3.Relationship between CD117 expression and chemotherapy effect on the patients with acute nonlymphoblastic leukemia

Background and Purpose:It has been proved that CD117 may be used as an immunology marker for diagnosis of leukemia of myeloid origin.The relationship between CD117 expression and effect of chemotherapy on the patients with Acute Nonlymphoblastic Leukemia(ANLL) remains unclear.This study is to investigate the relationship between CD117 expression and the response of patients with ANLL to chemotherapy.Methods:Flow cytometery(FCM) was used to detect the positive rate and the levels of CD117 expression of the bone marrow mononuclear cell(BMMNC) from 38 patients with acute lymphoblastic leukemia(ALL) and 81 patients with ANLL,respectively.All-trans Retinoic Acid(ATRA) was taken to treat M_(3) type of ANLL and protocol DA and/or HA was used to treat the other types.ANLL was divided into two groups: positive(+) and negative(-) expression of CD117.At the same time we compared the difference of rates of complete remission(CR) between CD117(+) and CD117(-) groups from ANLL after chemotherapy.Results:Positive percentage of expression of CD117 in ALL and ANLL groups were 13% and 70% respectively(P=0.000).Positive levels of CD117 decreased successively as follows: M_(3)/ M_(1)、M_(2)/ M_(6) / M_(4)、M_(5).CR rates of CD117(+) and CD117(-) groups of ANLL after chemotherapy were 51%(29/57) and 67%(16/24)(P=0.192),respectively.Conclusions:CD117 may serve as an immunology marker for the diagnosis of ANLL,but positive or negative expression of CD117 in ANLL was not associated with the response of the patients with ANLL to chemotherapy.
4.The clinic significance of CD117/CD34 co-expression in the patients with acute promyelocytic leukemia

Objective:To study expressions of CD117 and CD34 in the patients with acute promyelocytic leukemia(APL,M3) and in M1-M2 subtype from FAB classification for acute leukemia(AL).The focus of the study would be laid on the clinical significance of CD117/CD34 co-expression in the patients of M3 subtype.Methods:Researched cases of acute nonlymphoblastic leukemia(ANLL) were divided into two groups:M1-M2 subtype and M3 subtype.Flow cytometery(FCM) was used to detect the rates of positive expression of CD117 and CD34 on bone marrow mononuclear cell(BMMNC) in 54 patients of M3 and 63 patients of M1-M2 subtype respectively.Meanwhile,we compared the differences between the rates of expression of CD117 and CD 34.And,the rates of CD117/CD34 co-expression in patients of M1-M2 subtype and M3 subtype were studied.Results:Our results revealed that the positive rates of CD117 expression in M1-M2 subtype and M3 subtype were 71.4%(45/63) and 66.7%(36/54) respectively(P=0.58).The positive rates of CD34 expression in M1-M2 subtype and M3 subtype were 66.7%(42/63) and 11.1%(6/54) respectively(P=0.000).The positive rates of CD117/CD34 co-expression in M1-M2 subtype and M3 subtype were 71.1%(45/63) and 7.4%(4/54) respectively(P=0.000).Conclusion:CD117 may be used as immunology marker for leukemia of myeloid origin.CD34 had lower expression in M3 subtype than in M1-M2 subtype.The positive rate of CD117/CD34 co-expression in M3 subtype was significantly lower than that in M1-M2 subtype,which can help for diagnosis of M3 subtype and help differentiate M3 subtype from M1-M2 subtype as well.
5.Simultaneous Determination of 2 Constituents in Compound Chloramphenicol Auristillae by HPLC

OBJECTIVE:To establish a HPLC method for the determination of both chloramphenicol and furacillin in compound chloramphenicol auristillae.METHODS:The determination was performed on Tianhe Kromasil C18 column,the mobile phase consisted of methanol-water(60∶40) with flow speed at 1.0ml/min,detection wavelength at 278nm,column temperature at 25℃,and sample size at 20?l.The dexamethasone was used as internal standard.RESULTS:The linear ranges for chloramphenicol and furacillin were 31.75~127?g/ml and 26.25~105?g/ml,respectively;The average recovery rates were 100.4%(RSD=0.89%) and 100.7%(RSD=1.20%),respectively.CONCLUSION:This method is simple in operation,accurate and reliable in determination results and it can be used for the quality control of compound chloramphenicol auristillae.
6.Correlation of head nurse's authentic leadership and degree of solidarity among nurses in operating room

Objective To investigate the relationship between head nurse's authentic leadership and degree of solidarity among nurses in operating room.Method A total of 120 nurses in the operation room of 3 general hospitals in Jiamusi City were enrolled in the study by way of the questionnaire on authentic leadership and the scale of solidarity.Results The mean score on the head nurse's authentic leadership (41.28 ± 8.48) was above the middle-high level.The mean score on the solidarity (68.67 ± 8.82) was in the middle-high level.There was a positive correlation between the head nurses' authentic leadership and the degree of solidarity among nurses in the operating room.Conclusion Hospital and nursing administrators should take active and effective measures to improve the leadership of head nurses so as to improve the degree of solidarity among nurses in the operating room as well as the quality of nursing.
8.Effect of Flurbiprofen Axetil Combined with Fentanyl in Postoperative Patient-controlled Analgesia after Craniotomy

Objective To investigate the effect and side effects of flurbiprofen axetil combined with fentanyl in postoperative patient-controlled analgesia (PCA) after craniotomy.Methods 60 patients undergoing scheduled craniotomy were randomly divided into the treatment group and control group with 30 cases in each group, and received flurbiprofen axetil combined with fentanyl and only fentanyl respectively in PCA after craniotomy. The visual analogue scale (VAS), sedation score (SS) and the side effects were evaluated in 24 h after craniotomy.Results There was no significant difference in VAS between the two groups ( P>0.05). The SS, side effects such as nausea and vomiting in the treatment group were significantly less than that in the control group ( P<0.05).Conclusion Flurbiprofen axetil combined with fentanyl in PCA can reduce the side effects of fentanyl and offers overall favorable analgesic responses.
9.Study on the correlation between Chinese medicine syndrome types of the Uighur and Han people and levels of blood lipids and fibrinogen in premature coronary heart disease patients.

