Objective To assess the dlagnostic value of a triple stimulation technique(TST)for benign monomelic amyotrophy(BMA). Methods A total of 15 BMA patients and 15 healthy control subiects were exam-ined with the TST.The latent periods,amplitudes,areas and durations of the negative waves of the compound muscle action potential(CMAP)and motor evoked potential(MEP)induced by the stimulation were recorded,and the de-creasing ratio of amplitudes and areas in the two groups were analyzed. Results There were significant differences in amplitude and area ratios between the BMA patients and the healthy control subjects.Both ratios were significantly lower in the BMA patients. Conclusion TST has some value in diagnosis of BMA.

Objective To explore the association of chemokines and their receptors with immunologi- cal abnormality in newly diagnosed systemic lupus erythematosus(SLE) patients.Methods The serum con- centration of MIP-1?,MIP-1?,RANTES,IFN-?IL-4 were measured by enzyme-linked immunoabsorbent assay (ELISA) in 37 newly diagnosed.SLE patients and 20 normal controls.The expression rate of CCR1, CCR3,CCR5 on CD4~+T cells were detected by flow cytometry in 18 SLE patients and 10 normal controls.Re- suits Serum MIP-1?,MIP-1?concentrations were significantly higher in SLE patients than in normal control group (P＜0.01),the concentration of MIP-1?positively correlated with MIP-1?(r=0.609,P＜0.01);the per- centage of CD4~+CCR1~+ and CD4~+CCR5~+ cell were significantly lower in newly diagnosed SLE patients than in normal control group (both P＜0.01),the percentage of CD4~+CCRI~+ cells correlated negatively with the level of serum MIP-1?and IFN-?r=-0.525,P=-0.017;r=-0.442,P=0.045);the percentage of CD4~+CCR5~+ cell corre- lated negatively with the level of serum IFN-?(r=-0.645,P=0.001);the ratios of CD4~+CCR3~+/CD4~+CCR5~+ was significantly higher in newly diagnosed SLE patients than in the normal control group (P＜0.01).Conclusion Abnormal change and interaction of chemokines and their receptors with cytokines lead to immunologic dys- function and may participate in the initiation of SLE.

Kidney injury and decreased chemosensitivity of tumor cells are obstacles with cisplatin (CDDP) chemotherapy. Down-regulation of the organic cation transporter 2 (OCT2) and multidrug resistance-associated protein 2 (MRP2) is a key means to alleviate CDDP-induced kidney injury and increase chemosensitivity. Astragaloside IV (AS IV) is obtained from the well-known traditional Chinese herb Astragalus membranaceus. This study explored the role of AS IV in preventing kidney injury and enhancing the antitumor effect of CDDP by suppressing OCT2 expression in kidney and MRP2 in tumors. This project was reviewed and approved by the Animal Ethics Committee of the First Hospital of Jilin University. The effects of AS IV on CDDP inhibition of tumor growth and promotion of apoptosis were assessed in Lewis lung tumor (LLC)-bearing mice by H&E and TUNEL staining. Kidney injury was assessed by serum biochemical parameters and H&E staining. We used Western blotting and immunohistochemistry assays to detect OCT2 and MRP2 expression in kidney and tumor. The concentration of CDDP in kidney and tumor was measured by HPLC-MS/MS. AS IV enhanced CDDP chemosensitivity by increasing tumor cell apoptosis and slowing tumor growth, and decreased kidney injury as evidenced by lower blood creatinine (Cr) and blood urea nitrogen (BUN). Co-administration of AS IV suppressed MRP2 overexpression induced by CDDP in tumor tissues and may be an important mechanism for enhancing CDDP chemosensitivity. Moreover, AS IV reduced CDDP-induced kidney injury in mice along with suppression of OCT2 expression in kidney. The concentration of CDDP was increased in tumor but decreased in kidney. In total, AS IV not only enhanced the antitumor effect of CDDP by suppressing MRP2 expression in tumor cells, but also decreased kidney injury induced by CDDP. The results provide new insight into the combined use of a chemotherapy drug and natural ingredients to treat cancer.

