1.Oligodendrocyte transplantation for acute spinal cord injury:insulin-like growth factor 1 induces bone marrow mesenchymal stem cells differentiating into oligodendrocytes

BACKGROUND:Treatment and rehabilitation of spinal cord injury is a complicated problem,andthe reconstruction and remyelination of neural reflex pathwaysare the essentialprocess, during which oligodendrocytes play an important role in spinal cord injury repair.
OBJECTIVE:To observe theeffect ofoligodendrocyte transplantation for acute spinal cord injury in rats.
METHODS:Insulin-like growthfactor 1 induced bone marrow mesenchymal stem cels differentiating into oligodendrocytes, and those oligodendrocytes were transplanted into rats with acute spinal cord injury as induced cel transplantation group. Simple normal salineandnatural oligodendrocytes were transplanted into the rat injured spinal cord as control group and oligodendrocyte group, respectively. Rat behavioral changes were observed by inclined planetest and Basso-Beattie-Bresnahan (BBB) scores. Neurological recovery and survivalof the transplanted cels was detected and observed using spinal evoked potential and immunohistochemical staining, respectively.
RESULTS AND CONCLUSION:Compared with the control group, BBB scores and the criticalelevation angle oftheincline planetestsignificantly increased, latencies of spinal motor and sensory evoked potential were on the decline (P< 0.05), and there were no significant differencesin above indicators between the two groups at 4 and 8 weeks after transplantation. Moreover, survivedoligodendrocytes after transplantation could be found in the lesions of spinal cord in both two groups. In conclusion, insulin-like growth factor 1-induced bone marrow mesenchymal stem cels can differentiate into oligodendrocytesthat exact an excelentrole in acute spinal cord injury repair after transplantation, which achieve the equal clinical efficacy tothenatural oligodendrocytes.
2.Effect of Penqiangyan Granule on the immune function of patients with chronic pelvic inflammatory disease of blood-stasis and Shen-deficiency syndrome type.

OBJECTIVETo explore the effect of Penqiangyan Granule (PG) on the immunity of patients with chronic pelvic inflammatory disease (CPID) of blood stasis and Shen-deficiency syndrome (BSSDS) type.
METHODSSixty patients were randomly assigned to two groups: the treatment group treated with PG and the control group with Penyanjing Granule, 30 cases in each group. The treatment course was 4 weeks for both groups. The clinical efficacy, plasma levels of CD4 and CD8, and the serum levels of tumor necrosis factor-alpha (TNF-alpha) and interleukin-2 (IL-2) were measured before and after treatment.
RESULTSAfter treatment, the total effective rate in the treatment group and the control group was 96.7% and 63.3% respectively with significant difference between groups (P < 0.05); in the treatment group the plasma CD4, CD4/CD8 and serum IL-2 increased obviously, while the plasma CD8 and serum TNF-alpha decreased markedly (P < 0.05), all were significantly different with those in the control group (P < 0.05).
CONCLUSIONPG can improve the immune function and alleviate inflammation in CPID patients of BSSDS type.

Chitosan nanoparticles(CS-NP) has been a hotspot of the research due to its good biological characteristics and has a great potential to be used as the carrier for gene delivery. This article gives a simple review the application of the techniques, the currently used preparation methods of CS-NP, the factors that affect the rate of complexing of plasmid with CS-NP, the transfection efficiency of plasmid CS-NP compounds, and its in vitro drug releasing behavior.
4.Effects of fucoidan on angiogenesis of human multiple myeloma RPMI 8226 cells

AIM:To investigate the effects of fucoidan on the angiogenesis of multiple myeloma cells in vitro, and its related mechanisms .METHODS:The human multiple myeloma RPMI 8226 cells and human endothelial cells were cultured in vitro.The growth inhibition rate of RPMI 8226 cells was examined by MTT assay .The cell cycle and apoptosis rate were measured by flow cytometry .RPMI 8226 cells were treated with fucoidan for 72 h, and the cell culture superna-tant was collected .The VEGF concentration was examined by ELISA , and the tube formation assay was applied to assess the angiogenic activity .After treatment with fucoidan for 72 h at different concentrations , the protein levels of HIF-1α, VEGF, p-AKT and p-ERK1/2 were detected by Western blot .RESULTS: Fucoidan inhibited the growth of RPMI 8226 cells in a dose-and time-dependent manner .After treatment with fucoidan for 72 h, the cell cycle was arrested at G 1 phase , and the apoptotic rate of RPMI 8226 cells was increased with the increasing concentration of fucoidan , which was much higher than that in control group (P<0.05).The VEGF concentration was significantly decreased with the increa-sing concentration of fucoidan .The numbers and areas of the capillary-like structures decreased while the concentration of fucoidan increased, and those at 100 mg/L were less than those in the control (P<0.05).The protein levels of HIF-1α, VEGF, p-AKT and p-ERK1/2 in fucoidan group were significantly lower than those in control group (P<0.05).CON-CLUSION:Fucoidan inhibits the secretion of VEGF in multiple myeloma cells , and reduces angiogenesis induced by mul-tiple myeloma cells .It inhibits the protein expression of HIF-1αand VEGF , which may be related to inhibiting the phos-phorylation of AKT and ERK1/2.

Glaucoma,optical neuritis and trauma are optic nerve injury-related diseases.These ocular diseases cause irreversible damage of the vision and even blindness.The study on optic nerve regeneration is a powerful approach for rescuing vision and improving the quality of life of patients.Therefore,how to promote optic nerve regeneration is always the focus in ophthalmology.In recent years,great progression has achieved by modulation of cellular signal pathway,usage of glial cells,stem cells and neurotrophic factor,etc,which lays a basis for the treatment of optic nerve injury.Ophthalmologist should trace and participate in these researches to promote the development of regeneration medicine.This review summarizes these new approaches after discussing factors effecting optic nerve regeneration briefly and proposes questions to answer.

9.Correlation analysis between EMT and progression of chronic obstructive pulmonary disease (COPD) and its related non-small cell lung cancer (NSCLC)

Objective:Based on relevant database data analysis,to explore the correlation of the abnormal expression of EMT related factors and chronic obstructive pulmonary disease and its related non-small cell lung cancer.Methods: Based on some data sets of GEO in the NCBI database,to perform express analysis,survival analysis and correlation analysis.Results: ①Snai1 and other EMT related regulatory factors exist significantly higher expression in non-small cell lung cancer patients,and E-cadherin (CDH1) was showed significantly lower expression. ②A large number of COPD patients samples were analyzed,and some EMT-related molecules in patients with COPD also showed significant abnormal expression and consistented with the changes in patients with non-small cell lung cancer.Conclusion: The results showed that dysregulation expression of EMT related regulatory factors may have some correlation with disease progression of COPD patients through the EMT markers and their expression and correlation analysis in COPD patients.
10. Polyethyleneimine as targeted gene vectors: A review

Currently, polyethylenimine(PEI) is one of the most promising non-viral gene delivery vectors. However, high relative molecular mass PEI has obvious toxicity and is short of targeting or specificity to cells or tissues. As we know targeting or specificity is the key property of ideal gene delivery systems. Therefore, PEI should be modified to become a gene vector with targeting and low toxicity. Up to now investgations on modification of PEI for targeting effect mainly include:1.modification with endogenous ligands such as transferrin Tf RGD peptide etc 2.modification with carbohydrate such as galactose etc 3.modification with specific antibody. Of these, the specific antibody-directed PEI is anticipated to be one of promising and distinctive targeting gene delivery carriers. The present paper summarises literature on PEI as targeted gene delivery vectors.
