1.Effect of Early Intervention on Oligodendrocyte-Myelin Glycoprotein mRNA Expression of Neonatal Rats with Brain Injury Caused by Intrauterine Infection

Objective To observe the effect of early intervention on the oligodendrocyte-myelin glycoprotein(OMgp) mRNA expression of brain injury neonatal rats caused by intrauterine infection.Methods 1.Twenty-eight Wistar pregnant rats were randomly divided into 2 groups:lipopolysaccharide(LPS)-treated group and saline control group.Pregnant rats were consecutively injected with LPS (450 ?g?kg-1) or saline on the 18th gestation age.After birth,the placentas were taken out and made HE staining to observe intrauterine infection.2.Thirty neonatal rats in the saline control group and 55 rats in the LPS-treated group were randomly selected which were divided into intervention group (n=25) and no-intervention group (n=25).The second post-natal day (P2) rats in intervention group were treated by early touch and enriched environment.The neonatal rats in the no-intervention group and saline control group were fed in a routine way.Five cases of P1 rats were selected respectively from the LPS-treated group and saline control group,and brain tissue pathological section was made to observe the condition of brain injury.3.Five cases of P1,P3,P7,P28 and P42 rats were selected from the saline control group,intervention group and no-intervention group to detect the OMgp mRNA expression levels by using the real time polymerase chain reaction me-thod.Results 1.There were a great number of neurophilic granulocytes in the placentas in the LPS-treated group.2.Brain tissue pathological of P1 in the saline control group had complete substantia alba structure,ordered disposition and lightly stained clear chromatospherite.While in the LPS-treated group, there existed brain tissue looseness,colloid cell aggregation and oligodendrocyte cytoreduction in the position of substantia alba,callositas and capsula interna.Intraventricular hemorrhage,substantia alba blood vessel dilatation and blood capillary angiorrhexis and hemorrhage could also be found.3.There was a higher increase in OMgp mRNA expressions of brain tissue in the LPS-treated group at P1,P3,P7,P28,P42 than those in the saline control group (Pa

Obesity is an established risk factor for endometrial cancer. Leptin, a secreted protein of the ob gene by white adipose tissue, plays an important role in the regulation of food intake and energy consumption in the brain and acts as a potential growth stimulator in normal and neoplastic cancer cells. However, a direct role for leptin in endometrial cancer has not been demonstrated. In the present study, the effect of leptin on the proliferation of Ishikawa endometrial cancer cells was investigated as well as the possible mechanism(s) underlying this action in endometrial cancers which express both short and long isoforms of leptin receptors. The expression of leptin receptor (ObRb) in Ishikawa cells was detected by RT-PCR and Western blotting. The cells after serum starvation, were treated by leptin with various concentrations (0, 10, 50, 100, 150 ng/mL) for different durations (6, 12, 24 h). The effect of leptin treatment on cell proliferation was examined by MTT assay. Meanwhile, inhibitory effect of Janus tyrosine kinase 2 (JAK2)/signal transducer and activator of transcription 3 (STAT3) inhibitor AG490 or extracellular signal-regulated kinase 1/2 (ERK1/2) inhibitor PD98059 on the proliferation of Ishikawa cells induced by leptin was also studied. Ishikawa cells were treated with 100 ng/mL leptin for various periods (0, 20, 40, 60 min), and the levels of STAT3 phosphorylation and ERK1/2 phosphorylation were examined by Western blotting. The results showed that leptin induced the phosphorylation of STAT3 and the activation of ERK1/2 in a time- and dose-dependent manner in the Ishikawa endometrial cancer cells. Blocking STAT3 phosphorylation with the inhibitor AG490, or blocking ERK1/2 activation by the specific ERK1/2 kinase inhibitor, PD98059, abolished leptin-induced proliferation of Ishikawa cells. In addition, leptin was found to potently induce the invasion of endometrial cancer cells in a Matrigel invasion assay. Leptin-stimulated invasion was effectively blocked by pharmacological inhibitors of STAT3 (AG490) and ERK1/2 kinase (PD98059). These results suggested that leptin promotes endometrial cancer growth and invasiveness by activating STAT3 and ERK1/2 signaling pathways and therefore blocking its action at the receptor level can be a rational therapeutic strategy.
3.125Ⅰ seed brachytherapy for recurrent salivary gland carcinoma after external radiotherapy.

