1.Incidence of C-Shaped Root Canal Systems in Mandibular Second Permanent Molar and the Effect of One-Visit Root Canal Treatment

Objective To investigate the incidence of C-shaped root canal systems in mandibular second perma?nent molar of patients in the First People’s Hospital of Yunnan Province, and compare the success rate between one-and two-visit root canal treatment. Methods Two hundred and sixty mandibular second molars were recruited from the pa?tients in the department of endodontics of our hospital, and teeth with C-shaped root canal were categorized by using radio?graphic classification criteria and the modified Melton’s method. Teeth with C-shaped root canal systems were then random?ly divided into two groups to receive one-or two-visit root canal treatment, and success rates of 6-month post-operation were compared between two groups. Results Eighty out of 260 mandibular second molars exhibited C-shaped canals (30.77%). Radiographic examination showed that 25 teeth belonged to typeⅠ(31.25%), 32 teeth typeⅡ(40.00%) and 23 teeth typeⅢ(28.75%) in two groups. As for modified Melton’s method, the incidence rates were 21.25%, 48.75%, 16.25%and 13.75% for C1,C2,C3a and C3b, respectively, while C4 was not found. The success rates were 78.94% (30/38) and 81.08%(30/37) for A group and B group, respectively, with no statistically significant difference between two groups ( 78.94 vs 81.08, P>0.05). Conclusion Results indicate a frequent incidence of C-shaped root canal system of mandibular per?manent second molars in our hospital. One-visit root canal treatment is an effective way for treating C-shaped canals in man?dibular permanent second molar.
2.Effect of serum inflammatory cytokine by adjuvant minocycline ointment in the treatment of aggressive periodontitis

Objective To investigate the changes of serum C reactive protein(CRP) ,tumor necrosis factor alpha(TNF-alpha) and interleukin-17(IL-17) by minocycline ointment on the basis of conventional therapy in the treatment of aggressive periodontitis (AgP) .Methods All 45 cases of AgP patients were randomly divided into two groups ,23 cases in the therapeutic group ,were per-formed scaling and root planning plus local minocycline hydrochloride Ointment medication;22 cases in the control group ,were trea-ted by scaling and root planing only .All aggressive periodontitis patients were detected the serum CRP ,TNF-alpha and IL-17 levels before and after treatment ,and compared with normal controls .Results The serum levels of CRP ,TNF-αand IL-17 in normal hu-man were lower than those of AgP patients before and after treatment(P<0 .05) .After 3 months treatment ,the serum levels of CRP ,TNF-αand IL-17 in AgP patients were significantly lower than those before treatment(P<0 .05) .Conclusion The effect of minocycline on the basis of conventional therapy in the treatment of AgP patient is remarkable ,which can improve the serum level of CRP ,TNF-alpha and IL-17 .
3.Cilia Toxicity and Mucosal Irritation of Gastrodin Nasal Temperature-sensitive in situ Gel

Objective:To evaluate the nasal safety of gastrodin nasal temperature-sensitive in situ gel through the studies on cilia toxicity in toads and mucosal irritation in rats. Methods:The toads were randomly divided into four groups, saline group, gastrodin in situ gel group, blank gel matrix group and sodium deoxycholate group, and the cilia toxicity was observed in vivo by a toad palate meth-od. The rats were randomly divided into three groups, saline group, gastrodin in situ gel group and blank gel matrix group, and the mucosal irritation was studied in rats through the observation of nasal mucosal pathological changes and behavioral indices. Results:Compared with the saline group, gastrodin in situ and blank gel matrix showed no notable effect on the cilia movement function in toads, and the effect on cilia movement of sodium deoxycholate showed statistically significant difference when compared with that of sa-line, gastrodin in situ gel and blank gel matrix (P<0. 01). During and after the treatment, no sneezing appeared in the rats. Com-pared with that in the saline group, the number of scratching nose in the gastrodin in situ gel group and blank gel matrix group in-creased (P<0. 05) without difference between the groups (P>0. 05), and after the 2-day withdrawal, that in the gastrodin in situ gel group and blank gel matrix group decreased significantly when compared with that at the last administration (P<0. 05) and showed no notable difference when compared with that in the saline group (P>0. 05). The number of inflammatory cells in the nasal mucosa in the gastrodin in situ gel group and blank gel matrix group increased complicated with congestion and cilia falling off, and after the with-drawal, the mucosal morphology in the three groups showed no significant difference. Conclusion:The local application of gastrodin in situ gel has high security, which is valuable to be studied further.
4.Helicobacter pylori induces NOD1/NF-κB activation and IFN-βand IP-10 production in gastrics of mice

