1.Effects of valproic acid on expression of SMN2 mRNA in neuron-like cells derived from patients with spinal muscular atrophy

Objective To study the effects of valproic acid (VPA) to the expression of SMN2 mRNA in neuron-like cells (NLCs) derived from patients with spinal muscular atrophy (SMA). Methods Polymerase chain reaction-restriction fragment length polymerphism (PCR-RFLP) was performed to select the patients with SMA. Mesenchymal stem cells (MSCs) derived from patients were induced into NLCs which were set as the model of neurons. The transcripts of SMN2 by reverse transcription-polymerase chain reaction (RT-PCR) combined with sequenceing were detected. Semi-quantitative RT-PCR analysis was performed to detect the changes of SMN2 mRNA expression between before and after the NLCs were treated by VPA. Results Two bands (266 bp and 212 bp) were found in the gel picture of RT-PCR, which were respectively the products of full length transcript (fl-SMN mRNA) and skipping exon 7 (SMN?7 mRNA). Poor and monotonous work could easily lead to a decrease of arousal level of the monitoring work personnel. In order to improve the performance of monitoring work, low arousal level needs to be recognized and awakened. We proposed a recognition method of low arousal by the electroencephalogram (EEG) as the object of study to recognize the low arousal level in the vigilance. We used wavelet packet transform to decompose the EEG signal so the EEG rhythms of each component were obtained, and then we calculated the parameters of relative energy and energy ratio of high-low frequency, and constructed the feature vector to monitor low arousal state in the operation. We finally used support vector machine (SVM) to recognize the low arousal state in the simulate operation. The experimental results showed that the method introduced in this article could well distinguish low arousal level from arousal level in the vigilance and it could also get a high recognition rate. Have been compared with other analysis methods, the present method could more effectively recognize low arousal level and provide better technical support for wake-up mechanism of low arousal state.

4.Prospective study of improved elasticity scores in the differential diagnosis of malignant and benign breast lesions

Objective To explore the value of improved elasticity scores in differentiating malignant and benign breast lesions. Methods A prospective study was done in 394 patients with 508 breast lesions confirmed by pathology and the elastographic images of the lesions were evaluated by using old and improved elasticity scores respectively. Results The elastographic images of the lesions could be evaluated and classified in all patients by improved elasticity scores but could not be evaluated and classified in 82 lesions by old elasticity scores. The accuracy of ultrasonic elastography for diagnosing benign and malignant breast lesions was 88.8% and 78.1%, respectively by old elasticity scores and improved elasticity scores. Conclusions The elastographic images of breast lesions were accurately evaluated by using improved elasticity scores, and the use of improved elasticity scores is helpful to reduce the biopsy which is not necessary.
5.Initial study of contrast-enhanced ultrasound 5-point scoring system in the diagnosis of breast tumor

Objective To explore the application of contrast-enhanced scoring system in the diagnosis of breast tumor.Methods 249 patients with 260 lesions were examined by elastography and contrastenhanced ultrasound before surgery.All the lesions were scored according to the two different methods.Pathological results were made the golden criteria.The diagnosis accuracy of the two methods were compared.Results There were 145 benign and 115 malignant tumors.The diagnosis accuracy,specificity and sensibility of elastography were 85.0%,88.3%,80.1 % respectively.The critical value of contrastenhanced ultrasound was between 3 and 4.The diagnosis accuracy,specificity and sensibility of contrastenhanced ultrasound were 88.9%,87.6%,90.4% respectively.Area under ROC curve of the two methods were 0.901 and 0.910,the difference was not statistically significant.Conclusions Contrast-enhanced ultrasound 5-point scoring system was an initial quantitative criterion for breast tumor diagnosis.The diagnosis accuracy was parallel to that of elastography.

7.Research on adhesive strength of different composites resin core and fiber posts

Objective:To study the adhesive strength between fiber posts and four different composite resin core materials.Methods:Four resin core materials (Multicore Heavy Body、LuxaCore、 LIGHT-CORE and Z100) were placed around the fiber posts. The adhesive strengths of the samples were tested by the universal electrical testing machine. The interface between fiber post and resin core was observed by SEM. Results:Different resin core materials exhibited significant different influences on adhesive strengths(P<0.05). The LuxaCore reached the maximum of (314.31±15.36) N and the Z100 was the minimum of (267.39±10.54) N.Conclusion:Resin core material exhibits a significant influence on adhesive strength. The dual-curing composite core material with low viscosity has the highest bond strength in all groups.

9.Effects of zinc on carbon and nitrogen metabolism in leaves of Pinellia ternata and their yield

Objective To find out the proper concentration of ZnSO4?7H2O as foliar fertilizer used for Pinellia ternata, and to provide reference for high yield of P.ternate,effects of zinc on the carbon and nitrogen metabolism in leaves of P.ternata and the yield were studied.Methods Seedlings of P.ternata were employed as the experiment material.At the stage of full seedling,ZnSO4?7H2O of different concentration was sprayed on the leaves,the indexes of carbon and nitrogen metabolism were determined on the day 10,the weight of tubers and bulbils were measured at the sprout tumble stage.Results The yield of P.ternata has an exceedingly significant positive correlation with the contents of soluble sucrose and starch in the leaves,a significant positive correlation with the content of photosynthetic pigment and the aldolase activity,and a negative correlation with the amylase activity.In addition,the yield shows both an exceedingly significant positive correlation whith the nitrate reductase activity and a significant positive correlation with the content of protein.At the stage of full seedling,zinc sprayed on leaves can increase the content of photosynthetic pigment and improve the capability of photosynthesis.Meanwhile,spraying zinc can also promote the accumulation of soluble sucrose and starch in the leaves because of its stimulation on aldolase and inhibition on amylase.Besides,spraying zinc on leaves could improve the activity of nitrate reductase and the content of protein,and finally increase the yield of P.ternata.Conclusion The concentration of 600 mg/L is the best for ZnSO4?7H2O used as foliar fertilizer for P.ternate at the stage of full seedling.
10.Expressions of ER and PR in cast-off cells from fluid of latex ductal lavage and their significances in diagnosis and treatment of breast diseases

Objective To investigate the value of the expressions of estrogen receptor(ER)and progesterone receptor(PR)in the cast-off cells from the fluid of latex ductal lavage in the diagnosis and treatment of breast diseases.Methods A total of 58 female out-patients were examined by fiberoptic ductoscopy(FDS).All patients were divided into 6 groups by the complaints,physical examination,ultrasound and FDS:normal group(10 persons without symptoms and abnormalities in any examination),galactostasis group(n=19),latex fill up group(n=11),intraductal papilloma group(n=2),mammary ductal ectasia group(n=7)and contralateral examination after mastectomy for breast cancer group(n=9).A total of 168 ducts were successfully examined by FDS,and the expressions of ER and PR in the cast-off cells were detected among all of the samples.Results The positive rates of ER and PR expression in the cast-off cells from the fluid of latex ductal lavage in benign breast disease groups were much higher than those in normal group(P
