Objective:To establish a method for the determination of Mg and Al in 5-layer co-extrusion bags and their migration from the bags into infusion by ICP-MS. Methods:The samples were digested by microwave digestion, and the contents of Mg and Al were determined by ICP-MS under certain working conditions. Results:Both Mg and Al had a linear range between 2. 5 and 20. 0 μg ·L-1, and r was 0. 999 6 and 0. 999 7, respectively. The average recovery of Mg was 92. 90% (RSD=3. 01%,n=9),and that of Al was 94. 15% (RSD=4. 11%,n=9). Conclusion:The method is accurate, sensitive and simple, and can be used for the determi-nation of Mg and Al in 5-layer co-extrusion bags and their migration from the bags into infusion.

Objective To clone the coding dense granules protein 8(GRA8) from Toxoplasma gondii RH isolate for the potential use in the development of DNA vaccination. Methods Amplifing gene fragment coding GRA8 from the genomic DNA of Toxoplasma gondii RH isolate by means of ploymerase chain reaction (PCR), the gene is inserted into cloning vector pUC-19 digesting with restrictive enzymes and linking react ions. The positive colon is screed on LB plates containing ampicilline and IPTG, Xgal identified by blue-white and restrictive enzyme digestion. The inserted GR A8 gene was recombined with pcDNA3.1(+) eukaryotic expression vector by digestion with restrictive enzyme and linking reactions. The positive coloe is screene d o n LB plates containing ampicillin and identified by restrictive enzymes and link ing reactions. Results The GRA8 gene with about 804 base is specifically amplified from genomic DNA of Toxoplasma GRA8/RH and pcDNA3.1(+)-GRA8 recombinant is successfull y constructed. The sequencing results showed that GRA8 gene of isolate RH and R H from genebank shares quite high homology. Conclusion The gene encoding GRA8 is amplified from genomic DNA of Toxoplasma gene isolate GRA8/RH and pcDNA3.1 (+)-GRA8 recombinant is successfully constructed.

Journal of Experimental Hematology 2006;14(3):419-426
The molecular characterization of self-antigens expressed by human malignancies that are capable of elicitation of anti-tumor immune responses in patients has been an active field in hematology, oncology, and tumor immunology. More than 2000 tumor antigens have been identified. These significant progresses have led to the renaissance of tumor immunology and studies on novel anti-tumor immunotherapies in lymphomas, other hematologic malignancies and tumors. However, despite of the progress in the identification of these self-tumor antigens, current antigen-specific immunotherapies for tumors are far less satisfactory than that expected, which reflects the urgent need to improve our understanding on the basic principles underlying the selection of these self-tumor antigens. In order to develop more effective antigen-specific anti-tumor immunotherapies and to monitor the responses to these immunotherapies in patients with lymphomas and other malignancies, many additional questions need to be addressed. In this brief review, the progress in the identification of tumor antigens in lymphomas and other malignancies was outlined and the new principles of self-tumor antigens and their significance for future immunotherapies to these malignancies were summarized.
