Glioma is the most common form of brain cancer. Despite recent advances in the treatment of solid tumors, there are few effective treatments for malignant gliomas due to its infiltrative nature. It has important significance to improve the treatment of glioma through in-depth understanding the intracerebral metabolic characteristics and pharmacokinetics of chemotherapeutics. Brain microdialysis (B-MD), an effective method to monitor central nervous system anticancer drug disposition, conditions of drugs through the blood-brain barrier, basic pathophysiologic metabolism, bioactive compounds and the changes of neurotransmitter in brain, provides the unique opportunity to allow the simultaneous determination of unbound concentrations of drugs in several tissues, and directly measure gliomas biochemistry continuously. B-MD has been able to monitor the change of brain drugs, metabolites and neurotransmitters, dynamic analysis of the drug concentration and pharmacological effect after administration, pharmacodynamic interaction between drugs, receptor mechanism of drug transport, as well as feedback information of internal environment. B-MD is expected to provide reference for clinical individual chemotherapy of glioma, but also provide powerful tools for the evaluation of new anticancer drugs in vivo. In this review, a comprehensive overview of B-MD for studies on glioma is elucidated with special emphasis on its application to neurochemistry and pharmacokinetic studies.

Objective To measure the normal cerebrospinal fluid of the midbrain aqueduct peak velocity of different age groups by using magnetic resonance imaging (MRI) time‐spatial labeling inversion pulse (time‐SLIP) ,and to discuss the flow law of CSF . Methods Forty‐one cases of healthy volunteers ,including 23 cases of male ,18 cases of female ,aged 7 -73 .Patients were divided into 4 groups:7- <15 age group (n=11) ,15- <35 age group (n=10) ,35- <45 age group (n=9) ,≥45 age group (n=11) .We used the application of Toshiba 1 .5 T MRI head coil ,MRI sequence of time‐SLIP marked CSF to calculated the peak velocity of CSF in the midbrain aqueduct and measure the diameter and length of the midbrain aqueduct .Results The peak flow velocity be‐tween the 7- <15 age group and 15 - <35 age group ,35 - <45 age group ,≥45 age group were statistically significant (P<0 .05);inner diameter and length were no statistically significant difference (P>0 .05) between age .Conclusion In the midbrain aqueduct ,CSF is two‐way flow and supports the CSF of the pulsatile flow theory ;MRI sequence of time‐SLIP could measure CSF velocity and display the turbulence fluiding .

Objective: To observe the influence of electroacupuncture with different frequencies on rats swimming endurance and sift out the optimal frequency parameter.Methods: Wistar rats were randomly allocated to control group, non-acupoint group and electroacupuncture with low frequency group (EA with 2 Hz group), electroacupuncture with medium frequency group (EA with 15 Hz group), and electroacupuncture with high frequency group (EA with 100 Hz group), acupoints Zusanli (ST 36) and Taichong (LR 3) were punctured and rats load swimming time was used as an index, and the change of swimming endurance time before and after acupuncture was observed. Results: After electro-acupuncture, swimming time was increased in 15 Hz and 100 Hz groups respectively, with the significant differences between before and after acupuncture (P＜0.05). There was also a significant difference in the percentage of increase in swimming time between 15 Hz group and other groups (P＜0.05).Conclusion: It is suggested that electroacupuncture at acupoints Zusanli (ST 36) and Taichong (LR 3) with a frequency of 15 Hz could enhance rats's swimming endurance, and 15 Hz might be an optimal frequency parameter during the electrical stimulation.

There are many progresses in the therapy of MM with ASCT. ASCT improves the ratio of CR,EFS and OS,compared with conventional chemotherapy.Myeloma patients with renal failure or old age also can be candidate for ASCT.Tandem autologous transplantation improves the ratio of CR. Its impact to long-term EFS and OS still need to be judged.To improve conditional project,to choose suitable time,and- toselect consolidation schedule can boost therapeutic effect.

Objective To investigate the effect of costimulatory molecules 4-1BB/4-1BBL on T cell activation with systemic lupus erythematosus(SLE)patients.Methods The expression of NF-kB mRNA and p38 MAPK mRNA of T cells with 20 SLE patients and 20 normal controls before activation,after activation and blocked bv 4-1BB/4-1BBL were detected bv RT-PCR.The protein levels of p-p38 MAPK and NF-kB were detected by Western blot.Results The expression of NF-kB mRNA and p38 MAPK mRNA and the protein expression of NF-kB and p-p38 MAPK of T cell with SLE patients were higher than that of normal controls(P＜0.01).There were more expression of NF-kB mRNA,p38 MAPK mRNA,NF-kB protein and p-p38 MAPK protein of T cell with SLE patients stimulated by anti-CD3 monoantibody(P＜0.01).In anti-4-1BB monoantibody blockage group,there were decreased expression of p38 MAPK mRNA and p-p38 MAPK protein(P＜0.01).but not the decreased NF-kB mRNA and protein expression.Conclusion Costimulatory molecule 4-1BB plays a critical role in T cell activation of SLE by p38 MAPK signal pathway.

