1.Effect of SPPW on the invasive and metastatic capacities of human gastric cancer cells.

OBJECTIVETo clarify the action and possible mechanisms of SPPW, a Chinese herbal preparation consisting of Herba Scutellariae Barbatae, Radix Astragalus, Radix Glycyrrhizae, etc., in suppressing the metastasis of human gastric cancer, by way of observing its effect on the invasive and metastatic capacities of gastric cancer cells.
METHODSIn vitro serial sub-cultured human gastric cancer cell line SGC-7901 at the logarithmic growth phase were randomly divided into 5 groups, i.e. the negative control group, the 4 treatment groups intervened respectively with SPPW at three different doses (high, middle, and low), and 5-FU. The adhesion capacities of gastric cancer cells to matrigel were detected by MTT assay 48 h after intervention. The invasive and migratory capacities of gastric cancer cells were determined by Transwell assay. The mRNA and protein expressions of metalloproteinase-9 (MMP-9) and vascular endothelial growth factor (VEGF) in gastric cancer cells were detected by Western blot and Real-time polymerase chain reaction (RT-PCR) respectively.
RESULTSCompared with the negative control group, the adhesive, invasive and migratory capacities of gastric cancer cells were all significantly inhibited in the four treatment groups (P<0.05, P<0.01). In addition, the protein and mRNA expressions of MMP-9 and VEGF were down-regulated (P<0.01). Significant dose-dependent relation existed in the three SPPW treatment groups (P<0.01). Compared with the 5-FU treatment group, the high dose SPPW treatment group showed significant difference in inhibiting the adhesive and metastatic capacities of gastric cancer cells, lowering VEGF protein expression, and mRNA expressions of MMP-9 and VEGF (P<0.01).
CONCLUSIONSSPPW could lower the adhesion of gastric cancer cells to matrigel, and lower the invasion and migration of gastric cancer cells. Meanwhile, it could down-regulate the mRNA and protein expressions of MMP-9 and VEGF, which may possibly be one of its mechanisms for influencing the invasion and metastasis of gastric cancer cells.

3.Free vascularized fibular associated with iliac graft to treat old femoral neck fracture

Objective To evaluate the clinical results of free vascularized fibular associated with iliac graft in treatment of old femoral neck fracture.Methods From 1994 to 1997,76 cases of old femoral neck fracture were treated with free vascularized fibular associated with iliac graft,they were followed up for observ- ing fracture healing,hip function and femoral head necrosis.Results Sixty-eight of these 76 cases were fol- lowed up with detail data with the main term as 10.5 years.All fractures healed within 4 to 6 months.63 cases got a almost normal hip function with the Harris score was 87.5,in these cases,the structure of femoral head were normal and the grafted fibular and iliac bone healed with the femoral.4 cases got to femoral head necrosis and 1 experienced hip dislocation.Conclusion Free vascularixed fibular associated with iliac graft is a good method to treat these cases with old femoral neck fracture who want reserve their femoral head.
4.Therapeutic efficacy of small dosage of methylprednisolone in treatment of sepsis and effect on immune cells

Objective To investigate the effect of small dosage of methylprednisolone in treatment of sepsis and the influence on immtme cells.Methods Totally 72 patients diagnosed with sepsis in Liqun Hospital from October 2014 to October 2016 were selected in this research.They were randomly divided into two groups:36 patients in control group received conventional anti-infective treatment,and 36 patients in observation group were treated with small dosage of methylprednisolone on the basis of control group.The efficacy and level of immune cells were compared in two groups.Results The survival rate in observation group was significantly higher than that of control group (P < 0.05).The difference of the incidence of complications and the level of immune cells before treatment between two groups had not statistical significance.CD4+ T lymphocytes and CD8+ T lymphocytes in observation group were significantly higher than those of control group after treatment,whereas the ratio ofCD4+/CD8+ and levels of CRP,TNF-α,PCT and IL-1 were lower than those of control group (P < 0.05).Conclusion There is a significant effect of small dosage of methylprednisolone in treatment of sepsis,which can regulate apoptosis of T lymphocyte and inflammatory factor,reduce the immune response,improve the survival rate,and recommend the clinical popularization and application.
5.Characteristics of cases with chromosome 3q21q26 aberrations.