ObjectiveTo observe the effect of dressing and moxibustion-pretreatment on the local joint swelling and stress hormone in hypothalami of rats with adjuvant arthritis (AA) at early and secondary stages.MethodsForty Wistar rats were randomly divided into 5 groups: normal group, early and secondary model groups, early and secondary pre-dressing and moxibustion (PDM) groups. The dressing with Chinese herb and moxibustion was stuck on Dazhui point (GV14) before the AA model established. The effects of dressing and moxibustion-pretreatment on the feet swelling and corticotropin-releasing-hormone (CRH), beta-endorphin (β-EP) and neuropeptide-Y (NPY) in hypothalami were observed.ResultsThe right feet swelling rate at early and secondary stages obviously increased after modeling ( P<0.01), and it became lower in early and secondary PDM groups than in model groups at the same phases ( P<0.05～0.01). The level of hypothalamic CRH was higher after modeling ( P<0.05～0.01), compared with early model group, it had a tendency to going down in early PDM group, moreover, in secondary PDM group the level was similar with the normal group. The level of hypothalamic β-EP increased significantly in early model group ( P<0.01), and lightly changed in secondary model group, it decreased in early PDM group but increased in secondary PDM group, compared with model groups at the same stages ( P<0.05). The level of hypothalamic NPY increased significantly after modeling, and it declined in the secondary PDM group ( P<0.05).ConclusionDressing and moxibustion-pretreatment can relief feet swelling of AA rats, which may be related with its regulative effect on the level of hypothalamic CRH, β-EP and NPY.

OBJECTIVETo explore Mechanisms of electroacupuncture in anti-inflammation and immunoregulation, and to compare differences among "Dazhui" (GV 14), "Mingmen "(GV 4) and non-acupoint in these actions.

METHODSAdjuvant arthritis rat model was made. The rats were divided into 5 groups: normal group, model group, "Dazhui" (GV 14) electroacupuncture group, "Mingmen" (GV 4) electroacupuncture group and non-acupoint electroacupuncture group. Effects of electroacupuncture on local joint inflammation and corticotropin-releasing hormone (CRH), beta-endorphin (betaEP) and interleukin-2 (IL-2) contents in the hypothalamus were observed and differences of the actions among different points were compared.

RESULTSCRH content in the hypothalamus in the "Dazhui" (GV 14) electroacupuncture group decreased as compared with the model group (P < 0.05); there was no significant change in beta-EP and IL-2 contents of the hypothalamus in the 3 electroacupuncture groups as compared with the model group (P > 0.05); there was positive correlation between CRH and IL-2 contents, and between IL-2 and beta-EP contents in the hypothalamus. The swelling rate of the toe in the "Dazhui" (GV 14) group and the "Mingmen" (GV 4) group was lower than that in the non-acupoint group (P < 0 05).

CONCLUSIONElectroacupuncture exerts anti-inflammation and immunoregulation possibly via regulation of CRH, IL-2 and beta-EP each other, and the anti-inflammation action of "Dazhui" (GV 14) and "Mingmen" (GV 4) is stronger than that of non-acupoint.

OBJECTIVECeramic brackets debonding by Nd:YAG laser is based on the thermal effect of laser, which may cause injury of the pulp tissue. In this study, the histological changes of pulp tissue that subjected to Nd: YAG laser irradiation with different power and time were observed.

METHODS20 New Zealand rabbits were included in this study. Ceramic brackets were bonded to the 4 incisors as routine. The ceramic brackets of left upper teeth that debonded mechanically were used as control group, while the brackets of right upper, left lower and right lower incisors were debonded by laser with 3 W 3 s (group A), 2 W 5 s (group B) and 5 W 2 s (group C) energies, respectively. The teeth were pulled out at 5 minutes, 1 day, 3 days, 1 week and 1 month after the debonding operations. Slides prepared from the pulp tissues of the debonded teeth were used to evaluate the injury of laser.

RESULTSIn comparison with the control group, pulp tissue of teeth that exposed to laser with different energy for 5 minutes showed mild capillary dilation. One day later, group A, B and C showed moderate capillary dilation, and group C also showed moderate infiltration. At 3 days, inflammation was disappeared in group B, whereas capillary dilation was found in group A. Hemorrhage and inflammation cells infiltration were found in group C. At 1 week, alleviation of capillary dilation was found in group A but not in group C. One month later, inflammation disappeared in group A, while pulp tissue in group C showed mild edema and capillary dilation.

CONCLUSIONNd:YAG laser of high energy may cause injury of the pulp tissue during debonding. Laser energy of 3 W 3 s could effectively debond ceramic brackets without irreversible pulp injury.

