2.The dynamic change of Brn-3a expression in retina of chronic high intraocular pressure rat
Chinese Journal of Experimental Ophthalmology 2011;29(12):1077-1081
Background Brn-3a is a newly discovered specificity marker for retina ganglion cells(RGCs).It is well-known that RGCs damage is a important pathological basis of hypertension-visual disorder. But the study concerning expression of Brn-3a in RGCs in glaucoma eye is still rately. Objective The purpose of this work was to investigate the changes of Brn-3a expression in model eye with chronic high intraocular pressure(IOP) and its relation with morphology of retina and the expression of Brn-3a in chronic ocular hypertension rats. Methods Thirty-five clean adult SD rats were randomly divided into normal control group(5 rats) and model group(30 rats).Experimental chronic ocular hypertension models were induced unilaterally in the left eyes of 30 health adult SD rats by cauterizing super-scleral veins, and the conjunctival incision was made in the right eyes as sham operative group. The operated rats were subdivided into 6 groups according to the examination time points and 6 rats for each group.IOP was measured with Tono-Pen tonometer before and after 30 minutes,1,3,7,14,28 day after surgery respectively. The rat models were sacrificed in 1,3,5,7,14,28 days after operation by excessive anesthesia method, and retinal section was prepared for the histopathological examination and the RGCs were counted using Nissl staining method.Expression of Brn-3a in RGCs was detected by immunohistochemistry. This experimental complied with the Regulations for the Administration of Affair Concerning Experimental Animals by State Science and Technology Commission. Results The IOP was significantly raised from 30 minutes to 28 days after operation in model eyes with the top IOP( 34.46±4.65 )mmHg in the 30 minutes after operation, showing statistically significant differences in various time groups ( F =95.631,P =0.001 ) and different eyes ( F =287.473,P =0.001 ).Compared with sham operative group, the IOP were elevated from 1 day through 14 days after operation ( q =18.418,15.261,10.987,6.931,4.975,2.962,P < 0.05 ).The numbers of RGCs were ( 29.08 ± 1.98 ) in the normal control group and decreased gradually by 3.17%,7.84%,14.60%,22.40% in 1,7,14,28 day after surgery in the model eyes with the considerable differences in comparison with normal eyes ( t =5.943,8.034,15.023,17.004,19.371,P < 0.05 ).Immunochemistry revealed that Brn-3a was specially expressed in the RGCs layer and the positive RGCs for Brn-3a were evidently decreased as the prolong of high IOP duration ( F =127.583,P =0.000 ).Conclusions Chronic glaucoma model can be successfully established using Shareef-Sharma method. The loss of RGCs is associated with the high lOP duration. The expression level of Brn-3a is a reliable index of high-IOP-induced damage of optical nurve.

The diagnosis and treatment of prostate cancer are being improved due to the popularized screening of prostate specific antigen. Advanced prostate cancer, in spite of its response to androgen deprivation therapy, may finally develop into castration-resistant prostate cancer (CRPC) and shorten the overall survival of the patients. Many efforts have been made by worldwide researchers for new approaches to the management of CRPC, including new hormonal therapy, cytotoxic chemotherapy, immunotherapy, and bone metastasis-targeted therapy. This paper reviews the emerging agents undergoing clinical evaluation and drugs that have received approval for the treatment of CRPC in order to provide doctors and patients with more treatment options for CRPC and improve the overall survival rate and quality of life of the patients.

Objective To prepare the mB7-1-GPI-anchored Lewis vaccine and investigate its antitumor effects. Methods mB7-1-GPI was incorporated on Lewis tumor cells and mB7-1-GPI-anchoring tumor vaccine was prepared. The anti-tumor immunity induced by the prepared mB7-1-GPI-anchored Lewis tumor cell vaccine in tumor-bearing mice was observed. Results Flow cytometric analysis showed that mB7-1-GPI were positively expressed on the surface of Lewis tumor cells. After Lewis tumor cells incubated with mB7-1-GPI, the positive rate (PR) of mB7-1 antigen was 95.8% (0h), 93.6% (4h), 91.1% (8h) and the fluorescence intensity (FI) was 11.2(0h), 10. 6(4h), 9. 8(8h). The IL-2 and IFN-γ production of splenic lymphocytes + lewis cells was (25.9 ± 1.4) pg/ml, (56. 0± 3. 5 ) pg/ml. The IL-2 and IFN-γ production of splenic lymphocytes + lewis/mB7-1-GPI was ( 871.3 ± 10. 4 ) pg/ml, ( 1329. 0 ± 11.9 ) pg/ml. In 25 days, the mean diameter of tumor of Lewis/mB7-1 -GPI was shorter than Lewis( 1.4 ± 0. 21 )cm & ( 2. 5 ± 0. 27 )cm , P < 0. 05 ). Lewis tumor cell-bearing C57BL/6 mice treated with Lewis/mB7-1-GPI vaccine survived much longer than mice treated with Lewis vaccine ( 75.2 ± 2. 0 ) d & (40. 2 ± 2. 0 ) d ( P < 0. 05 ). Conclusion The Lewis tumor vaccine prepared with mB7-1-GPI fusion protein significantly inhibited the tumor growth in Lewis bearing mice. It represented an useful new strategy for attaching immunological factor onto tumor cell surfaces without genetic manipulation.

Journal of Acupuncture and Tuina Science 2006;4(5):261-263
Objective: To observe the influence of electroacupuncture with different frequencies on rats swimming endurance and sift out the optimal frequency parameter. Methods: Wistar rats were randomly allocated to control group, non-acupoint group and electroacupuncture with low frequency group (EA with 2 Hz group), electroacupuncture with medium frequency group (EA with 15 Hz group), and electroacupuncture with high frequency group (EA with 100 Hz group), acupoints Zusanli (ST 36) and Taichong (LR 3) were punctured and rats load swimming time was used as an index, and the change of swimming endurance time before and after acupuncture was observed. Results: After electro-acupuncture, swimming time was increased in 15 Hz and 100 Hz groups respectively, with the significant differences between before and after acupuncture (P<0.05). There was also a significant difference in the percentage of increase in swimming time between 15 Hz group and other groups (P<0.05).Conclusion: It is suggested that electroacupuncture at acupoints Zusanli (ST 36) and Taichong (LR 3) with a frequency of 15 Hz could enhance rats's swimming endurance, and 15 Hz might be an optimal frequency parameter during the electrical stimulation.

Acta Pharmaceutica Sinica 2016;51(1):29-32
Autophagy is a crucial biological process in eukaryotes, which is involved in cell growth, survival and energy metabolism. It has been confirmed that autophagy mediates toxicity of anticancer drugs, especially in heart, liver and neuron. It is important to understand the function and mechanism of autophagy in anticancer drugs-induced toxicity. Given that autophagy is a double-edged sword in the maintenance of the function of heart, liver and neuron, the autophagy-mediated toxicity are very complicated in the body. We provide a review on the concept of autophagy and current status about autophagy-mediated toxicity of anticancer drugs. The knowledge is crucial in the basic study of anticancer drugs-induced toxicity, and provides some strategies for the development of alleviating the toxicity of anticancer drugs.
