1.Preparation and Quality Control of Thermo-sensitive Ornidazole Hydrogel

OBJECTIVE: To prepare thermo-sensitive ornidazole hydrogel and establish its quality control method.METHODS: Thermo-sensitive ornidazole hydrogel was prepared with ornidazole as chief constituent using poloxamer 407 and poloxamer 188 as base.The content of ornidazole in the hydrogel was determined by ultraviolet spectrophotometry.RESULTS: The preparation was white or yellowish semisolid gel,and its test results were up to the related standard specified in Chinese Pharmacopeia(2005 Edition).The linear range of ornidazole was 3.98~43.77 mg?L-1(r=0.999 8),and its mean recovery was 98.52%(RSD=1.1%).CONCLUSION: The preparation is simple and feasible in preparation process,and the quality of the preparation is stable and controllable.
2.Establishment of anin vivo rabbit model of intervertebral disc degeneration under tensile load and its significance

BACKGROUND:Abnormal stress is an important factor causing intervertebral disc degeneration. To establish an ideal in vivoanimal model of intervertebral disc degeneration is of great significance for in-depth study on the related pathogenesis. OBJECTIVE:To develop anin vivo rabbit intervertebral disc model and to investigate the relationship between continuous tensile load and intervertebral disc degeneration. METHODS:Twenty-five New Zealand white rabbits aged 6 months old were randomly divided into three groups: blank control (n=5), sham (n=10) and experimental (n=10) groups. The blank control group received no intervention, and the L4/5 segments were removed at the 1st day. The intervertebral disc assistor was used to fix the L4 and L5 vertebral bodies in the experimental and sham groups, the L4/5 segments in the experimental group were loaded 1 MPa axial tensile force, and the L4/5 segments in both two groups were then removed at 14 and 28 days. The changes of L4/5 intervetebral space height and surrounding bone substance were observed by X-ray examination, the morphological changes of the intervertebral disc were observed by hematoxylin-eosin staining, the cell survival was detected by nitro blue tetrazolium staining and mRNA expression levels of aggrecan, collagen type Ⅱ and SOX9 in the intervertebral disc tissues were assessed by RT-PCR at each time point. RESULTS AND CONCLUSION:The radiological manifestations, histological changes, cell survival and mRNA expression levels of aggrecan, collagen type Ⅱ and Sox9 showed no significant difference between the blank control and sham groups. Comparied with the blank control group, in the experimental group, the L4/5 intervertebral space was narrowed with time, the articular surface was coarse, and the upper and lower corpus vertebrae edge appeared to have lip-shaped hyperplasia; the intervertebral disc cells distributed irregularly; the nucleus pulposus was in dehydration and deflation, annulus fibrosus arranged irregularly, and the vacuoles in notochord cells tended to disappear; the expression levels of aggrecan, collagen type Ⅱ and SOX9 were markedly downregulated. These findings suggest that the in vivo rabbit model of intervertebral disc is successfully established, in which continuous mechanical tensile load is further proved to directly cause intervertebral disc degeneration.
3.Mixed linear model and its application to repeated measurements in single-sample

Objective To explore analysis method of repeated measurements in single-sample. Methods Mixed linear model was presented and an example of repeated measurements in single-sample was analyzed. Results The reasonable results were obtained for repeated measurements in single-sample by the methods of mixed linear model. Conclusion Mixed linear model can be used to analyze repeated measurement data in single-sample.
4.Clinical study of congestive heart failure accompanied with malnutrition With danshenfenzhen and nutritional support

Objective To observe the clinical therapeutic effect of danshenfenzhen and nutritional support in treating congestive heart failure accompanied with malnutrition on the basis of common treatment.Methods 42 cas- es of congestive heart failure accompanied with malnutrition were randomly divided into 2 groups:test group 21 cases (danshenfenzhen and nutritional support) and control group 21 cases(free diet),in the test group intravenous drip danshenfenzhen and vein nutrition were given if the internal nutrition can not provide enough energy on the basis of count of energy consumption,the serum albumin,total blood lymph cells and body weight were measured.Results After 14 days' treatment in the test group,the serum albumin,body weight and the total lymph cell increased signifi- cantly while in the control group,the serum albumin,body weight and the total lymph cells decreased significantly. Conclusion Intravenous drip danshenfenzhen and nutritional support can improve nutritional status and cardiac function of patients with congestive heart failure accompanied with malnutrition.
5.Analysis on medication principles for cough based on experience of Xu Di-hua, descendant of Meng He Medical School.

Based on the software of traditional Chinese medicine inheritance support system (TCMISS), this article aims to analyze the experience and composition rules for cough from the descendant of Meng He Medical School, Xu Di-hua. The cough cases treated by Xu Di-hua were collected, and recorded into TCMISS (V2.0). Data mining methods such as Apriori algorithm and complex system entropy cluster were used to analyze the medication principles of Xu Di-hua for cough from pathogenesis and therapeutie aspects, and dig out the frequency of the herbs in prescription, core medicine and new combinations. The experience of curing cough from Professor Xu Di-hua were well found in the research. He is good at choosing prescriptions accurately, and pays attention to simultaneous use of cold and moisture drugs with combination of tonification and purgation. He is skilled in adding or reducing materia medica flexibly, as well as regulating lung to relieve cough and eliminating phlegm by clearing heat.

