3.Effect of Manzhi Kechuan Ling on Pulmonary Function and Quality of Life of Patients with Chronic Obstructive Pulmonary Disease at Stationary Stage

Objective To observe the effect of Manzhi Kechuan Ling(MKL)on pulmonary function and quality of life (QOL)of patients with chronic obstructive pulmonary disease (COPD)at stationary stage.Methods Seventy-six COPD patients were randomized into two groups.The treatment group (N=40)received oral use of MKL (mainly composed of Radix Ginseng,Radix Astragali,Rhizoma Atractylodis Macrocephalae,Colla Cornus Cervi,Placenta Hominis,Gecko,Semen Juglandis,Gecko,Fructus Perillae,Bulbus Fritillariae Cirrhosae,Rhizoma Pinelliae)for 6 months,and was followed up for another 6 months after suspension of medication.The control group (N=36)did not receive any medication.The pulmonary function,TCM syndrome scores,QOL scores and annual acute attack times were examined before treatment,6 and 12 months after treatment.Meanwhile,artery blood gas analysis was performed before treatment and 6 months after treatment.Results Inthe treatment group,the pulmonary function indexes of forced expiratory volume in one second (FEV1)andforced vital capacity (FVC)were improved 6 months after treatment (P0.05).In the control group,FEV1 and FVC presented a decreasing trend 6 and 12 months after treatment,and FEV1 and FCV 12 months after treatment differed from those before treatment (P0.05).There showed statistical differences of PaO2 and PaCO2 between the two groups 6 months after treatment(P

Objective To investigate the therapeutic effect of Shouling Jiannao Capsule (SJC)for the treatment of multiple infarction dementia (MID),and to observe the effect of SJC on blood lipid level and hemorrheology.Methods Fifty MID patients aged 45~80 subjecting to the diagnostic standards were randomly divided into two groups:treatment group (N=30)received SJC (mainly composed of Radix Polygoni Multiflori,Ganoderma Lucidum,Radix Notoginseng,Radix Salviae Miltiorrhizae and Radix Curcumae),and control group (N=20)received dihydroergotoxine.The treatment lasted 2 months.The therapeutic effect in both groups was evaluated after treatment,and the changes of scores of TCM syndromes,Hasegava Dementia Scale(HDS),and Mini-mental State Examination (MMSE)were observed before and after treatment.Meanwhile,blood lipid level,hemorheological indexes,erythrocyte superoxide dismutase(SOD),plasma lipid peroxide(LPO)and transcranial color Doppler(TCD)were examined and statistically analyzed before and after treatment in both groups.Results After treatment,the total effective rate was 90.0% in the treatment group and 55.0% in the control group,the difference being significant(P
5. Metabonomic study of metformin in type II diabetic rats

Aim To study the mechanism of metformin in the treatment of type 2 diabetic melitus (T2DM) rats. Methods Rats were randomly divided into normal group, model group and metformin group with 10 rats in each group. Four weeks after induction with HFSD, 35 mg 'kg-1 STZ was injected intramuscularly, and the experiment was completed after 8 weeks of administration. UPLC/ESI-TOF-MS was used to study the effects of metformin on metabolites in serum of T2DM rats, and qPCR was used to find its target. Results Compared with normal group, the body weight, pancreatic index and insulin level in model group decreased significantly (P < 0. 05) , liver index, GLU, TC, TG and FFA levels increased significantly (P < 0.01), OGTT was significantly impaired, while met-formin could significantly increase body weight, pancreatic index and insulin level (P <0. 05) , reduce liver index, GLU, TC, TG and FFA levels (P < 0. 05) , and improve OGTT. After metahonomics, 17 biomark-ers were obtained. After verification, metformin was found to regulate the synthesis and metabolism of cholesterol, fatty acids, bile acids and phospholipids. Conclusions Metformin can improve the lipid metabolism disorder of T2DM by regulating the synthesis and metabolism of cholesterol, fatty acids, bile acids and phospholipids.
6.Effect of pretreatment by solid-state fermentation of sawdust on the pelletization and pellet's properties.