OBJECTIVE:
To investigate the clinical application and efficacy of 125Ⅰ radioactive seeds implantation in the treatment of recurrent salivary gland carcinoma after external radiotherapy.
METHODS:
From July 2004 to July 2016, 43 cases of recurrent salivary gland carcinoma of the neck after external radiotherapy or surgery combined with external radiotherapy were treated. According to the conventional segmentation radiotherapy for head and neck cancer (once a day, 1.8-2.0 Gy each time, 5 days per week), the cumulative radiation dose of the patients in this group was calculated. In the study, 26 patients received 50-60 Gy, 7 patients received less than 50 Gy, 4 patients received 60-70 Gy, and 6 patients received more than 80 Gy (range: 80-120 Gy). The interval between the last external irradiation and local recurrence was 4-204 months, and the median interval was 48 months. Among them, 25 cases were treated with 125Ⅰ radioactive seeds implantation only and 18 cases were treated with 125Ⅰ radioactive seeds implantation after operation. The prescription dose was 100-140 Gy. The control rate, survival rate and disease-free survival rate were recorded to evaluate the side effects.
RESULTS:
The median follow-up time was 27 months (ranging from 2.5 to 149.0 months). Among them, the median follow-up time of adenoid cystic carcinoma patients was 31 months (range: 2.5-112.0 months), and the median follow-up time of mucoepidermoid carcinoma patients was 18 months (range: 5-149 months). The local control rates for 1, 3 and 5 years were 66.5%, 48.8% and 42.7%, respectively. The 1-, 3- and 5- year survival rates were 88.0%, 56.7% and 45.8%, respectively. The disease-free survival rates of 1, 3 and 5 years were 58.3%, 45.4% and 38.1%, respectively. There was no statistically significant difference in local control rate, survival rate, and disease-free survival between the radioactive seeds implantation group and the radioactive seeds implantation group after surgical resection. There were 2 cases of acute radiation reaction Ⅰ/Ⅱ and 3 cases of reaction Ⅲ or above. In the late stage of radiotherapy, there were 8 cases with Ⅰ/Ⅱ grade reaction and 3 cases with Ⅲ grade or above reaction. The incidence of radiation reactions of Grade Ⅲ and above was 7%.
CONCLUSION
125Ⅰ radioactive seeds implantation provides an alternative method for the treatment of recurrent salivary gland carcinoma after external radiotherapy. The local control rate and survival rate are improved on the premise of low incidence of side effects.

Objective To investigate the expression and function of integrin-iinked kinase (ILK) in the transdifferentiation process of primary tubular epithelial cell( TEC) and its effect on the accumulation of extracellular matrix (ECM) in the aging process. Methods Primary TEC was cultured from kidneys of male Wistar rats aged 3 and 24 months. Then the primary TEC was stimulated by TGF-? at the concentrations of 0, 5, 10 and 20-40/?g/L for 24 h. The TEC expressions of ?-SMA,ILK and F-actin were detected by immunocytochemistry or indirect immunofluorescence. FN or ILK protein expression levels were tested by Western-blot. DIG-labelled cRNA probe of rat ILK in situ hybridization was obtained by transcription in vitro, with which ILK expression level and the location of TEC were detected. Results ?-SMA expression levels were very low in young or aging rat TEC on the base situation, no significant difference was found between the two groups. However, ILK and F-actin levels in 24 months group were higher than in 3 months group. Along with increasing concentration of TGF-?, ?-SMA, ILK and F-actin levels were increased. Toxicity of different processed was evaluated Polygoni Multiflori Radix by determining the hepatotoxic potency for selecting processing technology. Process Polygoni Multiflori Radix using high pressure steamed, Black Bean high pressure steamed, atmospheric steamed for different time. Using normal human hepatocytes (L02) as evaluation model, hepatotoxic potency as index to evaluate hepatotoxic potency of different processed Polygoni Multiflori Radix. Analysis chemical composition of some processed products by UPLC-MS. Hepatotoxic bioassay method cloud evaluate the toxicity of different Polygoni Multiflori Radix samples. Different processing methods can reduce the toxicity of Polygoni Multiflori Radix, high pressure steamed three hours attenuated was better. Different processing methods have different effects on chemical constituents of Polygoni Multiflori Radix. Comparing with crude sample, the contents of gallic acid, 2,3,5,4-tetrahydroxyl diphenylethylene-2-O-glucoside, emodin-8-O-beta glucoside and emodin were decreased in processed products with 3 kinds of different methods. The change trend of 2,3,5,4-tetrahydroxyl diphenylethylene-2-O-glucoside content was similar with hepatotoxic potency. Different processing methods can reduce the toxicity of Polygoni Multiflori Radix. Processing methods and time attenuated obvious impact on toxicity. Recommended further research on the attehuated standard control of Polygoni Multiflori Radix concocted.

