Gene regulation relies on the precise binding of transcription factors (TFs) at regulatory elements, but simultaneously detecting hundreds of TFs on chromatin is challenging. We developed cFOOT-seq, a cytosine deaminase-based TF footprinting assay, for high-resolution, quantitative genome-wide assessment of TF binding in both open and closed chromatin regions, even with small cell numbers. By utilizing the dsDNA deaminase SsdAtox, cFOOT-seq converts accessible cytosines to uracil while preserving genomic integrity, making it compatible with techniques like ATAC-seq for sensitive and cost-effective detection of TF occupancy at the single-molecule and single-cell level. Our approach enables the delineation of TF footprints, quantification of occupancy, and examination of chromatin influences on TF binding. Notably, cFOOT-seq, combined with FootTrack analysis, enables de novo prediction of TF binding sites and tracking of TF occupancy dynamics. We demonstrate its application in capturing cell type-specific TFs, analyzing TF dynamics during reprogramming, and revealing TF dependencies on chromatin remodelers. Overall, cFOOT-seq represents a robust approach for investigating the genome-wide dynamics of TF occupancy and elucidating the cis-regulatory architecture underlying gene regulation.

Plutonium isotopes(238Pu,239Pu,240Pu and 241Pu)in the environment are important"fingerprint"nuclides in the study of nuclear activity traceability. The content of plutonium isotopes in the environmental metrics is usually very low,and the measurement of these isotopes, especially 238Pu,using mass spectrometry is seriously interfered with by the coexisting 238U.The analysis of several plutonium isotopes in soil usually requires combination of multiple measurement techniques, which leads to a long analysis time and large uncertainty in the isotope ratio. In this work, the hydrous titanium oxide(HTiO)precipitated by the hydrolysis of titanium oxydichloride(TiOCl2)under near-neutral condition was used to preconcentrate plutonium from the soil digestion solution, and the highly efficient decontamination of 238U in the sample was achieved by TK200 resin column chromatography with a decontamination factor of 108.Simulation resuts of density functional theory(DFT)showed that NH3 was considered as a promising reaction gas to eliminate the interference of 238U from 238Pu measurement using mass spectrometry due to the significant discrepancy of the chemical reactivity of U+and Pu+with the reactive gas NH3.Experiments confirmed that by optimizing the flow rates of collision gas(He)and reaction gas(NH3),the interference of 238U could be effectively suppressed, and the decontamination factor of 238U was 104.Combined with chemical separation, the overall decontamination factor of 238U could reach 1012 by using the developed method.By combining chemical separation and tandem quadrupole inductively coupled plasmon-mass spectrometry(ICP-MS/MS)measurement,the simultaneous determination of four ultra-trace plutonium isotopes in soil was realized, and the detection limit of plutonium isotopes was at the femtogram level.Analysis of the international standard reference materials(NIST-SRM-4357 and IAEA-384)showed that the established method could be successfully used for the accurate analysis of ultra-trace four plutonium isotopes(238Pu,239Pu,240Pu and 241Pu)in soil samples.

Body weight abnormalities, including overweight, obesity, and underweight, have become a dual public health challenge in Chinese adults: overweight and obesity lead to a variety of chronic complications, while underweight increases the risks of malnutrition, sarcopenia, and organ dysfunction. To systematically address these issues, multidisciplinary experts in endocrinology, sports science, nutrition, and psychiatry from various regions have held multiple weight management seminars. Based on the latest epidemiological data and clinical evidence, they expanded the guideline to include assessment and intervention strategies for underweight, in addition to the core content of obesity management. This guideline outlines the etiological mechanisms, evaluation methods, and multidimensional management strategies for overweight and obesity, covering key areas such as diagnosis and assessment, medical nutrition therapy, exercise prescription, pharmacological intervention, and psychological support. It is intended to provide a scientific and standardized approach to weight management across the adult population, aiming to curb the rising prevalence of obesity, mitigate complications associated with abnormal body weight, and improve nutritional status and overall quality of life.