Objective:To construct the Helicobacter pylori infected C57BL/6 mice model to observe the activation of NOD1/NF-κB signaling pathways in the gastric tissues,and study its roles in inflammatory response during Hp infection.Methods:6-8 week-old C57BL/6 mice were randomly divided into two groups,the Hp infection group and the control group,and mice were given by gavage every 48 h for five times with Hp or PBS,respectively.All the animals were sacrificed at different time point and the gastric tissue were stained with hematoxylin-eosin( HE);The mRNA expression of NOD1 and RIP2 in gastric tissues were examined by RT-PCR;Levels of IFN-βand IP-10 in mice serum were assessed by ELISA;Nuclear translocation of p65 in gastric tissue was detected by Western blot.Results:Hp infection elicits an inflammatory cell response,glands in gastric tissue were reduced or atrophic,as compared with that in the control group.The levels of IP-10 and IFN-βincreased in the model group, and peaked at 16 weeks after Hp infection.Hp infection increased the mRNA expression of NOD1 and the p65 content in nuclear between 24-120 h(P<0.05),and the highest level at 48 h,subsequently the expression levels were began to decrease.The mRNA expression level of RIP2 was up-regulated after Hp was administrated, peaked at 48 h and declined after 72 h.However, the expression levels would rise again at 120 h.Conclusion: Hp infection can activate the NOD1/NF-κB signaling pathways and induce the production of IFN-βand IP-10 in gastrics of mice.

7.Analysis on literature distribution and research status of depression in the elderly published in journals of Chinese Medical Association from 2003 to 2012

Objective To analyze the literature distribution and research status of depression in the elderly published in Chinese Medical Association journals.Methods The literatures of elderly depression published in 85 kinds of Chinese Medical Association journals from 2003 to 2012 were collected.The publication year,distribution,content,author,institution and region of these literatures were analyzed.Results From 2003 to 2012,there were 85 articles on depression in the elderly published in 15 kinds of journals of the Chinese Medical Association.These literatures were published mostly in 2011 (18 articles,21.2%),secondly in 2010 (13 articles,15.3%) and 2012 (12 articles,14.1%),which were published mostly in Chinese Journal of Geriatrics (29,34.1%).These articles mainly included basic science,inventory survey,diagnostic methods and clinical treatment strategy,and the most published form was treatise (77 articles,90.6%).The author who published the most articles was YUAN Gui yong from Nanjing (3 articles,3.5%),the institution which published the most articles was People's Hospital of Hainan Province (4 articles,4.7%),and the provinces or cities which published the most articles were Beijing (15 articles,17.6%),Shandong (9 articles,10.6%) and Zhejiang (7 articles,8.2%) by turns.Among the 85 articles,31 articles (36.5%) were supported by foundations.The definition,epidemiology,mechanism,influence factors,clinical characteristics,diagnosis and treatment strategy of elderly depression,and especially secondary depression were well discussed in these literatures.Conclusions The literatures of elderly depression published in Chinese Medical Association journals are insufficient in quantity,and more than half of the literatures are published from 2010 to 2012.The distribution of journals is relatively concentrated,but the distribution of author,institution and region in these articles is dispersed.Secondary depression in the elderly is concerned especially.
8.Three-dimensional ultrasound locatization in endoscopic thyroidectomy for multinodular goiter.

OBJECTIVE:
To explore the three-dimensional ultrasound locatization in endoscopic thyroidectomy for multinodular goiter (MG).
METHOD:
Thirty two patients with MG underwent ET via bilateral areolar approach. Preoperative ultrasonography were used to locate lesions and marked in the 3-D map designed by us. Dur ing ET procedure, surgery skills were adopted to expose inferior pole of thyroid, glands in the area of Berry ligament or (and) upper pole were retained according to the preoperative location map.
RESULT:
According to the preoperative ultrasonic orientation, 3 lobectomy were performed in 15 cases of unilateral lesions, 12 subtotal thyroidectomy were performed in 17 cases of bilateral lesions, and 5 patients performed unilateral lobectomy add partial thyroidectomy. All patients had successful ET using bilateral areola approach. No conversions to open surgery, no complication occurred, and no nodules were found missing in the postoperative ultrasonography examinations.
CONCLUSION
Preoperative ultrasonic orientation and intraoperative operating skills are contributing to a successful ET for MG.