Objective To investigate the effect and mechanism (a selective cyclooxygenase-2 inhibitor) on invasive ability of human nasopharyngeal carcinoma (NPC) line CNE-2Z. Methods The proliferation of NPC cells was examined by MTT assay. The invasive and migrating ability of NPC cells was detected with transwell chamber. E-cadherin protein expression was detected by immunocytochemistry and the expressions of Cox-2 and E-cadherin mRNA were analyzed by reverse transcription-polymerase chain reaction (RT-PCR). Results MTT showed that celecoxib inhibited CNE-2Z proliferation in dose-dependent manner, the survival rate of cells treated with 25, 50, 100 μ mol/L celecoxib (-x±s) for 24 h was(94. 75 ±1.34)%, (91.77 ±2. 70)% , (64. 54 ± 1.20)%, respectively, and the survival rate of cells treated for 48 h was ( 88.41±1.28 ) %, ( 78. 84 ± 1.56 ) %, ( 52. 46 ± 2. 25 ) %, respectively, the concentration of 50% inhibition concentration of a substance (IC50) was 100 μmol/L, the difference was statistically significant between different concentration groups in the same time-point ( repectively, F were 462. 204 and 1328. 306, P ＜0. 01 ). Treated with different concentrations of celecoxib(0, 25, 50 μmol/L) for 24, the cell numbers (-x±s) through PVPF by tumor invasion assay were (263.7 ± 13.5), (185.3 ±8.7) and (144. 0 ± 8. 2), the difference was statistically significant between the experimental and control group (F =102. 089, P ＜0. 01 ). Immunocytochemistry showed that celecoxib significantly induced the increase of Ecadherin protein expression, also with a dose-dependence in 0 μmol/L, 25 μmol/L, 50 μmol/L group was (21.7 ±2. 6), (28. 7 ±2. 4), (40. 3 ± 1.3), and 50 μmol/L group increased significantly ( F =78. 637,P ＜0. 01 ). RT-PCR showed that celecoxib reduced the expression of Cox-2 mRNA expression in 25, 50 μmol/L group decreased significantly compared with the control group (respectively, t were 23. 950 and 36. 651, P ＜ 0. 01 ), but it enhanced the expression of E-cadherin mRNA expression in 25, 50 μmol/L group was significantly higher ( respectively, t were 35. 829 and 81. 497, P ＜0. 01 ). Conclusion Celecoxib can inhibits the invasive ability of NPC cell line CNE-2Z, which possibly relates with the upregulated expression of E-cadherin.

Objective To compare the bioequivalence and pharmacokinetics of national made and imported atorvastatin in healthy male Chinese volunteers after single oral administration.Methods This randomized sequence,open-label,two-period crossover study with a one-week washout period between doses was performed in 24 fasting healthy Chinese males.They were randomly assigned to receive 20 mg of either the test ( national made) or reference (imported) formulation orally.The blood samples were collected over a 72-hour period.Plasma concentrations of parent atorvastatin (AT),ortho-hydroxy-atorvastatin (o-OAT)and para-hydroxy-atorvastatin (p-OAT) were simultaneously determined using the validated liquid chromatography-tandem mass spectrometry method,the bioequivalence was also evaluated throughout the study.Results The main pharmacokinetic parameters of test and reference formulations were as follows:the values of Cmax for AT were ( 10.6 ± 11.9 ) μg/L and ( 10.6 + 9.8 ) μg/L,t1/2z were ( 11.4 ± 3.9 ) h and ( 11.4 ± 5.3 ) h,AUC0-t were (54.2 ± 37.4) μg · h - 1 · L- 1 and (51.7 ± 34.1 ) μg · h -1 · L- 1,respectively.The values of Cmax for o-OAT were (7.8 ±4.5) μg/L and (7.6 ±4.3) μg/L,t1/2z were (12.3 ±4.2) h and ( 11.9 ± 3.4) h,AUC0-t were (96.8 ± 48.2) μg · h - 1 · L- 1 and (92.3 ± 44.4) μg · h - 1 · L - 1,respectively.The values of Cmax for p-OAT were (0.5 ±+ 0.4 ) μg/L and (0.4 + 0.3 ) μg/L,t1/2z were ( 18.4 + 12.4 ) h and (23.3±17.8) h,AUC0-t were (15.9+12.3) μg· h-1 · L-1 and (13.8+8.11) μg· h-1 · L-1,respectively.The relative bioavailability of AT and o-OAT in test formulation were ( 105.3 ± 20.7 ) ％ and ( 107.8 ±23.2)％,respectively.The 90％ confidence interval of the test/reference geometric mean ratios of AUC0-t for AT and o-OAT were (97.7 - 110.5)％ and (98.3 - 111.3)％,Cmax for AT and o-OAT were (75.8- 114.0)％ and (90.6 -122.9)％,they were all located within the bioequivalence criteria range (80％ -125％ for AUC,and 70％ - 143％ for Cmax ).Conclusion The result demonstrated that two formulations were bioequivalent.

OBJECTIVETo investigate the precise locations of the blood vessels and nerves surrounding the seminal vesicles (SV) in men and provide some anatomical evidence for SV-related minimally invasive surgery.

METHODSWe observed the courses and distribution of the blood vessels and nerves surrounding SVs and obtained the data for positioning the SV neuroplexes in 20 male pelvises.

RESULTSOne branch of the neuroplexes was distributed to the SVs bilaterally with the neurovascular bundles, (2.85 ± 0.18) cm from the median sulcus of the prostate (MSP), while another branch ran through the Denonvillier fascia behind the SV, (0.81 ± 0.06) cm from the MSP. The arterial SVs (ASV) originated from the inferior vesical artery and fell into 4 types, 55% going directly to the SVs as one branch, 15% running between the SV and the ampulla of the deferent duct as another branch, 25% downward as 2 branches to the SV and between the SV and the ampulla of the deferent duct respectively, and 5% as the other ASVs. The shortest distance from the ASV through the prostatic neuroplexus to the posterior SV was (1.08 ± 0.09) cm.

CONCLUSIONIn SV resection, neuroplexus injury can be reduced with a bilateral distance of < 2.85 cm and a posterior distance of < 0.81 cm from the MSP, and so can bleeding by vascular ligation between the SV and the ampulla of the deferent duct.