OBJECTIVETo construct eukaryotic expression vector of ribosomal protein sl5(RPs15) gene and study its effect on mouse skin fibroblasts in vitro.
METHODSThe RPs15 cDNA encoding region of fetal mouse skin was amplified by reverse transcription-polymerase chain reaction (RT-PCR) method and cloned into eukaryotic expression vector pcDNA3.1(-). The recombinant plasmid was transfected into adult mouse skin fibroblasts by FuGENE6 transfection reagents. Then the expression of RPs15 gene, was detected and its biological effect on fibroblasts was measured.
RESULTSThe DNA sequencing result of pcDNA3.1/RPs15 was identical with the reported. The RPs15 gene was expressed in transfected fibroblasts. The growth density of fibroblasts decreased with the conformation changing accordingly.
CONCLUSIONSThe eukaryotic expression vector pcDNA3.1/RPs15 is successively constructed and can be expressed in mouse skin fibroblasts. The results set up a basis for further study of the effect of RPs15 gene on skin fibroblasts.

8.Efficacy of three-dimensional conformal radiotherapy for 132 patients with esophageal carcinoma

Objective To evaluate the efficacy of three-dimensional conformal radiation therapy (3DCRT) for esophageal carcinoma and identify prognostic factors in this patient group.Methods From May 2002 to Jun 2005,132 patients with unresectable or inoperable esophageal cancer were treated with 3DCRT in our hospital.Their chnical data were analyzed retrospectively.ResultsThe 1-and 2-year local control rates was 65.4% and 52.1% in the whole group,respectively.The overall 1-and 2-year survival rate was 50.7% and 32.2% ,respectively.The median survival time was 13 months.The 1-and 2-year survival rate was 56.7%and 36.7% in stageⅠ + Ⅲ ,respectively,with 35.2% and 14.7% in stage IV.The median survival time were 15 months and 9 months for stage Ⅰ +Ⅲ and Ⅳ,respectively(x2 = 8.17,P = 0.004). Of patients with stage Ⅰ + Ⅲ disease who were absent of perforation sign before radiotherapy,with lesion length less than 8.0 cm and whole course given by 3DCRT,the 1-and 2-year survival rate was 73.0% and 49.9%,respectively.Univariate analysis revealed that condition of alimentation,absence of perforation sign, short lesion length,early TNM stage were associated with good survival.Multivariate analysis confirmed that absence of perforation sign and lesion length were independent prognostic factors for survival. Conclusions 3DCRT is effective for esophageal carcinomas in terms of survival and local control.Further improvement could be achieved with muhi-modality treatment.Absence of perforation sign and lesion length are independ ent prognostic factors for survival.

Objective To investigate the constituents of water and ethanol extracts from the stems and leaves ofAucklandia lappa and their effect on gastric emptying and intestinal propulsion of gastrointestinal normal,hyperthyroidism and inhibited mice.Methods The water,ethanol and aether petrolei extracts from stems and leaves of A.lappa were prepared,and the chemical constituents of the stems and leaves were identified by chemical reaction.The hyperthyroidism state was induced by neostigmine and inhibiting state was induced by atropine.Mice were ig administered with water and ethanol extracts (0.5 g/kg),with the improved phenol red method to determine gastric emptying and intestinal propulsion in gastrointestinal normal,hyperthyroidism and inhibited mice.Results Both water and ethanol extracts significantly improved intestinal propulsion in normal mice (P < 0.05 and 0.01),while significantly decreased the intestinal propulsion in hyperthyroidism mice (P < 0.05) and ethanol extract showed a stronger decreasing effect than that of water extract.The inhibitory effect of atropine on intestinal propulsion was intensified by these two extracts (P < 0.05).The restrained gastric emptying of normal,hyperthyroidism and inhibited mice was also observed.Chemical composition analysis indicated that many kinds of chemical components including protein,sugar,essential oil,flavonoids,lactones,alkaloids,saponins and tannins existed in the leaves and stems of A.lappa.Conclusion The leaves and stems of A.lappa could promote the intestinal propulsion of normal mice and restrain the intestinal propulsion of hyperthyroidism and inhibited mice while inhibit the gastric emptying in any condition tested,and its promoting effect on the gut may be related to the M cholinergic receptor.
10.One Hundred and Thirty-Six Cases of Incomplete Kawasaki Disease

Objective To investigate the clinical characteristics of incomplete Kawasaki disease(KD).Methods Clinical data includi-ng test results,therapeutic methods were analyzed retrospectively in 579 patients with Kawasaki disease.They were divided into classic KD and incomplete KD and made a compared analysis.Results There were no significant differences in gender,age,symptom and laboratory examination between classic and incomplete KD.But the rate of coronary artery lesions was higher in incomplete KD(18.4%) than that of classic KD(11%).Conclusion The rate of coronary artery lesions was higher in incomplete KD,and it should be paid more attention to earlier diagnosis and earlier treatment.