9.Effects of transforming growth factor-β2 on human Tenon fibroblasts transformation and scarring after glaucoma filtration surgery

Background Research showed that transforming growth factor-β2 (TGF-β2) promotes scar formation.But its mechanism in scarring after glaucoma filtration surgery is worthy of studying.Objective This study was to investigate the effect of TGF-β2 on myofibroblast transition of human Tenon fibroblasts (HTFs) and scarring after glaucoma filtration surgery.Methods Tenon capsular tissue was obtained from 3 patients with strabismus during the surgery and was incubated in DMEM with 10% fetal bovine serum (FBS).The cells were collected and passaged in the free-serum medium for 24 hours,and then 1,2,5,10,20 μg/L TGF-β2 was added into the medium respectively,to induce the transformation of HTFs,and 2 μg/L or 5 μg/L TGF-β2 was used to treat the HTFs for 6,24,48 and 72 hours.The control group was not treated with TGF-β2.The expressions of α-smooth muscle actin (α-SMA) and phosphorylation of the signaling proteins (pSmad2) in HTFs were detected by Western blot assay.The expressions of α-SMA and F-actin were located by cell immunofluorescine technique under the confocal immunofluorescence microscopy.Cell contractility was determined by collagen gel contraction assays.This study was approved by Ethic Committee of Institute of Surgery Research of Daping Hospital,and informed consent was obtained from each patient or custodian initial of the study.Results The expression of α-SMA protein in the HTFs was increased significantly after the treatment of TGF-β2 in comparison with the control group and reached a peak at 24-48 hours.The α-SMA expression was gradually weakened in the 10 μg/L TGF-β2 groups.Little of α-SMA and F-actin were expressed in the control group.However,strong staining for α-SMA and F-actin were observed in the 1,2 and 5 μg/L TGF-β2 groups and then the staining weakened at the concentration of 10 μg/L.In addition,pSmad2 showed a stronger expression in the 2 μg/L TGF-β2 group than that in the PBS group and FBS group,with the strongest expression in 30 minutes through 2 hours.The untreated gel contracted (78.00±3.13)% from its initial size,and contraction in the 1,2,5,10 μg/L TGF-β2 group were (63.88±1.78)%,(20.69±0.65)%,(19.49-±0.54)%,(16.24±0.84) %,respectively,TGF-β2 increased HTFs contraction significantly (Fgroup =859.400,P =0.000).Conclusions TGF-β2 can induce transdifferentiation of Tenon fibroblast into myofibroblast and increase cell contractility,with a concentration-dependent and time-dependent pattern to an extent.It may be the mechanism of scar formation after glaucoma filter surgery.
10.Establishment of human neuroblastoma SH-SY5Y cell line stably silencing beclin1.

The pGenesil-1-Beclin1 eukaryotic expression vectors were constructed to establish an SH-SY5Y cell line stably expressing shRNA-Beclin1. The shRNA was connected to pGenesil-1 to construct the recombinant plasmid pGenesil-1-Beclin1, which was transformed into JM109 E. coli. Positive clones were identified by digestion with restriction endonuclease and DNA sequencing. SH-SY5Y cells were cultured by the conventional method. The pGenesil-1-Beclin1 and pGenesil-1 plasmids were transfected into SH-SY5Ycells, and the cells were screened by G418 until the stable G418-resistant monoclonal cells were acquired. Beclin1 mRNA and Beclin1 protein were detected by RT-PCR and Western blot analysis respectively. The results of restriction endonuclease analysis and DNA sequencing confirmed the correct construction of the eukaryotic expression vector pGenesil-1-Beclin1. Two SH-SY5Y transfected cell lines were successfully selected. Compared with the control group, RT-PCR and Western blot showed that the expression of Beclin1 mRNA and protein were down regulated 71.28% ± 1.45% (P < 0.05)and 75.50% ± 2.63% (P < 0.05), respectively. The results indicated that the eukaryotic expression vector pGenesil-1-Beclin1 was successfully constructed and the SH-SYSY cell lines with inhibited Beclin1 expression were established. It provides a useful cell model for studying the biological function of Beclin1.
