1.Effect of kuntai capsule on the number of retrieved oocytes, high-quality oocytes and embryos in in vitro fertilization of poor ovarian response patients.

OBJECTIVETo observe the effect of Kuntai Capsule (KC) on the number of retrieved oocytes, the quality of high-quality oocytes and embryos in in vitro fertilization of poor ovarian response (POR) patients.
METHODSTotally 70 POR patients preparing for in vitro fertilization-embryo transfer (IVF-ET) were randomly assigned to the observation group and the control group, 35 cases in each group. KC was administered to patients in the observation group in the preparation cycle (i.e., three menstrual cycles before IVF-ET) and during the superovulation process. Those in the control group took placebo during this period. Before and after medication the improvement of Shen yin deficiency syndrome (SYDS) was observed in the two groups. The basal follicle-stimulating hormone (bFSH), luteinizing hormone (LH), estradiol (E2), anti-Mullerian hormone (AMH), the ratio of FSH to LH, and antral follicle count (AFC) were observed. Besides, the E2 level of a single ovum on the day of HCG injection, the number of retrieved oocytes, the high-quality oocyte rate, and the high-quality embryos were observed.
RESULTSCompared with the control group, the SYDS, decreased bFSH and LH levels, increased ACF numbers, the E2 level of a single ovum on the day of HCG injection, the number of retrieved oocytes, high-quality oocytes, and high-quality embryos were superior in the observation group (P < 0.05). There was no statistical difference in the decreased FSH/LH level (P > 0.05). E2 and AMH increased after medication of KC in the observation group, while they decreased after administration of placebos in the control group. There was statistical difference in the post-pre treatment difference of E2 and AMH between the two groups (P < 0.05).
CONCLUSIONKC could increase the number of retrieved oocytes, and elevate the quality of occytes and embryos in the IVF-ET.

Many bioactive peptides from neural and endocrine tissue are amidated at C-terminals,which is essential for their activities.The ?-amide comes from post-translational modification that is catalyzed by ?-AE (?-amidating enzyme) or PAM (pepdilylglycine ?-amidating monooxygenase).The gene encoding ?-AE was amplified with PCR and cloned into the plasmid pET-30a.After the recombinant plasmid pET-A was transformed into E.coli BL21,the ?-AE was expressed and purified by the Ni2+affinity chromatography,which has the ability catalyzing Dns-Tyr-Val-Gly to Dns-Tyr-Val-NH2.It identified that the recombinant protein producing by E.coli BL21 is ?-AE,which will benefit for studies of amidation at the C-terminals of peptides.

Objective To explore the risk factors of neonatal necrotizing enterocolitis(NEC)and assess the effect of prophylactic use of probiotics(Bifico)in prevention of NEC and observe changes of weight.Methods Five hundred and twenty-four cases of prematures who were inpatients were divided into 2 groups randomly(prevention group,n=276,no-prevention group,n=248)and the morbidity of NEC and the increase of eight during hospitalization were observed.A case-control study and conditional Logistic regression model multifactorial analysis were made to 20 cases NEC neonates and 80 cases non-NEC neonates.Results The prevention group(276 cases)had 5 cases of NEC and the average weight increase was(8.114?8.137)g/d,the no-prevention group 248 cases had 17 cases of NEC,and the average weight increase was(6.595?5.337)g/d.The occure rate of NEC in prevention group was significantly lower than that of no-prevention group(?2=5.57 P
6.Effect of suprasorb G polyurethane hydrogel application as an early nursing intervention on the prophylaxis of phlebitis

Objective To investigate the efficacy of suprasorb G polyurethane hydrogel application as a method of early nursing interventions on the prophylaxis of mechanical phlebitis resulting from PICC tube.Methods 102 elderly patients with hematological malignancies were randomly divided into experimental group and contrast group by parity sequence of setting PICC tube.The patients in experimental group received prophylactic administration of suprasorb G polyurethane hydrogel application along the punctured vein 3 to 5 cm above acupuncture point for 7 to 10 days.And then psychological and exercise instruction were also given after PICC tube.The patients in contrast group were only given psychological and exercise instruction after PICC tube.Results The incidence of mechanical phlebitis in experimental group was 3.92% and significant difference ( x2 =3.888,P < 0.05) was present when compared with contrast group ( 15.7% ).Conclusions Suprasorb G polyurethane hydrogel application as an early nursing intervention can markedly reduce incidence of mechanical phlebitis induced by PICC tube and relieve pain.
7.Similarity between leaves of Nauclea officinalis and stems of Nauclea officinalis.

The study is to develop a method to determine 3 batches leaves of Nauclea officinalis and stems of N. officinalis by HPLC. The differences between strictosamide contents and fingerprints was compared, then chromatographic peak of fingerprints was validated with the assistance of LC-MS. The strictosamide contents in stems of N. officinalis were higher than leaves of N. officinalis. The main chemical composition in leaves of N. officinalis and stems of N. officinalis were alkaloid which revealed by LC-MS. There are 7 chemical compositions were same between them, but the chemical composition in leaves of N. officinalis is more than stems of N. officinalis. This provides a scientific basis for the development of the potential medicinal value of leaves of N. officinalis and the sustainable utilization of N. officinalis.

Rapid screening and characterization of bioactive compounds from traditional Chinese medicines ( TCM) is one of the highlighted re-search challenges in the field of drug development.Target molecule affin-ity coupled with mass spectrometry ( MS) has become a new method for the screening of bioactive compounds from complex mixture, the recent development of which since 2005 will be reviewed.According to the prin-ciple of affinity screening, the methods could be classified into pre -column affinity screening including ultrafiltration -MS, size -exclusion chromatography-MS, turbulent-flow chromatography-MS, restricted-access material-MS and immobilized target-MS and post-column af-finity screening (HPLC-continuous-flow bioassay-MS).Each meth-od with its typical applications would be introduced, providing technique references for the screening of bioactive compounds from complex mixture such as TCM.
9.Simultaneous determination of six coumarins in Radix Glehniae using pressurized liquid extraction and HPLC

Objective To develop a HPLC method for simultaneous de-termination of six coumarins in Radix Glehniae from different sources. Methods The samples were analyzed by HPLC after pressurized liquid extraction ( PLE ) .The chromatographic conditions were as following:The column was Kinetex core-shell-C18 column (50 mm ×4.6 mm, 2.6 μm) , the mobile phase consisted of acetonitrile-water using gradi-ent elution, the flow rate was 0.4 mL? min-1 and the column tempera-ture was 30℃.The detection wavelength was set at 310 nm and the sam-ple injection volume was 20 μL.The method was validated by investiga-ting its specificity, linearities and limit of detection, precision, extraction recovery, stability and repeatability.Results Six components were de-termined by HPLC, including psoralen, xanthotoxin, bergapten, impera-torin, cnidilin and isoimperatorin. The linearities of them were good ( r≥0.999 2 ) in the concentration ranges of 0.50 -10.00 , 1.00-20.00, 0.32-6.40, 0.25 -5.00, 0.05 -1.00, 0.17 -3.50μg? mL-1 , respectively.The coumarins showed the overall recoveries ranging from 91.7%-107.6%.The contents of six compounds of Radix Glehniae from different areas had an obvious difference.Conclusion Six coumarins in Radix Glehniae were analyzed by PLE and HPLC. The method has good specificity, reproducibility and high sensitivity, which can be applied for the quality control of Radix Glehniae.