We pretreated sawdust (Castanopsis fissa Rehd.et Wils) by solid state fermentation (SSF) with Phanerochaete chrysosporium, and then compressed it into pellets with the moisture content of 15% and the pressure of 98 MPa, to solve the problem of low density, low Meyer hardness, high water uptake, and short storage period of pellet in the woody pellet industry. We studied the effects of fermentation time on pelletization and pellets's characteristics (including energy consumption, density, Meyer hardness, and hydrophobicity). SSF affected the heating values of pellet. Compared with fresh sawdust, SSF consumed more energy at the maximal value by 6.98% but saved extrusion energy by 32.19% at the maximum. Meanwhile, SSF could improve the density, Meyer hardness and hydrophobicity of pellet. Pellet made of sawdust pretreated by SSF for 48 d had best quality, beneficial for long-term transportation and storage of pellets.

7.Expression of PED/PEA-15 and XIAP in prostate cancer cells and their effects on prostate cancer cell (PC-3) apoptosis

Objective To study the effect of antiapoptosis factors PED/PEA-15 and XIAP on prostate cancer cells(PC-3)apoptosis.Methods The expressions of XIAP and PED/PEA-15 in prostate cancer cells(PC-3)were respectively assayed using the RT-PCR technique.XIAP and PED/ PEA-15 specific siRNA vectors were designed and constructed and then were transiently cotransfected into PC-3 cells under induction of liposome.The effects of siRNA vectors on PED/PEA-15 and XIAP transcription were assayed by RT-PCR technique,and the effect of XIAP and PED/PEA-15 on cancer cell apoptosis were determined by flow cytometry and microscope observation.Results PED/PEA- 15 and XIAP were both highly expressed in PC-3 cells.Enzyme digestion analysis and DNA sequencing confirmed that the PED/PEA-15 and XIAP-specific siRNA expression vectors were constructed successfully.The designed siRNA sequences of PED/PEA-15 and XIAP could specifically inhibit their transcription.The PC-3 cells which were cotransfected with PED/PEA-15 and XIAP- specific siRNA vectors were more sensitive to doxorubicin.The apoptosis rate of cotransfected cells was significantly increased.Conclusions PED/PEA-15 and XIAP might be involved in the development of prostate cancer.
8.Expression of sterol regulatory element-binding protein-2 in the process of chondrocyte degeneration

BACKGROUND: Recent studies have demonstrated that sterol regulatory element binding protein-2 (SREBP-2) plays a key role in osteoarthritis, but its exact pathogenesis remains incompletely understood yet. OBJECTIVE:To investigate the expression of SREBP-2 in the process of interleukin-1β-induced articular chondrocyte degenerationin vitro. METHODS: Articular chondrocytes obtained from C57BL/6J mice were culturedin vitro. After the second passage, cels were randomly divided into four groups: control group, and three experimental groups treated with 10 μg/L interleukin-1β for 24, 48 and 72 hours, respectively. RESULTS AND CONCLUSION:The cels became hypertrophic after being stimulated by interleukin-1β, and the staining of colagen X was positive at 72 hours. MTT assay demonstrated that the cel activity after stimulation with interleukin-1β decreased with time. Results of RT-PCR showed that the expression of SREBP-2 and SREBP cleavage activating protein mRNA was significantly increased after stimulation with interleukin-1βas compared with the control group and increased with time. On the contrary, the expression of aggrecan and colagen II mRNA was decreased with time. It is revealed that interleukin-1β could inhibit the proliferation of regular chondrocytes and the expression of its extracelular matrix, and furthermore, induce chondrocyte hypertrophy. The expression of SREBP-2 showed a negative relationship with key cartilage genes during this interleukin-1β-induced degeneration.
9.Advances on pathogenesis research of acute myeloid leukemia with t(8;21)-- review.

Acquired chromosomal translocations can be identified in nearly 50% of human acute myeloid leukemias. The common chromosomal translocation in this disease is t(8;21) (q22;q22). It involves the aml1 (runx1) gene on chromosome 21 and the eto (mtg8, runx1t1) gene on chromosome 8 generating the aml1/eto fusion gene. An initial model for its pathogenesis emphasized the conversion of a hematopoietic transcriptional activator AML1 into a leukemogenic repressor which blocked myeloid differentiation at the level of target gene regulation. Aml1/eto fusion genes inhibit key hematopoietic transcription factor that function as tumor suppressors at several nodal point during hematopoietic differentiation. A new model is presented in which aml1/eto coordinates expansion of the stem cell compartment with diminished lineage commitment and with genome instability. In this review, the molecular role of aml1/eto fusion gene and his transcribed isoforms in regulating stem renewal, blocking hematopoietic differentiation and interacting with various lineage-specific transcription factors are summarized.
