Chinese Journal of Neurology 2000;0(04):-
Objective To investigate the apoptosis of peripheral blood lymphocyte cells in patients with HAM/TSP and the roles of Fas/FasL system, TNF ? and Bcl 2 protein in occurrence and progression of HAM/TSP Methods The single cell gel electrophoresis (comet assay) was used to detect the apoptosis of lymphocyte cells in patients with HAM/TSP The presence of sFas, sFasL in the serum and cerebrospinal fluid (CSF) of patients with HAM/TSP was evaluated by using an enzyme linked immunosorbent assay (ELISA) The Fas, TNF ? and Bcl 2 protein expressions were quanlitatively determined by flow cytometry from 8 cases of HAM/TSP, and compared with the controls and other inflammatory neurological diseases (OIND) Results (1) The apoptotic ratio of lymphocyte cells in patients with HAM/TSP (13 63%?0 48%) was significantly increased as compared to the controls (1 63%?0 57%, P 0 05) Levels of sFas and sFasL in CSF from patients with HAM/TSP ((0 19?0 07) ng/ml and (0 24?0 08) ng/ml) were both significantly increased than those from OIND ((0 11?0 06) ng/ml and (0 01?0 01) ng/ml, P 0 05) or patients with OIND (34 84?9 63, P
2.Kinase modulators approved by FDA in 2022
Acta Pharmaceutica Sinica 2024;58(1):61-75
The FDA approved a total of 37 new drugs in 2022, including 22 new molecular entities and 15 new biological products. This is the year with the lowest number of new drugs approved by the FDA since 2017. Among these approved drugs, 21 new drugs belong to the "first-in-class" category, accounting for 56% of the total approved drugs, which is the highest ratio in the past 10 years. Among the drugs approved in 2022, there are 5 small molecule kinase modulators, including the tyrosine kinase 2 (TYK2) allosteric inhibitor deucravacitinib, the first oral pyruvate kinase (PK) activator mitapivat, the Janus kinase 1 (JAK1) selective inhibitor abcrocitinib, the JAK2 selective inhibitor pacritinib and the broad-spectrum fibroblast growth factor receptor (FGFR) inhibitor futibatinib. This review briefly describes the discovery background, research and development process, synthesis routes and clinical efficacy and safety of small molecule kinase modulators approved by the FDA in 2022, hoping to provide ideas and methods for further research on kinase modulators.

Objective To measure the renal cortex blood flow leVel in Patients with stage 3-4 chronic kidney disease, and to obserVe the effect of Probucol on renal cortex blood flow and renal function. Methods Twenty Patients with stage 3-4 chronic kidney disease in our hosPital were randomly diVided into treatment grouP ( 10 Patients ) and obserVation grouP ( 10 Patients). Ten healthy subjects were chosen as the control grouP. Dynamic sonograPhic tissue Perfusion measurement (DTPM) was used to eValuate the mean flow intensity (Imix),mean flow Velocity (Vmix) and tissue resistance index (TRI) in the three grouPs. Patients in the obserVation grouP were treated with standard theraPy;the treatment grouP was giVen Probucol 500 mg,twice daily in addition to standard theraPy for 6 months. The renal cortex blood flow leVel after treatment was measured in the treatment grouP and obserVation grouP. The serum BUN,SCr,TC,TG,LDL,HDL,hs_CRP,ox_LDL,PA,ALB before and after treatment were measured and comPared between the treatment grouP and obserVation grouP. Results Imix and Vmix were significantly lower, and TRI was significantly higher in the treatment grouP and obserVation grouP than in the control grouP (P<0. 01). After 6 months of treatment,Imix and Vmix in the treatment grouP were significantly increased and TRI significantly decreased as comPared with the baseline and the control grouP (P<0. 01). After treatment,Imix and Vmix were significantly decreased and TRI was significantly increased in the obserVation grouP (P<0. 05). In the treatment grouP,the serum BUN,SCr,TC,TG,LDL,hs_CRP and ox_LDL decreased and PA,ALB and HDL increased after treatment as comPared with the baseline and the obserVation grouP (P<0. 05 or P<0. 01). Conclusion The renal cortex blood flow leVel is significantly lower in Patients with stage 3-4 chronic kidney disease than in the healthy PeoPle. Probucol can imProVe the renal cortex blood flow leVel significantly,Protect the renal function, and imProVe microinflammatory state, nutrition status and dysliPidemia.

AIM: To analyze the pathogenic bacteria and drug sensitivity in cases of canalicular inflammation.
●METHODS: Lacrimal sac secretion from 57 cases ( 57 eyes) with canalicular inflammation. used to do bacterial cultures and drug sensitivity tests. Grind open the sulfur particles from canaliculus for bacterial smear.
●RESULTS:After squeeze canalicular, there are 56 sulfur granules from 57 patients. All of the Sulfur particles smears were found in actinomycetes. A total of 55 from 57 cases of lacrimal secretions for bacterial culture were positive, and 63 strains were cultured. The main pathogen are Staphylococcus epidermidis, Streptococcus viridans and pneumococcus. Drug susceptibility test results showed that:rifampicin, cefoxitin, chloramphenicol, and mezlocillin are sensitivity.
●CONCLUSION:Actinomycetes were the main pathogens to canalicular inflammation, and most of the presence of co- infection with other bacteria. Rifampin, cefoxitin, chloramphenicol, and mezlocillin are sensitivity canalicular inflammation.

Objective To evaluate the predicting value of the marker of endothelial injury: plasma matrix metalloproteinase-9( MMP-9) and high sensitivity C-reactive protein( hsCRP) level on the progression of acute anterior circulating territory infarction progressing to malignant middle cerebral artery infarction (m-MCAI). Methods 90 patients with acute anterior circulating territory infarction, in which 46 patients progressed to m-MCAI, were collected and sampled consecutively. The plasma MMP-9 and hsCRP of all patients were determined by ELISA and immunotur-bidimetry,respectively,at admission. And the clinical characters and cranial CT features of the patients were analyzed. Results At admission, the plasma MMP-9 level in the patients with m-MCAI(242.0 ±58.0)ng/ml was significantly higher than that in the patients with non m-MCAI( 169.0 ± 50.0) ng/ml( P < 0. 01) ,the plasma hsCRP level in the patients with m-MCAI(6.25 ±1.2) ng/ml was significantly higher than that in the patients with non m-MCAI( 1.55 ± 0.9) mg/ml( P <0. 01).Conclusion The increased level of plasma MMP-9 and hsCRP could be predictors for the m-MCAI proceeding.

The efficient and safe delivery of drugs to the therapeutic site through the biofilm has traditionally been a difficult and hot topic in the field of drug delivery. In recent years, alkyl polyglycoside (APG) have become ideal penetration enhancers for drug delivery systems because of their high permeability, good safety and biodegradability, which has attracted wide attention of domestic and foreign researchers. In this paper, the physical and chemical properties, characteristics, action mechanism and application of APG in drug delivery system are reviewed, and its application prospect in drug delivery system is prospected.