To investigate the cytogenetic and clinical characteristics of inv(3q) (q21q26) and t(3;3) (q21; q26) aberrations as well as prognosis, cases were collected and chromosome specimens of bone marrow cells were prepared by 24-hour culture, while G-banding technique was used to perform karyotyping. The results showed that the simple inv(3q) and t(3; 3) aberrations were rare, they commonly combined with other chromosome aberrations such as -7/7q- and t (9; 22). The involved diseases included myelodysplastic syndromes, acute myeloid leukemia and chronic myelogenous leukemia in blast crisis. Out of 24 patients, 2 patients diagnosed with M(5) subtype did not achieve complete remission after multiple chemotherapy; 2 patients received allogenic stem cell transplantation relapsed. It is concluded that 3q21q26 aberration commonly combined with chromosome aberration 7/7q-, for these patients the efficacy of chemical therapy is poor, the efficacy of bone marrow transplant is too poor, these patients with inv(3q) and t(3; 3) aberrations have poor prognosis and short overall survival.

6.Amplitude Changes of Low Frequency Fluctuation in Brain Spontaneous Nervous Activities Induced by Needling at Hand Taiyin Lung Channel.

OBJECTIVETo observe amplitude changes of low frequency fluctuation in brain spontaneous nervous activities induced by needling at Hand Taiyin Lung Channel, and to preliminarily explore the possible brain function network of Hand Taiyin Lung Channel.
METHODSBy using functional magnetic resonance imaging (fMRI), 16 healthy volunteers underwent resting-state scanning (R1) and scanning with retained acupuncture at Hand Taiyin Lung Channel (acupuncture, AP). Data of fMRI collected were statistically calculated using amplitude of low frequency fluctuations (ALFF).
RESULTSUnder R1 significantly enhanced ALFF occurred in right precuneus, left inferior parietal lobule, bilateral superior temporal gyrus, bilateral middle frontal gyrus, left superior frontal gyrus, left inferior frontal gyrus, left medial frontal gyrus. Under AP significantly enhanced ALFF occurred in right precuneus, bilateral superior frontal gyrus, cerebellum, bilateral middle frontal gyrus, right medial frontal gyrus, and so on. Compared with R1, needing at Hand Taiyin Lung Channel could significantly enhance ALFF in right gyrus subcallosum and right inferior frontal gyrus. Significant decreased ALFF appeared in right postcentral gyrus, left precuneus, left superior temporal gyrus, left middle temporal gyrus, and so on.
CONCLUSIONNeeding at Hand Taiyin Lung Channel could significantly change fixed activities of cerebral cortex, especially in right subcallosal gyrus, right inferior frontal gyrus, and so on.

7.Levels of hemoglobin concentration and anemia during pregnancy in Lhasa

Objective To investigate the distribution of hemoglobin(Hb)and prevalence of anemia in pregnant women living in the Tibetan Plateau area.Methods Random sampling method was used and fingertip blood sample was tested.Related sociodemographic information was collected.We adjusted the hemoglobin measurements based on the different levles of altitude,using three different methods from CDC.Dirren's and Dallman's to estimate the prevalence rates of anemia.Results Three hundred and eighty pregnant women were included.The mean Hb was 127.6 g/L.Tibetan had a lower Hb (126.6 g/L)than people from other ethnicities(134.6 g/L)with rural pregnant women having a higher Hb(130.4 g/L)than that of urban ones(125.9 g/L).After controlling the gestational age,results remained the same.Different method used for correction of hemoglobin would cause different prevalence rates of anemia which were all higher than the pre-adjusted prevalence rates:70.0% for CDC method,77.9% for Dirren's method and 41.3% for Dallman's method respectively.After analyzing the erythrocyte arameters,we could not conclude that population in our study was under the early stage of the iron deficiency anemia.Conclusion Higher altitude could affect the levels of hemoglobin concentration significantly.Currently adopted hemoglobin correction methods might not be suitable for Tibetan pregnant women.We suggested that it was imperative to establish a relationship between altitude and hemoglobin of Tibetan women during pregnancy in order to correctly evaluate the prevalence of anemia.
8.Case-Control Study on the Fibroblast Growth Factor Receptor 2 Gene Polymorphisms Associated with Breast Cancer in Chinese Han Women.

