1.Effect of aluminum exposure on abnormal phosphorylationof tau protein in PC12 cells of rats

Objective :
To investigate the effect of aluminum exposure on expression of miR-497-5p, wingless murine breast cancer virus integration site family member 3a (Wnt3a), β-catenin protein, glycogen synthase kinase-3β (GSK-3β) protein and tau protein in rat adrenal pheochromocytoma PC12 cells, so as to provide insight into unraveling the mechanisms underlying aluminum exposure-induced abnormal phosphorylation of tau protein.
Methods:
PC12 cells were exposed to Al(mal)3 at concentrations of 0, 100, 200, 400 μmol/L for 24 h. The viability of PC12 cells was measured using cell counting kit-8 (CCK-8) assay. The relative expression of miR-497-5p and Wnt3a was detected using a real-time fluorescent quantitative PCR (RT-qPCR) assay, and the expression of Wnt3a, β-catenin, GSK-3β, P-GSK-3β (Ser9), tau and p-tau (Ser396) proteins were determined using Western blotting.
Results :
The viability of PC12 cells appeared a tendency towards a decline with the increase of aluminum dose (Ftrend=323.473, P=0.001). RT-qPCR assay detected that the relative miR-497-5p expression appeared a tendency towards a rise with the increase of aluminum dose (Ftrend=14.888, P=0.031), and the relative Wnt3a expression appeared a tendency towards a decline with the increase of aluminum dose (Ftrend=165.934, P<0.001). The miR-497-5p expression negatively correlated with the relative Wnt3a expression (r=-0.693, P=0.012). The expression of Wnt3a (Ftrend=357.656, P=0.001), β-catenin (Ftrend=208.750, P=0.001) and p-GSK-3β (Ser9) proteins (Ftrend=512.583, P<0.001) appeared a tendency towards a decline with the increase of aluminum dose, and the expression of GSK-3β (Ftrend=39.965, P<0.001), tau (Ftrend=277.929, P=0.006) and p-tau (Ser396) proteins (Ftrend=96.247, P=0.002) appeared a tendency towards a rise with the increase of aluminum dose.
Conclusion
Up-regulation of miR-497-5p and GSK-3β expression and down-regulation of Wnt3a and β-catenin expression may be a mechanism underlying aluminum exposure-induced abnormal phosphorylation of tau protein.

To discuss the distribution of patents' holders for Chinese herbal compounds in treating cardiovascular and cerebrovascular, the patents' holders for Chinese herbal compounds in treating cardiovascular and cerebrovascular were cluster analyzed by means of simple statistics and cluster analysis. Clustering variables were composed of patent applications, patent maintained number, related papers' quantity, etc. Chinese herbal compound patents' holders were divided into four categories according to their different scientific research and patent strength. It is the magic weapon for Chinese herbal compound patents' holders that have scientific research patents' transforming and make coordination of patent protection and scientific innovation.

5.Cultural Conditions for Production of Glutathione by Mutant Saccharomyces J-X25

The production conditions of glutathione with shaking flask fermentation by the mutant Saccharomyces J-X25, a methionine-defected strain were studied, and the optimum culture conditions are as follows: initial pH6. 0, temperature 30℃, 100ml in 500ml flask, the inoculum size 10% and agitation rate 220r/min. The emphasis was on the stimulating effect on the cells by dioxogen and the sodium lactate as surfactant. Both of which were added at the logarithmic phase of fermentation, and the GSH production was up to 0. 253g/L , 52% higher than the control without the additions. Compared with the production of GSH initial strain that by the mutant in optimum conditions was increased by 79%.
6.Effect of Acetylcholine on Electric Discharge of Pain-related Neurons in Hippocampal CA1 Area

Objective To study the effect of intracerebroventricular(icv) injectionon acetylcholine(ACh) on the elecric activities of pain-related neurons in the hippocampal CA1 area of rat.Methods Trains of the electric impulses applied to the right sciatic nerve were used as noxious stimuli.The discharges of neurons were led out by extracellular recording method with glass microelectrodes.Results Icv injection of ACh(20?g/10?l) caused an decrease of the evoked discharge frequency of pain-excited neurons(PEN) and a prolongation latency as well as an increase of the evoked discharge of pain-inhibited neurons(PIN) and a shortening of inhibitory duration.Conclusion The response of pain-related neurons in hippocampal CA1 area to the noxious stimulation is weakened by icv injection of ACh,exhibiting analgesic effect.

8.Develepment of Hand Function Rehabilitation Evaluation and Training System Based on Virtual Reality Technol-ogy

Objective To develop a system for hand function evaluation and training based on virtual reality. Methods 5DT Data Glove 5 Ultra, Visual Studio 2010 were integrated as the development environment and DirectX 9.0 as components, and the system was developed based on the MFC programming framework. Results and Conclusion The system can assess the fingers function, recognise gestures and can be used as a 3D environment for virtual training, which may guide the patients to take the training actively.
9.Application Practice of Quality Control Circle in the Management Improvement of Dismounted Drug in Outpatient Pharmacy of Our Hospital

OBJECTIVE:To reduce the types of drugs which was not consistent with the accounts in monthly checking of out-patient department,reduce the wastage of dismounted drugs and improve dismounted drug management. METHODS:Through qual-ity control circle(QCC),analyzing the reasons for inconformity of accounts and drugs,adopting relevant countermeasures of staff training,cipher prescription,counter storage,managed by designated person,expiry data management and so on,the process of dismounted drug management was formulated and implemented. The types of drugs which was not consistent with the accounts and activity growth value of circle members were analyzed statistically before,during and after QCC. RESULTS:The types of drugs which was not consistent with the accounts were 33.6,21.2 and 13.6 before,during and after QCC,decreasing by 59.5%. The ac-tivity growth value of circle members were all positive value. CONCLUSIONS:QCC of outpatient pharmacy can effectively reduce the types of drugs which is not consistent with the accounts in monthly checking,standardize dismounted drug management and im-prove pharmaceutical care quality.
10.The Study of Association Factors Affecting IVF/ICSI-ET

Objective To investigate the association factors affecting the course of IVF/ICSI-ET. Methods The clinical data of 348 cycles of IVF/ICSI-ET in patients with different causes of infertility were analyzed retrospectively. Results Ovarian reserve and response to stimulation declined with aging. The ability of bFSH predicting ovarian response was low. E 2 peak concentration, number of big follicle and retrieved oocytes decreased significantly when bFSH concentration evelated. The number of big follicle and retrieved oocytes obviously increased with the E 2 peak concentration increasing. The number of antral follicle and volume of ovary were positive relation with ovarian response. None of the factors above could predict the outcome of IVF indenpedently. Conclusion Age was strongly associated with ovarian reserve and response. The bFSH level was one of the indices reflecting ovarian reserve, and had the limited capability for predicting ovarian response and IVF outcome.E 2 peak concentration could not predict the IVF consequence. The number of total antral follicle and volume of ovary were the most direct index reflecting ovarian response. The conditions of patients should comprehensively be evaluated to raise the pregnant rate of IVF.
