1.Sulfur-containing amides from Entada phaseoloides.

To study the chemical constituents of the Entada phaseoloides (L.) Merr., seeds of Entada phaseoloides were extracted with 70% ethanol at room temperature. Isolation and purification were performed by silica gel, reversed-phase silica gel column chromatography and semi-preparative HPLC. Structures of the pure compounds were established on the basis of spectral analysis. Four sulfur-containing amide compounds were isolated from the n-BuOH-soluble fraction and identified as entadamide A-beta-D-glucopyranosyl-(1-->3)-beta-D-glucopyranoside (1), entadamide A (2), entadamide A-beta-D-glucopyranoside (3) and clinacoside C (4). Compound 1 is a new compound. Compound 4 is isolated from the genus Entada for the first time.
2.Medical image retrieval based on low level feature fusion and relevance feedback.

Based on chest images' features, this paper presents a retrieval method with domination color of partition, gray level co-occurrence matrix and gradient image shape invariant moments, as well as the fusion of them and the technique of relevance feedback. In Windows Vista environment with Visual C + +, a chest image retrieval prototype is implemented and the experiments have proved that this method has a better retrieval effect.

3.Study on HPLC fingerprint of crude and processed products of Entada phaseoloides.

OBJECTIVEA novel method was established and the results were compared for HPLC fingerprint determination of crude and processed products of Entada phaseoloides.
METHODHPLC-UV was proposed for studying the fingerprints of fresh E. phaseoloides and their processed products, respectively. HPLC-ESI-MS was introduced to analyze the common peaks in each batch of crude E. phaseoloides.
RESULTSixteen characteristic peaks were found in crude E. phaseoloides samples and twenty-one common peaks existed in processed E. phaseoloides samples. Nine characteristic peaks of which were identified by comparison of the retention time and their molecular weights of chemical standards, most of which were identificated belong to triterpenoid saponins and glucosides.
CONCLUSIONAfter processed, the chemical composition of the extraction with solution of 60% methanol from crude E. phaseoloides are less or more than that from processed E. phaseoloides, and the changes of the main peaks of fingerprint chromatographic suggest that HPLC can be used to reflect the difference of chemical composition of E. phaseoloides and their processed products. It would be an efficient way for qualitative control of E. phaseoloides.

4.Role of magnetic resonance imaging in the diagnosis of lesions in the sellar region.

OBJECTIVETo evaluate the role of magnetic resonance imaging (MRI) in the diagnosis of lesions in the sellar region.
METHODSThe MRI data of 142 patients with surgically and pathologically proved lesions in the sellar region were retrospectively analyzed. Based on the MRI findings, the lesions were divided into pituitary adenoma and sellar region diseases except pituitary adenoma. According to the pathologic features, the sellar region diseases except pituitary adenoma were further divided into cystic lesions, parenchymatous lesions, and cystic and parenchymatous lesions.
RESULTSOf these 142 patients, pituitary adenoma was found in 30 cases, cystic lesions of sella region diseases except pituitary adenoma in 24. Further classification resulted in 66 cases of parenchymatous lesions and 22 cases of cystic and parenchymatous lesions.
CONCLUSIONMRI-based classification of lesions in the sellar region is useful for the diagnosis and differential diagnosis.

In order to establish a method by which the recombinant suicide plasmids integrated on the chromosome could be recircled, A simple method of transconjugative cloning was established with the helper plasmids pMT999 or pRK2013 and fusion strains of Shigella flexneri which were obtained by screening with in vivo expression technology. And the cloning efficiency with this method is very high.
6.Lesion localization and surgical resection for non-palpable breast cancer.

OBJECTIVETo investigate the methods of lesion localization and surgical treatment for non-palpable breast cancer, presented with only small calcification lesion on the images.
METHODSFrom November 2003 to August 2007, 61 patients with non-palpable lesion were finally pathologically diagnosed as early breast cancer (T1-2N0M0), based on the small calcification lesions shown by full field digital mammography (FFDM) through molybdenum target, and the rich blood supply shown by type-B ultrasonic examination. Accurate lesion-localization prior to surgical resection was conducted, and sample re-examination by FFDM was done after resection. Patients with single lesion underwent breast-conserving surgery, precise excision with the aid of image-guided wire localization, and stage I breast reconstruction was performed simultaneously using wide-based gland-tissue flap. Patients with multiple lesions received modified radical mastectomy.
RESULTSAmong the 50 patients treated with breast-conserving surgery, the accuracy of localization for lesions was 100% (50/50), and all lesions were excised completely with a negative margin proven by FFDM re-examination and pathological examination. The superior rate of mammaplasty was 86.0% (43/50) according to JCRT criteria, with a compliance difference of 1.5 cm. Modified radical mastectomy was performed in 11 patients. The follow-up period in this series was from 6 to 58 months with a mean follow-up time of 39 months. Distant metastases were detected in only one patient and local recurrence was not observed yet.
CONCLUSIONLesion localization by FFDM in patients with non-palpable breast cancer is accurate and practical. In patients with single lesion, breast-conserving resection followed by synchronous stage I breast reconstruction with wide-based gland-tissue flap is appropriate.

