1.In vitro and in vivo models of osteosarcoma for research of enhanced green fluorescent protein expression

Background and purpose: Mouse osteosarcoma model was widely used in osteogenic malignant tumor research, and it was helpful for studying the invasion and metastasis of the tumor cells when it was well marked in vivo. The purpose of this study was to establish mouse sarcoma cell lines (S180) that were infected with enhanced green fluorescent protein(EGFP). Methods: EGFP-S180 expressing strong EGFP fluorescence was acquired by electroblot, and supplemented with G418 (800 mg/mL), c-Myc was detected by laser scanning confocal microscopy. Meanwhile, the cancer-bearing model was established subcutaneously within the abdominal cavity. Results: EGFP-S 180 cells were cloned. There was no significantly difference between c-Myc expressions in S180 cells and those in EGFP-S180 cells (P>0.05), and between the cancer-bearing time subcutaneously and the time within abdominal cavity (P>0.05). Conclusion: According to in vitro and in vivo assay, it showed that EGFP-expressing S180 cells could be used for studying further the tumor biological behavior with fluorescence technology.
3.Changes of synapsin Ⅰ expression and synaptic ultrastructure in hippocampus of the rat models of epilepsy

Objective To illuminate relationships between epilepsy and functional and morphologic plasticity of synapse through investigating temporal-spatial expression of syanpsinⅠand the alteration of synaptic ultrastructure in hippocampus after seizure. Methods The models of epilepsy were established by injection of pilocarpine and lithium. Electromicroscope and the software of image manipulation were applied to observe the alteration of synaptic ultrastructure in hippocampus during acute phase, resting phase and chronic phase. The expressions of synapsinⅠ were determined by immunohistochemistry. 6.Strain Ratio Measure and Improved 5-scoring System in Differentiation of Benign and Malignant Breast Tumors with Ultrasonic Elastography

Purpose To investigate the value of strain ratio (SR) measure and improved 5-scoring system of ultrasonic elastography in the differentiation of benign and malignant breast tumors.Materials and Methods The data of 213 patients with breast cancer confirmed by pathology or surgery were retrospectively studied.All the patients underwent ultrasonic elasticity imaging examination and were evaluated by strain ratio (SR) method and improved 5-scoring system.Taking pathology as gold standard,we measured the sensitivity,specificity and accuracy of the two methods and evaluated the diagnostic efficiency.Results The area under curve (AUC) and 95% CI of SR measure was 0.926,0.871-0.980 and that of improved 5-scoring system was 0.921,0.870-0.973.The best diagnostic limit of SR was 3.055,when SR > 3.055,the lesions was malignant,when SR<3.055,the lesions was benign.The sensitivity,specificity and accuracy of SR and improved 5-scoring system in diagnosing malignant breast lesions were 92.1%,92.7%,92.5% and 85.5%,83.9%,84.5%,respectively.The specificity and accuracy of SR was higher than the improved 5-scoring method,the difference was statistically significant (P<0.05),but there was no significant difference in sensitivity (P>0.05).Conclusion SR is more objective in diagnosing malignant breast tumors,which has higher diagnostic value than improved 5-scoring system.
7.Effects of E23K polymorphism in KCNJ11 gene on membrane current.

OBJECTIVEE23K polymorphism in KCNJ11 gene is associated with cardiovascular disease and diabetes. In order to explore the mechanism of E23K correlation to related diseases, the effect of E23K polymorphism in KCNJ11 gene on membrane current was investigated.
METHODSThe exon of KCNJ11 was obtained by PCR amplification and the G-->A mutation was completed by overlap extension PCR. The sequences of KCNJ11 exon contained 23E or 23K was inserted into pcDNA3.1/CT-GFP vector respectively. The recombinant plasmid, pcDNA3.1-KCNJ11(E) and pcDNA3.1-KCNJ11(K), were transfected into HEK293T cells by lipofectamine and the membrane current density was determined by whole-cell patch clamp technique.
RESULTS1,173 bp sequences of KCNJ11 gene's exon were amplified by PCR and the recombinant expression plasmid, pcDNA3.1-KCNJ11(E) and pcDNA3.1-KCNJ11(K), were constructed successful. Positive and negative currents were detected in HEK293T cells transfected with difference plasmid by whole-cell patch clamp technique. Results showed that the reversed voltage was 50mV. The current in HEK293T cells with pcDNA3.1-KCNJ11(E) was significantly greater than that with pcDNA3.1-KCNJ11(K) (P < 0.05, n = 10).
CONCLUSIONThe polymorphism of E23K in exon of KCNJ11 gene changed the membrane currents in HEK293T cells. It could be an experiment support for the possible mechanism between the locus and related diseases.

8.A qualitative research on posttraumatic growth in patients with cervical cancer who had a hysterectomy at the childbearing age

Objective We aimed to explore the experience of posttraumatic growth in patients with cervical cancer who had a hysterectomy at the childbearing age.Methods The phenomenological methodology was used in the study.An in-depth interview was conducted among 12 patients chosen by purposive sampling method.Results Through careful analysis and collation,three themes were identified:smashing the original assumption,searching the meaning of existence and rebuilding the new cognitive schemata.The experience of patients' was conformed to the model which was put forward by Tedeschi,and the theme changes in personal relationships,personal strength,altruism born from sufferings and new philosophy of life were conformed to theoretical connotation of PTG.Conclusions Clinical medical staff should update psychological nursing concept timely and taking advantages of PTG to help patients pass through their difficult period.
10.Intratracheal anti-tumor necrosis factor-α antibody effect on ultra-structure of lung after cardiopulmonary bypass

Objective To study the effect of intratracheal anti-tumor necrosis factor-α antibody(TNF-αAb)on ultra-structure of lung after cardiopulmonary bypass(CPB).Method 28 healthy rabbits were selected and randomly evenly divided into four groups:I group only received open chest operation;Ⅱ-Ⅳ groups underwent CPB.In the IV group,rabbit TNF-α Ab (2400 ps/kg)Was dropped into the intracheal tube before operation and just after releasing the aortic clamp.Saline was given to the Ⅲ group by the same way.Water volume,TNF-α mRNA,TNF-α protein,apoptosis and pathomorphological changes were measured in the lung tissues.Results TNF-α Ab can re-duce releasing of TNF-α.It could also reduce the occurrence of apeptosis and attenuate pathomorphological changes in the lung tissue.Conclusion Intratracheal TNF-α Ab markedly lessenes the injury of nltrastructure of lung after CPB.