PURPOSE: Genetic variation in fibroblast growth factor receptor 2 (FGFR2) is a newly described risk factor for breast cancer. This study aimed to evaluate the association of four single nucleotide polymorphisms (SNPs) in FGFR2 with breast cancer in Han Chinese women. METHODS: Two hundred three women with breast cancer and 200 breast cancer-free age-matched controls were selected. Four SNPs (rs2981579, rs1219648, rs2420946, and rs2981582) and their haplotypes were analyzed to test for their association with breast cancer susceptibility. The presence of the four FGFR2 SNPs was determined by polymerase chain reaction-restriction fragment length polymorphism analysis. RESULTS: A statistically significant difference was observed in the frequency of rs2981582 in the FGFR2 gene (p<0.05) between case and control groups. In subjects stratified by menopausal status, rs2981582 TT, rs2420946 AA, and rs1219648 CC were significantly associated with the risk of breast cancer in postmenopausal subjects, but no significant associations between these four SNPs and the risk of breast cancer were identified in premenopausal subjects. Further, there was no significant association between hormone receptor status (estrogen receptor and progesterone receptor) and breast cancer risk. Six common (> 3%) haplotypes were identified. Three of these haplotypes, CGTC (odds ratio [OR], 0.613; 95% confidence interval [CI], 0.457-0.82; p=0.001), TGTC (OR, 6.561; 95% CI, 2.064-20.854; p<0.001), and CATC (OR, 12.645; 95% CI, 1.742-91.799; p=0.001) were significantly associated with breast cancer risk. CONCLUSION: Our findings indicated that the SNP rs2981582 and haplotypes CGTC, TGTC, and CATC in FGFR2 may be associated with an increased risk of breast cancer in Han Chinese women.

9.Dynamic changes of renal blood perfusion in Goto-Kakizaki rats using contrast-enhanced ultrasonography

Objective To explore dynamic changes of renal cortex blood perfusion in Goto-Kakizaki rats(GK rats) by contrast-enhanced ultrasonography.The clues for early diagnosis and intervention of diabetic renal impairment may be obtained from the study.Methods Male GK rats were taken as experimental groups,and Wistar male rats were used as controls.Contrast-enhanced agent SonoVue was administered via the femoral vein cannulation in a bolus fashion.Image changes of renal cortex were observed in real time, and dynamic image changes within 0 - 5 minute after injection were observed and stored.The renal cortex blood perfusion changes were analyzed using the time-intensity curve (TIC).Results (1) The kidney was quickly enhanced after the injection of contrast agent.The renal artery, cortex, renal vein were displayed in sequence.(2)The shape of renal perfusion TIC in all the objects was a single peak,steep ascending, flat descending curve.The overall shape of TIC in GK rats was wider than controls and they were wider and wider with age increasing of the GK rats.(3) Compare to controls,the time to peak (TTP)and the half time of peak descending (HDT) in GK rats was longer (P <0.01), the slope rates of the ascending curve (S1) and the slope rates of the descending curve (S2) decreased and the area under the curve increased in 12-week-old and 20-week-old GK rats (P < 0.05 or P < 0.01).Conclusions The abnormal blood perfusion of renal cortex microcirculation was present in the early stage of diabetic in GK rats.Real-time contrast-enhanced ultrasonography and TIC could display this change effectively.
10.Effects of music therapy on depression and duration of hospital stay of breast cancer patients after radical mastectomy.

BACKGROUNDBreast cancer remains the most important cancer among women worldwide. The disease itself and treatment may have a profound impact on the patients' psychological well being and quality of life. Depression is common in breast cancer patients and affects the therapeutic effects as well as prolongs the duration of hospital stay. However, few studies reported the effectiveness of music therapy on depression and duration of hospital stay of female patients with breast cancer after radical mastectomy.
METHODSOne hundred and twenty subjects were recruited to this clinical trial and randomly allocated to two groups. The experimental group (n = 60) received music therapy on the basis of routine nursing care, whereas the control group (n = 60) only received the routine nursing care. The whole intervention time was from the first day after radical mastectomy to the third time of admission to hospital for chemotherapy. Data of demographic characteristics and depression were collected by using the General Questionnaire and Chinese version of Zung Self-rating Depression Scale (ZSDS) respectively. One pre-test (the day before radical mastectomy) and three post-tests (the day before discharge from hospital, the second and third admission to hospital for chemotherapy) were utilized. Duration of hospital stay was calculated from the first day after radical mastectomy to the day of discharged from hospital.
RESULTSThe mean depression score of all subjects was 37.19 ± 6.30. Thirty-six cases (30%) suffered from depression symptoms, with 26 (72.2%) mild depression cases, 9 (25.0%) moderate depression cases, and 1 (2.8%) severe depression case. After music therapy, depression scores of the experimental group were lower than that of the control group in the three post-tests, with significant differences (F = 39.13, P < 0.001; F = 82.09, P < 0.001). Duration of hospital stay after radical mastectomy of the experimental group ((13.62 ± 2.04) days) was shorter than that of the control group ((15.53 ± 2.75) days) with significant difference (t = -4.34, P < 0.001).
CONCLUSIONSMusic therapy has positive effects on improving depression of female patients with breast cancer, and duration of hospital stay after radical mastectomy can be reduced. It is worthy of applying music therapy as an alternative way of nursing intervention in clinical nursing process of caring female patients with breast cancer.