OBJECTIVETo study the correlation between the Chinese medicine (CM) syndrome types of the Uighur and Han people and levels of blood lipids and fibrinogen in premature coronary heart disease (PCHD) patients.
METHODSRecruited were 196 Uighur and 189 Han patients with PCHD first confirmed by coronary angiography at the Heart Center of First Affiliated Hospital of Xinjiang Medical University and Traditional Chinese Medicine Hospital Affiliated to Xinjiang Medical University. Another 60 healthy volunteers were recruited as the healthy control group. CM syndromes were typed by physicians or those having higher professional titles at the cardiac internal medicine. The contents of triglyceride (TG) and total cholesterol (TC) were detected using oxidase method. The high-density lipoprotein cholesterol (HDL-C) level was detected using chemical modification enzymic method. The low-density lipoprotein cholesterol (LDL-C) level was detected using selective dissolution. The fibrinogen (FIB) level was detected using Clauss congeal principle.
RESULTSThe CM syndrome types of Uighurs were sequenced as filthy turbidity and phlegm obstruction syndrome (82 cases, 21.3%) > qi deficiency and blood stasis syndrome (61 cases, 15.84%) >cold accumulation in Xin meridian syndrome (39 cases, 10.13%) >Xin-Shen yin deficiency syndrome (14 cases, 3. 63%). The CM syndrome types of the Han nationality were sequenced as qi deficiency and blood stasis syndrome (69 cases, 17.92%) >filthy turbidity and phlegm obstruction syndrome (67 cases, 17.40%) > cold accumulation in Xin meridian syndrome (42 cases, 10.91%) > Xin-Shen yin deficiency syndrome (11 cases, 2.86%). There was no statistical difference in the syndrome type distribution between the two groups (P >0.05). Compared with the healthy control group, the levels of TG, LDL-C, and FIB increased, and the HDL-C level decreased in patients of filthy turbidity and phlegm obstruction syndrome and qi deficiency and blood stasis syndrome of the patient group. The HDL-C level decreased in the Uighur patients of Xin-Shen yin deficiency syndrome, showing statistical difference (P < 0.01, P < 0.05). Compared with the Han people of the same syndrome type, the TG level increased and the HDL-C level decreased in the Uighur patients of filthy turbidity and phlegm obstruction syndrome. The FIB level increased in the Uighur patients of qi deficiency and blood stasis syndrome with statistical difference (P<0.05). In the Uighur patients, filthy turbidity and phlegm obstruction syndrome was positively correlated with TG (r=0.469) and FIB (r=0.464), and negatively correlated with HDL-C (r=-0.382). Qi deficiency and blood stasis syndrome was positively correlated with FIB (r=0.72) with statistical difference (P<0.01, P<0.05). In the Han patients, filthy turbidity and phlegm obstruction syndrome was positively correlated with TG (r=0.445) and FIB (r=0.372), and negatively correlated with HDL-C (r=-0.37). Qi deficiency and blood stasis syndrome was positively correlated with TG (r=0.471) and FIB (r=0.459) with statistical difference (P<0.01, P<0.05).
CONCLUSIONCM syndrome types were correlated with TG, HDL-C, and FIB in PCHD Han and Uighur patients.

10.Expression of hypoxia-inducible factor 2 alpha and vascular endothelial growth factor in chondrocytes of articular cartilages in human osteoarthritis

BACKGROUND:Studies have found that vascular endothelial growth factor and hypoxia inducible factor are involved in the development process of osteoarthritis, but their correlation is rarely reported. OBJECTIVE:To observe the expression and correlation of hypoxia inducible factor-2α and vascular endothelial growth factor in chondrocytes of articular cartilages in human osteoarthritis. METHODS: Articular cartilage specimens were colected from 50 patients with knee osteoarthritis undergoing total knee joint replacement. According to the joint Kelgren-Lawrance (K-L) X-ray grouping classification standard, there were 18 cases of K-LIII level and 32 cases of K-LIV level. Besides, articular cartilage specimens from 10 patients undergoing amputation for legs tumor or traffic accident served as control group. Hematoxylin-eosin staining, Safranin O-Fast Green staining and Mankin scoring were performed to observe and evaluate the histological characteristics of articular cartilages of each group, immunohistochemical staining was conducted to detect the expression of hypoxia inducible factor-2α and vascular endothelial growth factor in chondrocytes of articular cartilages, and their correlations were analyzed. RESULTS AND CONCLUSION:The Mankin score of K-LIV group was significantly higher than those of K-LIII group and control group. Immunohistochemical staining revealed that the number of chondrocytes with positive expression of hypoxia inducible factor-2α or vascular endothelial growth factor in K-LIV group was significantly higher than that in K-LIII group and control group (P < 0.05). The expression of hypoxia inducible factor-2α and vascular endothelial growth factor increased in chondrocytes of articular cartilages of osteoarthritis patients, and to up-regulate the expression of vascular endothelial growth factor may be the regulatory mechanism of hypoxia inducible factor-2αinthe pathogenesis of osteoarthritis.
