Background and Purpose:Recent studies have shown that survivin is an anti-apoptosis gene,which is involved in carcinogenesis and drug resistance of leukemia.Antisense oligodeoxynucleotide(ASODN) can be used to inhibit the expression of survivin,inducing apoptosis and enhancing the chemosensitivity of leukemic cells.This study was designed to explore the effect of survivin ASODN on the growth,apoptosis,and caspase-3 activity of leukemia cell line K562/A02 with the phenotype of drug resistance.Methods:Survivin ASOND was transfected to K562/A02 cells by liposomal reagent,The rate of inhibition,expression of survivin mRNA,apoptosis,and activity of Caspase-3 were detected by colormetric MTT,RT-PCR,flow cytometry and fluorometer,respectively.Results:Survivin ASODN could inhibite the cell proliferation in a dose and time dependent manner.Compared with controls,expression of survivin mRNA decreased by 36.2%(P
3.Clinical study on nutritional support for severe acute pancreatitis

Objective To explore the effect of enteral nutrition combined with parenteral nutrition support on the prognosis of severe acute pancreatitis(SAP).Methods Sixty-one patients diagnosed with SAP in our hospital from July 2011 to June 2012 were chosen and randomly divided into EN and PN group(30 cases)and PN group(31 cases).At the early stage of treatment,the former group was provided parenteral nutrition(PN)alone while the latter group was given enteral nutrition together with parenter-al nutrition.Then we measured the patients′body mass index on the 1st,2nd,3rd,5th,7th,10th,14th day respectively after admis-sion,and recorded the following indexes:whether complications occurred or not,the time for the first passage of gas by anus and defecation,the total time and cost of hospitalization,the drug proportion and the time the serum and urine amylase returned to nor-mal.Results The two groups of patients′body mass index showed no significant differences in the first 3 days,but there were sig-nificant differences in statistics on the 5th,7th,10th and 14th day(P <0.05).In addition,the two groups of patients formed a sharp contrast in such aspects as the bowel sound recovery time,the anal exhaust time,the total cost of hospitalization and the incidence rate of the complications(P <0.05)whereas there were no remarkable differences in the serum and urine amylase recovery time and the total time of hospitalization(P >0.05).Conclusion Compared with PN,the combined therapy of EN and PN could improve the patients′nutrition,reduce the weight loss caused by consumption,promote intestinal peristalsis,effectively reduce infectious compli-cations caused by floraectopic and ease the patients financial burden.
5.Effects of combination of maternal hydration and hands and knees posture on nursing of breech pregnancy

Objective To compare the effects of combination of maternal hydration and hands and knees posture with simple hands and knees posture on correction of breech pregnancy. Methods One hundred patients who were diagnosed breech pregnancy from November 2015 to November 2016 were assigned to experimental group and control group with 50 cases each according to individual willingness. The patients in experimental group received the combination of maternal hydration and hands and knees posture;while the patients in control group received simple hands and knees posture. Results Amniotic fluid index was (18.94±2.44)cm in experimental group and (15.97±2.33)cm in control group, the difference had statistic significance (t=6.239, P<0.05). The effective transfer rate and successfully used time was 92%(46/50) , (2.10 ± 0.75) weeks in experimental group and 64%(32/50), (4.27 ± 0.98) weeks in control group, the difference had statistic significance (χ2=11.422, P<0.05; t=-12.463, P<0.05). For primipara and multipara, the effective transfer rate was 90.91%(30/33), 16/17 in experimental group, and 68.75%(22/32), 10/18 in control group, the difference had statistic significance (χ2=4.986, 6.806, all P<0.05). The rate of vaginal and cesarean section deliveries was 86%(43/50), 14%(7/50) in experimental group, and 56%(28/50), 44%(22/50) in control group, the difference had statistic significance(χ2=10.928, P=0.001). Patients who had fetal movement or who didn′t have time to do correction or missed was 17.10%(46/269), 1.49%(4/269) in experimental group, and 12.55% (22/255), 7.06% (18/255) in control group. Conclusions @Therapic effects of combination of maternal hydration and hands and knees posture is much better than that of the simple hands and knees posture therapy on correction of breech pregnancy, which is worthy of popularization and application in clinic.