Comminuted fracture of distal femur is a common lower limb injury from traffic accidents, especially from motor accidents. Routine dynamic condylar screw (DCS) or 95-degree condylar plate (CP) sometimes cannot solve the bone defect in the center of alignment and contralateral diaphysis for the reason of absent screw anchor point, especially for AO C2.2-2.3 types. Many authors recommended open reduction and fixation with less invasive stabilization system (LISS) as the treatment of choice, but there are still problems in fusion and alignment. In this study, we reported our experiences with the use of bone splint technique in the high-energy commimuted fracture of distal femur with central and medial bone defect in adolescents.

OBJECTIVETo evaluate the pre-clinical effect of YJ-XCC1Z3 on the treatment of depression with the mice mouse.

METHODYJ-XCC1Z3 was administered at the dose of 405 mg x kg(-1) and 135 mg x kg(-1) to observe the locomotor activity with the mouse locomotor activity recorder apparatus, to observe the effect of YJ-XCC1Z3 on the duration of immohility in the mouse forced swimming test and tail suspension test, to observe the effect of YJ-XCC1Z3 on the body temperature and the metabolism of monoamine neurotransmitters in mouse brain in the mouse model of reserpine induced hypothermia, and to observe the effect of YJ-XCC1 Z3 on the times of 5-HTP induced head-twitches in mice.

RESULTThere were no significant changes in the locomotor activity, but a significant reduction in the immobility time was observed in the mice treated with YJ-XCC1Z3 405 mg x kg(-1) and imipramine in the forced swimming test and the tail suspension test. YJ-XCC1Z3 135 mg x kg(-1) and 405 mg x kg(-1) could improve the range of reserpine induced hypothermia in mice, and the latter could also enhance the times of 5-HTP induced head-twitches in mice. YJ-XCC1Z3 405 mg x kg(-1) and 135 mg x kg(-1) could increase the content of 5-HT and NE and decrease the ratio of 5-HIAA/5-HT in mouse brain, but the dose of 405 mg x kg(-1) could decrease the content of DA. The dose of 405 mg x kg(-1) could increase the content of 5-HIAA and had no obvious effect on the content of HVA and DOPAC.

CONCLUSIONYJ-XCC1Z3 shows potent antidepressant effect by improving the behaviour of the mouse in depression and not inducing hyperlocomotion in the mice. This effect results in the increase of the content of 5-HT and NE in the mouse brain. YJ-XCC1Z3 can decrease the metabolism of 5-HT to effect the content of 5-HT.

Objective:To evaluate the 5-year survival outcome of patients with unresectable locally advanced non-small cell lung cancer (NSCLC) treated with Endostar in combination with platinum-based concurrent chemoradiotherapy.Methods:From March 2009 to June 2015, 115 patients with the unresectable locally advanced NSCLC from two prospective studies[Clinical trials 2009-2012(ClinicalTrials.gov NCT01894) and 2012-2015(ClinicalTrials.gov, NCT01733589)] were treated with Endostar in combination with platinum-based concurrent chemoradiotherapy. A total dose of 60-66 Gy was delivered in 30-33 fractions. Endostar was given 1 week prior to the beginning of radiotherapy, and repeated fortnightly during the concurrent chemoradiotherapy. After long-term follow up, survival outcome was evaluated in 104 patients treated with radiation dose of ≥60 Gy. Kaplan-Meier method was used for survival analysis. Univariate survival analysis was performed using the log-rank test.Results:Of 104 eligible patients, 60.6% of them had squamous carcinoma and 65.4% were classified in stage Ⅲ B. All the patients received ≥2 cycles of Endostar and 93.3% of them received 4 cycles of Endostar. The median follow-up time was 68.3 months. The median overall survival (OS) and median progression-free survival (PFS) were 31.3 and 13.9 months, respectively. The 3-year and 5-year OS were 45.6% and 35.7%, respectively. The 3-year and 5-year PFS were 27.1% and 24.9%, respectively. Univariate analysis indicated that sex, ECOG, pathological type, clinical stage, radiotherapy technique, chemotherapy regimen, chemotherapy cycle and cycle of Endostar use were not associated with OS. Late radiation injury occurred in 14.4% of patients, and no grade 4-5 late injury was observed. Conclusion:Patients with unresectable locally advanced NSCLC treated with Endostar fortnightly in combination with platinum-based concurrent chemoradiotherapy achieve better OS than historical data with tolerable toxicities.
