3.Value of Heparin Blocking Attachment of Respiratory Syncytial Virus to Human Airway Epithelial Cells

Objective To investigate the mechanisms of attachment of respiratory syncytial virus(RSV) to human airway epithelial cells.Methods Attachment of RSV to CFBE was quantitatively assessed by flow cytometry.Various polyclonal and monoclonal antibodies to RSV and heparin were pre-incubated with RSV.The blocking effects of these antibodies and heparin on attachment were evaluated.Results CFBE cells reduced capacity being bound by RSV.All the antibodies used were failed to block attachment of RSV on CFBE,whereas heparin blocked RSV attachment in a dose-response manner and the blokade by heparin was almost complete at the concentration of 0.4 mg/L.Conclusions Flow cytometry provides direct evaluation of attachment without growth of virus.Heparin-like molecules on cell surface of CFBE appears to be involved in attachment between RSV and human epithelial cells.
4.Expression of Fas/FasL in Children with Aplastic Anemia and Its Clinical Significance

Objective To investigate the expression of Fas/FasL in children with aplastic anemia(AA) and its clinical significance.To explore the pathogenesis of AA.Methods CD34+ cell counts and the expression of Fas/FasL in 12 children with severe AA(SAA),18 with chronic AA(CAA)and 10 normal children were detected by flow cytometry combining monocolon antibody.Results The expression of Fas in children with AA were significantly higher than that in control group(P0.05).Conclusions Fas/FasL systeam take part in inducing CD34+ cell apoptosis.This may contribute to understanding the decrease number of stem cells and bone marrow failure.

9.Relationship between Apoptosis of Bone Marrow Cells and Tumor Necrosis Factor-?,Fas/FasL Expression in Children with Aplastic Anemia

Objective To investigate the relationship between apoptosis of bone marrow cells and tumor necrosis factor-?(TNF-?),Fas/FasL expression in children with aplastic anemia.Methods The concentration of TNF-? in supernatant of cultured bone marrow mononuclear cell(BMMC) was measured by enzyme linked immunosorbent assay.The expression of Fas/FasL,apoptosis ratio of bone marrow CD34+ cell were evaluated by flow cytometry.Results The concentration of TNF-? in supernatant of cultured BMMC in AA group[SAA (27.9?8.20) ng/L;CAA (23.6?6.78) ng/L] increased significantly compared with control group(t=3.432 P0.05).Apoptosis ratio of CD34+ cell in SAA group[(24.6?9.56)%] and CAA group[(20.9?7.32)%] significantly increased compared with control group(t=3.492,3.458 Pa0.05).The concentration of TNF-? was positively correlated with the expression of CD34+ Fas+ in all types AA children(r=0.542 P0.05).Conclusions TNF-? and Fas/FasL system can be involved in inducing CD34+ cell apoptosis.This may contribute to understanding the decreased number of stem cells and bone marrow failure.
10.Xylometazoline hydrochloride nasal spray combined with laser artificial nasolacrimal duct implantation for nasolacrimal duct obstruction

AIM: To study the role of xylometazoline hydrochloride nasal spray in combination therapy of nasolacrimal duct obstruction and to investigate the effect of nasal inflammation on nasolacrimal duct obstruction.
METHODS: Totally 279 patients with nasolacrimal duct obstruction were collected, who received lacrimal passage irrigation, CT angiography for lacrimal passage and nasal endoscope before treated by lacrimal laser forming and artificial nasolacrimal duct implantation combined with xylometazoline hydrochloride nasal spray. In group A, 137 patients were treated with antibiotic eye drop and non-steroidal anti-inflammatory drugs after operations. In group B, 142 patients were treated with xylometazoline hydrochloride nasal spray besides the same treatment for group A.
RESULTS:In the 279 patients 217(77.8%), in which 105 cases(76.6%)were in group A and 112 cases(78.9%)were in group B, were suffered with nasal inflammation, including nasal mucosal hyperemia, inferior turbinate hypertrophy, middle turbinate hypertrophy. At 3mo after the ducts were drawn, efficacy of group B was 95.8%, which was significant better than that of group A(86.1%, P<0.05).
CONCLUSION: Nasal inflammation was an important factor in the incidence of nasolacrimal duct obstruction, which shoud pay more attention in the process of diagnosis and treatment. Combination therapy could improve the cure rate of nasolacrimal duct obstruction.