7.Influence of crosstalk phenomenon on the measurement of gross alpha and gross beta radioactivity in drinking water

Objective To study the influence of crosstalk phenomenon on the measurement of gross radioactivity in drinking water.Methods The gross activity in different standard materials with different thickness and area was measured using national standard method.Results There was no obvious change in crosstalk factor with the increase of 241Am powder amount in the measurement,whereas the larger amount of uranium used might lead to larger crosstalk factor.The different measurement channels resulted in different crosstalk factors.The influence of beta radioactivity on alpha radioactivity measurement was significant.On the contrary,the alpha-to-beta crosstalk factor was negligible.The area of sample plate imposed no significant influence on crosstalk factor.Conclusions The gross beta activity can be corrected to decrease the influence of alpha radioactivity using powder standard samples,when simultaneous alpha and beta counting mode is applied in measurement grass radioactivity in drinking water.
8.The survey of gross radioactivity level in drinking water in Inner Mongolia Region

Objective To ascertain the gross α/β levels in drinking water in Inner Mongolia and to estimate the annual effective dose to the local residents from radionuclides in drinking water.Methods A total of 768 water samples were collected from 101 counties distributed over 12 cities of Inner Mongolia.Low background α/β measuring instrument was used to measure the radioactivity level;On this basis,use EPA Federal Guidance Report 11 universal method to estimate the annual effective dose to the local residents via intake of radionuclides from drinking water.Results The gross α radioactivity range was 0.016-1.003 Bq/L for tap water,O.016-0.975 Bq/L for factory water,0.017-1.544 Bq/L for river water,0.120-0.672 Bq/L for lake water,0.016-0.492 Bq/L for reservoir water,0.016-1.139 Bq/L for well water,0.032-3.156 Bq/L for spring water,respectively.The gross β radioactivity range was 0.030-0.828 Bq/L for tap water,0.031-0.571 Bq/L for factory water,0.066-0.873 Bq/L for river water,0.169-2.268 Bq/L for lake water,0.046-0.519 Bq/L for reservoir water,0.071-0.526 Bq/L for well water,0.087-1.063 Bq/L for spring water,respectively.Conclusions In Inner Mongolia,the gross α/β mean value in tap water is less than the World Health Organization-recommended value and the average annual effective dose from tap water is also less than the WHO-recommended value O.1 mSv/a.The gross α/β radioactivity from the other water samples is also within the range of the nationwide average.
9.Expression of multi-drug resistance genes in cervical cancer before and after neoadjuvant chemotherapy and its relationship with the effect of chemotherapy

Objective To explore the impact of preoperative neoadjuvant chemotherapy on the expressions of multi-drug resistance genes in patients with cervical cancer and its relationship with the effect of chemotherapy. Methods Ninety-eight cervical cancer patients with TP regimen selected to perform preoperative chemotherapy were enrolled in the Affiliated Hospital of Guiyang Medical College between January 2010 and June 2014. Immunohistochemisty (En vision method) was used to determine the expressions of P-gP, GST-π and TopoII of the same patients before and after neoadjuvant chemotherapy and explore the relationship with the effect of chemotherapy. Results The positive expression rates of P-gp and GST-π were 71.43% and 64.29% before chemotherapy and 80.61%and 74.49%after chemotherapy, respectively. The former two had significant differences (P<0.01). The positive expression rates of TopoII was 48.98%before chemotherapy and 28.57%after chemotherapy , respectively, showing significant differences (P < 0.01). The expressions of P-gp, GST-π and TopoⅡ gene were not affected by the clinical and pathological features of cervical cancer (P > 0.05). Before neoadjuvant chemotherapy, the positive expression of GST-π in the ineffective group was statistically higher than that in the effective group (P<0.05). The positive expressions of P-gp and Topo II showed no statistical significance between the effective group and the ineffective group (P > 0.05). There was significant correlation in the expressions of P-gp, GST-π and TopoⅡ(P < 0.05) before and after neoadjuvant chemotherapy. Conclusions The expression of P-gp, GST-πand TopoⅡgene may not be affected by the clinical and pathological features of cervical cancer, but may change expressions of multi-drug resistance genes in cervical cancer by neoadjuvant chemotherapy. Monitoring their expression has a guiding significance for drug selection, prognostic judgment, and the following treatment regimen decision. The GST-π, expression level can be used as a biological parameter to predict the effect of TP regimen neoadjuvant chemotherapy.
10.Design of the nurse occupation well-being scale and evaluation of its reliability and validity

Objective To prepare an occupation well-being scale for nurses, and test its reliability and validity. Methods After literature review and semi-structured interviews, the item pool of the occupation well-being scale for nurses was created based on theory of structure of the subjective well-being. After group discussion, and evaluation by the specialists, a pilot scale was obtained. A total of 350 clinical nurses were selected by using stratified sampling method, and their occupation well-being were surveyed by the scale. The data were used to assess the reliability and validity of the scale. Results The occupation well-being scale for nurses had 3 dimensions, including 8 exploring factors. The values of Cronbach Alpha and the split half reliability coefficient of the total scale, endogenous exogenous satisfaction, satisfaction of nurses occupation happiness were all larger than 0.9, which meaned the internal consistency was good. In the emotion dimension, the value of Cronbach Alpha and the split half reliability coefficient were less than 0.7, which meant the internal consistency was not so good. The reliability coefficient of repeated measurement of the whole scale and each dimension were greater than 0.7, suggesting the external consistency of the scale was good. The content validity index of 3 dimensions were all larger than 0.9,and the cumulative ex-planatory variables were all larger than 55%. Conclusion The occupation well-being scale for nurses proposed in this study has good reliability and validity, and can be used to evaluate the occupation well-being of the nurses.