Objective To investigate the impact of eccentric placement for various types of artificial aortic valves on downstream flow dynamics. Methods A physiological pulsatile circulation simulation system was employed and particle image velocimetry(PIV)was utilized to analyze the downstream flow field variations for bioprosthetic and mechanical valves under two placement conditions:centralized placement(0 mm)and eccentric placement(1 mm).Hemodynamic parameters such as velocity, vorticity, and viscous shear stress were assessed to evaluate the flow field characteristics. Results By analyzing the flow field variations at four characteristic time points, namely, early systole, acceleration phase, peak systole, and deceleration phase,a significant difference in flow field distribution between bioprosthetic and mechanical valves was observed. The bioprosthetic valve exhibited a centrally symmetric jet with a higher flow velocity, whereas the mechanical valve displayed a three-jet structure with a lower central flow velocity.Under eccentric placement, the blood flow in the aortic sinus region was sluggish, with a reduction in average velocity, hindering the formation and maintenance of vortices.During the peak systolic phase, the maximum viscous shear stresses in the sinus region for the bioprosthetic and mechanical valves were 0.45 and 0.67 Pa,respectively, approaching the threshold for endothelial cell damage. Conclusions Eccentric placement of both mechanical and bioprosthetic valves resulted in reduced sinus blood flow velocity and diminished viscous shear stress, creating favorable conditions for thrombus formation. In clinical practice, careful attention should be given to the placement of valve replacement to prevent eccentric placement.

Chagas disease(American trypanosomiasis)is a zoonosis caused by Trypanosoma cruzi, which severely affects public health.Recently, with changes in economic globalization and increased population mobility, this disease has gradually spread from the original Latin American epidemic areas to non-epidemic areas, such as Europe, thus showing a trend of globalization. The main trans-mission routes have changed from transmission via the Triatomine vector to blood transfusion transmission,mother-to-child transmis-sion,oral transmission, and other routes.Consequently,Chagas disease is spreading globally, and more people are increasingly vul-nerable to infection. This article retrospectively reviews research on the transmission routes of Chagas disease, analyzes the changing trends in transmission routes, and provides a scientific basis for the formulation and optimization of Chagas disease prevention and con-trol strategies from a One Health perspective.

Journal of Chinese Physician 2025;27(5):727-730
Objective:To investigate the effect of lumbar sympathetic ganglion block (LSGB) on postoperative ileus (POI) in patients undergoing colonic surgery. Methods:Ninety patients scheduled for elective laparoscopic colonic surgery in the same treatment group at the Nanjing First Hospital from March 2022 to August 2023 were selected and randomly divided into a control group and an observation group using a random number table, with 45 patients in each group. The observation group received ultrasound-guided LSGB before anesthesia, while the control group received no intervention. Both groups received standardized anesthesia protocols and management after entering the operating room. The time to gastrointestinal motility recovery (first defecation time), first flatus time, first feeding time, number of cases requiring gastric tube decompression, postoperative hospital stay, number of cases with nausea and vomiting, intraoperative additional and titrated sufentanil dosage, remifentanil dosage, first patient-controlled intravenous analgesia (PCIA) press time, sufentanil dosage at 4, 8, 12, 16, and 24 hours postoperatively, and the number of analgesic pump presses were recorded. Results:Compared with the control group, the observation group had shorter first defecation and flatus times, earlier first feeding time, shorter postoperative hospital stay, and fewer cases of nausea (all P<0.05). The observation group also had less intraoperative sufentanil supplementation and titration, later first PCIA press time (all P<0.05), and significantly fewer sufentanil dosages and analgesic pump presses at 4, 8, 12, 16, and 24 hours postoperatively compared with the control group (all P<0.05). Conclusions:LSGB reduces the incidence of POI in colonic surgery patients, decreases opioid use, and shortens hospital stay.