9.Prospective efficacy comparison between the two-cuff swan neck catheter and the Tenckhoff catheter in peritoneal dialysis patients

Objective To compare the efficacy between the two-cuff swan neck catheter and the Tenckhoff catheter in continuous ambulatory peritoneal dialysis (CAPD) patients prospectively. Methods One hundred and ten patients with end-stage renal disease (ESRD) were selected as candidates, who received catheter implantation and CAPD therapy for the first time. Patients were divided into group A (swan neck catheter group) and group B (Tenckhoff catheter group), 55 patients for each group. Catheters of beth groups had a straight end and were implanted by routine surgical procedure. One-year follow-up was performed and information was recorded such as complications, survival time, quit of dialysis, death, etc. Survival analysis was carried out by Kaplan-Meier method and Log-Rank tests. Results At the end of follow-up, 17 patients died, 3 received renal transplantation, 8 were transferred to hemodialysis, 3 went to other hospitals, and 79 patients (71.8%) remained in our department for CAPD. Twenty-six patients of both groups had peritonitis with a total of 35 occurrences. The total incidence of peritonitis was 0.32 times/patient year, with the detailed figure of 0.35 times/patient year for group A and 0.29 times/patient year for group B respectively (P0.05). The time interval between the catheter implanting and the onset of peritonitis was (30±29) weeks and (29±24) weeks for group A and group B respectively (P0.05). The risk of developing peritonitis in both groups was 26.97% within 1 year. Tunnel infection occurred in 2 patients and exit-site infections in 9 patients of two groups. The incidence of tunnel plus exit-site infections was 0.1 times/patient year. Incidence of tunnel infection and the exit-site infection for group A was lower than that of group B (0 vs 0.036 times/patient year and 0.06 times/patient year vs 0.11 times/patient year respectively). However, the difference was not significant (P0.05). Mechanical complications of catheter (catheter migration, omcntum wrapping, leakage of peritoneal dialysates, slip out of outer cuff), incidence of inguinal hernia and bellyache between two groups were not significantly different (P0.05). There were 4 cases of catheter drawing in each group. Both two groups had the same 12-month technical survival rate as 92.73%. Of 17 dead cases, 7 were in group A and 10 in group B (P0.05). The main death causes were cardiocerebral events (47.1%) and infections (23.5%). The 12-month survival rate was 86.34% for group A and 80.68% for group B (P0.05). Conclusions There are no significant differences of infection, mechanical complications, technical survival rate and patients' survival rate between two groups. The efficacy of swan-neck catheter is similar to Tenckhoff catheter in CAPD patients.
10.Clinical research on plasma and urine neutrophil gelatinase-associated lipocalin of lupus nephritis

Objective To study the clinical significance ofneutrophil gelatinase-associated lipocalin (NGAL), which in serial plasma and urine samples was measured in participants with lupus nephritis (LN)and healthy persons. Methods NGAL in serial plasma and urine samples was measured by enzyme-linked immunosorbent assay (EL.ISA) in 35 patients with LN by 1997 ACR systemic lupus erythematosus (SLE)standard with varied degree of kidney damage and 30 healthy persons with matching sex and age in physical examination center. Disease activity was measured by the SLE disease activity index (SLEDAI-2K),and 35LN patients were classified in active group and (25 cases) non-active group (10 cases) according to the SLEDAI-2K. Results Urinary NGAL were significantly increased in LN patients [(78.94 ± 81.97) μg/L]compared with healthy persons[(28.50 ± 18.08) μ g/L] (P = 0.002). And urinary NGAL were significantly increased in active group [(92.90 ± 94.88) μg/L] compared with non-active group [(48.20 ± 24.77)μ g/L] (P = 0.049). NGAL in serial plasma had no statistically significant difference between active group and non-active group (P >0.05). Conclusions NGAL in urine but not in plasma represents a novel biomarker for renal disease activity in LN. The increase might be related to renal tubule pathological changes.
