos flap technique was used in 4,X-V skin flap technique in 14,and adjoining U-shaped subcutaneous flap technique in 10. Results All the cases were followed up for 3 to 36 months.Of the 45 cases of hupospadias, fistulas occurred in 3.Of the 28 cases of urethral fistulas, no fistulas occurred.The success rate of operation was 95.9%. Conclusions The use of these techniques can reduce the incidence rate of urethrocutaneous fistulas in hypospadias repair and hypospadias fistulas repair.Based on our experience these techniques are the optimal repair for classical patients with hypospadias and hypospadias fistulas.
5.Determination of Ginsenoside Rb_1,Ginsenoside Re and Ginsenoside Rg_1 in Yixin Fumai Granula by HPLC

OBJECTIVE: To determine Ginsenoside Rg1,Ginsenoside Re and Ginsenoside Rb1 in Yixin fumai granula by HPLC.METHODS: Samples were determined on a Diamond C18(150 mm?4.6 mm,5 ?m).The mobile phase consisted of accetonitrile-water by gradient elution with flow rate at 1 mL?min-1,UV detection wavelength at 203 nm,column temperature at 25 ℃ and sample size at 10 ?L.RESULTS: The linear ranges of Ginsenoside Rg1,Ginsenoside Re and Ginsenoside Rb1 were 0.645~6.450 ?g(r=0.999 8),0.54~5.40 ?g(r=0.999 7) and 0.605~6.050 ?g(r=0.999 9),respectively;the average recoveries were 100.59%(RSD=2.03%),98.70%(RSD=1.46%)and 98.99%(RSD=1.19%),respectively.CONCLUSION: The method is sensitive,simple and accurate,and it can be used for the quality control of Yixin fumai granula.
6.Dermal scaffold-experimental study on preparing and transplantation of spongiform collagen membrane

Objective: To prepare a collagen rnembrane and to investigate its feasibi1ity as a derma1 scaffold. Metbods:Pig hide collagen was dispersed in 0. 5 mol/L acetic acid, co-precipitated with chordroitin-6-sulfate and lyophilized for 48 h toproduce a highly porous membrane. Cross-linking was in 0. 25% glutaraldehyde for 24 h. The membrane was implanted be-neath the flaps on the dorsal sites of SD rats. After implantation, biopsies were obtained periodically to study the tissue com-patibility, vascularity and stability. Results: The average pore size of the spongiform collagen membrane was 100 pm. Sub-dermal implantation of it showed that the inflammation was minimal, with no evidence of an acute rejective reaction. Conclu-siou: The spongiform collagen membrane prepared is easy to secure and produce, it has satisfied tissue compatibility, abilityof vascularity and optimum dermal scaffold structure.
7.Effect of α-keto acid combined with low protein diet on kidney function, calcium-phosphorus metabolism and adenosine deaminase activity in malnourished patients with long term peritoneal dialysis

Objective To investigate the effect of α-keto acid combined with low protein diet on kidney function, calcium-phosphorus metabolism and adenosine deaminase activity in malnourished patients with long term peritoneal dialysis.Methods Forty-six cases malnourished patients with long term peritoneal dialysis were selected from May 2013 to August 2014 in the hospital and divided into two groups.The control group (n=23) were treated by conventional therapy, acceptable daily intake of protein 1.2 g/(kg? d).The experiment group (n=23) were treated by α-keto acid combined with low protein diet, acceptable daily intake of protein 0.8 g/(kg? d).Two groups were treated for three months.Serum calcium, phosphorus, nutritional status assessment, adenosine deaminase activity and renal function were observed and compared after treatment.ResuIts After twelve months ’ treatment, compared with control group, serum creatinine, blood urea nitrogen, serum uric acid of experiment group were lower ( P<0.05); serum calcium level was higher (P<0.05), serum phosphorus level was lower (P<0.05);ADA activity and white blood cell count were lower (P<0.05). The incidence of malnutrition rate had no significant difference between two groups(χ2 =3.136,P>0.05).ConcIusion α-keto acid combined with low protein diet could effectively increase serum calcium level, reduce serum phosphorus level, ADA activity and WBC count, and improve nutritional status in long-term peritoneal dialysis patients, which has important significance.

9. Chemical constituents in the mycelia of an acidoduric streptomycetes

Objective: To investigate the chemical constituents in the myceiia of an acidoduric streptomycetes. Methods: Multiple chromatographic methods were used to isolate and purify the constituents, and their chemical structures were identified by NMR, IR, and MS. The anti-inflammatory activities of four compounds were observed in rats with C II -A- induced toe swelling experiments. Results: Four compounds were identified: 8-(4-hydroxy-3, 5-di-methoxy phenyl)-2, 3- dimethoxy-1, 6-naphthalenediol (1), 7, 4′-dihydroxy isoflavone (2), 5, 7, 4'-trihydroxyisoflavone (3), and 3, 4, 5-trihydroxybenzoic acid (4). Compound of 2, 3, and 4 could inhibit C II-A-induced toe swelling in rats at 10 mg/(kg • d). Conclusion: Compound 1 is a novel compound, and other compounds have been isolated from the acidoduric Streptosporangium sp. for the first time. Compounds 2,3, and 4 possess anti-inflammatory activities.
10.Surgical excision and botulinum toxin A injection for vocal process granuloma.

OBJECTIVE:
To study the efficacy of treatment with microsurgery in combination with local injection of type A botulinum toxin for vocal process granuloma.
METHOD:
28 patients with vocal process granuloma received endotracheal intubation under general anesthesia. The lesion was removed with micro-scissor and CO2 laster under a self-retaining laryngoscope and microscope. The incision and mucous membrane surrounding the wound was closed with 8-0 absorbable suture. 4-point injection of botulinum toxin type A 8-15 u was then performed along the thyroarytenoid muscle and arytenoid muscle of the same side. Postoperative medication was administered based on disease causes.
RESULT:
All patients experienced vocal cord dyskinesia of the injected side 2-3 days after surgery. At 1 month after the surgery, wound healing was good in all the 28 patients, and the vocal cord movement was limited at the injected side. At 3 months, movement of the bilateral vocal cords was normal, and the vocal cord process mucosa was smooth. Patients were followed up for more than a year, and only one patient had recurrence in 2 months after surgery. The cure rate was 96. 4%.
CONCLUSION
Combination of laryngeal microsurgery and type A botulinum toxin local injection can shorten the treatment course of vocal process granuloma.