7.Qualitative and quantitative analysis of fluoxetine hydrochloride by 19F NMR.

The chemical shift of fluoxetine hydrochloride appears at delta 14.15 in 19F NMR analysis. The delta moved upfield slightly from 14.158 to 14.145 when the concentration of solution became diluted from 2.00 to 0.05 mmol x L(-1). Spiking test was suggested to confirm the existence of the compound for qualitative analysis. 19F NMR detection sensitivity test illustrated that a concentration of 17 mg in 1 L water could be detected while the sample was scanned 500 times with optimum parameters. In quantitative analysis, standard curve of concentration versus fluorine signal intensity was proposed to determine the amount of fluoxetine. Long capillary tube containing trifluoroacetic acid was used as internal standard for the integration measurements and straight line was obtained with good fitting. Direct additions of trifluoroethanol to fluoxetine solutions gave a poorer standard curve.

9.Effect of different glucose concentrations on the expressions of insig-1 and insig-2 mRNA during the differentiation of 3T3-L1 cells.

OBJECTIVE:
To determine the effect of different concentrations of glucose on the differentiation of 3T3-L(1) and the expression of insig-1 and insig-2 mRNA, and to explore the effect of insulin-induced gene in the differentiation and formation of adipocytes and lipogenesis.
METHODS:
The 3T3-L(1) cells were induced to differentiate in high glucose concentration (25 mol/L G.S), low glucose concentration (5.5 mol/L G.S), and mannitol (19.5 mol/L Mannitol +5.5 mol/L G.S), respectively. The differentiation of 3T3-L(1) cells was examined by oil red "O" straining, and the expression of insig-1,insig-2 mRNA and AP2 mRNA was examined by RT-PCR and in situ hybridization.
RESULTS:
With the differentiation of 3T3-L(1) cells, the expression of insig-1 and insig-2 mRNA was gradually up-regulated. The expression of insig-1 and insig-2 mRNA significantly increased while AP(2) mRNA decreased in the low glucose concentration inducing group and mannitol inducing group. In the high glucose concentration inducing group, the cell differentiation was poor (P<0.05). There was no difference between the low glucose concentration and the mannitol group in the differentiation of 3T3-L(1) cells, and in the expression of insig-1 and insig-2 and AP(2) mRNA.
CONCLUSION
Different concentrations of glucose may influence the cell differentiation and the low glucose concentration promotes insig-1 and insig-2 gene expression, which may lead to the inhibition of the differentiation and lipogenesis of preadipocytes.

10.Apoptosis induction of rhesus peripheral blood lymphocytes.

OBJECTIVETo establish a method for inducing apoptosis of rhesus peripheral blood lymphocytes (PBLs).
METHODSRhesus PBLs were irradiated with X-ray, (60)Co gamma-rays and ultraviolet (UVC254 nm), respectively, and the cell apoptosis was evaluated with flow cytometry using annexin-V staining and propidium iodide staining.
RESULTSX-ray and (60)Co gamma-ray irradiation induced only low apoptotic rates of the PBLs, and UVC resulted in the highest apoptotic rate of about 60%. UVC irradiation of the PBLs in RPMI supplemented with 10% heat-inactivated fetal calf serum for 60 min at a distance of 20 cm led to an early apoptotic rate of 58.85% and necrotic rate of 11.5%. The apoptotic rate of PBLs increased in a dose- and time-dependent fashion.
CONCLUSIONFor inducing apoptosis of the rhesus PBLs, UVC can be more effective than X-ray and (60)Co gamma-ray. The highest apoptotic rate can be achieved when the rhesus PBLs in RPMI supplemented with 10% heat-inactivated fetal calf serum are exposed to UVC for 60 min at the distance of 20 cm.
