1.Effect of red peony root decoction on rats with severe acute pancreatitis

Objective To observe the therapeutic effect of red peony root decoction (RPRD) on model rats with acute pancreatitis (AP),and the effect on the expression of NF-κB,TNF-α and IL-6.Methods Forty SD rats were randomly divided into five groups:normal control group (group A),AP model groups (group B,24 h; group C,72 h) and RPRD treatment groups (group D,24 h; group E,72 h).There were 8 rats in each group.The AP models were induced by intraperitoneally injecting with L-arginine.After the model was established,rats of group B and group C were treated with intra-gastric instillation of NS,and rats in group D and group E with RPRD.At 24 and 72 hours after induction of AP,rats in each group were sacrificed and blood samples were taken for the measurement of serum amylase,TNF-α and IL-6.The expression of NF-κB p65 protein in the pancreas were detected by Western Blot,and pathological status of the pancreas were analyzed.Results The serum amylase level in group B and group D was significantly higher than that in group A (P<0.05),and the level in group D was obviously lower than that in group B (P<0.05).The serum amylase level in group C and group E was decreased,and there was no difference between group A (P>0.05).The pathological severity and pathological scores of the pancreas in group D and group E were more lighter than those in group B and group C (P<0.05).The level of serum TNF-α and IL-6 was significantly decreased in RPRD treatment groups (P<0.05),group D compared with group B,[ (12.54±2.12)pg/ml,(552.88±81.27)pg/ml] vs.[ (20.53±4.16)pg/ml,(1255.75± 118.82)pg/ml]; group E compared with group C,[ (11.01 ±2.02)pg/ml,(582.87± 88.86) pg/ml] vs [ (23.85 ± 5.59) pg/ml,(1366.33 ± 163.16) pg/ml].The expression levels of NF-κBp65 protein in the pancreas of group D and group E were significantly lighter than those in group C at both time points (group D compared with group B,(0.07±0.055) vs (0.71 ±0.029),P<0.05; group E compared with group C,(0.15±0.038) vs (0.65±0.026).Conclusion RPRD may alleviate the pathologic changes in AP
2.Effects of Radix Paeoniae Rubra decoction on the expression of cvtokines in rats with acute pancreatitis

Objective To observe the therapeutic effects of Radix Paeoniae Rubra decoction (RPRD) on a-cute pancreatitis (AP) in rats, and to study effects of RPRD on the expression of NF-κB, TNF-α and IL-6. After the model was estabhshed, rats of group B and group C were treated with inn'a-gastric instillation of NS, and rats in group D and group E with RPRD. At 24 and 72 hours after induction of AP, rats in each group were sacri-riced and blood samples were taken for the measurement of serum amylase. The expression of NF-κB 1065 protein and TNF-α and IL-6 mRNA in the pancreas was detected by Western blotting and real-time quantitative PCR, re-spectively, and the pathological status of the pancreas was analyzed. Results The serum amylase level in group B and group D was obviously increased at 24 hours, and were significantly higher than that in group A (P < 0.05) ; the level in group D was lower thaa that in group B (P < 0.05). the level reduced at 72 hours, and there was no difference between group E and group C (P > 0.05). The pathological severity and the pathological score of pan-creatic tissue in group D and group E at 24 hours and 72 hours were more hghter than those in group B and hours C (P < 0. 05). The expression levels of NF-κB p65 protein (Western blotting) in the pancreas of group D and group E were significantly lighter than those in group B and group C at both time points (group D compared with group B, (0.07 ± 0.006) vs. (0.71 ± 0.029), P < 0.05;group D compared with B, (0.07 ± 0.006) vs. (0.71 ±0.029), P <0.05; group E compared with C, 0.18±0.014 vs. 0.65±0.026, P <0.05). The results of real-time quantitative PCR shown that the expression levels of TNF-α and IL-6 mRNA in model treatment group were significantly stronger than that in RPRD group at the two time points (P < 0.05). Conclusions RPRD is effective for the treatment of AP rats to inhibit the activity of NF-κB. Inhibition of NF-κB offers a promising strategy to treat AP by down-regulating eytokine expression.

4.Study on effect of total matrines and extracts from Periplaneta americana on negative endometrial cancer cell JEC of progesterone receptors.

To study the effect of total matrines and extracts from Periplaneta americana on negative endometrial cancer cell JEC of progesterone receptors. After detecting the effect of total matrine, extracts from P. americana and their combination on JEC cells' growth inhibition, cell cycle, P53 and c-erbB-2 gene protein expressions through MTT, flow cytometry instrument and Western blot method, the author found that, (1) MTT: total matrines and extracts from P. americana could inhibit the growth of JEC cell, with significant increase in the inhibitory effect in the combination group. (2) Flow cytometry instrument: the cell cycle at G0/G1 increased after the treatment with total matrines, the cell cycle at G2/M increased after the treatment with extracts from periplaneta americana, and the ratio of G0/G1 cell cycle in the combination group was significantly higher than the other groups, with inhibition in cell growth and statistical difference in inter-group comparison (P < 0.05). (3) Western blot: the expression level of P53 increased and c-erbB-2 decreased after the treatment with total matrines, extracts from P. americana and their combination on JEC cell, with statistical difference in inter-group comparison (P < 0.05). The above results suggested that total matrines, extracts from P. americana and their combination could induce cell cycle arrest and inhibit the growth of JEC cell by up-regulating P53 and down-regulating the c-erbB-2 level.

7.The relationship between atrial fibrillation and serum uric acid level in patients with coronary heart disease

Objective To investigate the relationship between serum uric acid level and atrial fibrillation in patients with coronary heart disease.Methods Two hundred and forty-seven inpatients with coronary heart disease were selected.All the patients were divided into simple coronary heart disease group (90 cases),coronary heart disease with paroxysmal atrial fibrillation group (85 cases) and coronary heart disease with continuous/permanent atrial fibrillation group (72 cases).The age,history of cardiovascular events,uric acid,echocardiographic characteristics and drug-taking history were carefully recorded.The risk factors of atrial fibrillation in patients with coronary heart disease were analyzed by Logistic regression.Results The level of history of smoking,diastolic blood pressure,body mass index,total cholesterol,high density lipoprotein cholesterol,low density lipoprotein cholesterol,creatinine,fasting plasma glucose,postprandial 2 h plasma glucose,interventricular septal thickness,carotid intima-media thickness and drug-taking history among the 3 groups showed no statistical differences (P > 0.05).The age,systolic blood pressure,uric acid,left atrial diameter (LAD),left ventricular end diastolic diameter (LVEDD) in coronary heart disease with paroxysmal atrial fibrillation group and coronary heart disease with continuous/permanent atrial fibrillation group were significantly higher than those in simple coronary heart disease group,the left ventricular ejection fraction (LVEF) was significantly lower than that in simple coronary heart disease group,and there were statistical differences (P < 0.01 or < 0.05).The result of Logistic regression analysis showed high uric acid,high age,expanded LAD and LVEDD were the independent risk factors of atrial fibrillation in patients with coronary heart disease,and uric acid showed the most significant correlation (P =0.001,OR =1.061,95% CI 1.026-1.096).Conclusion High serum uric acid level maybe a risk factor of atrial fibrillation in patients with coronary heart disease.
8.Study on Quality ofYuan-Hu Zhi-TongTablet from Different Manufacturers by HPLC Fingerprint

This study was aimed to evaluate the quality ofYuan-Hu Zhi-Tong (YHZT) tablet from different manufacturers by high performance liquid chromatography (HPLC) fingerprint. An HPLC method was developed to establish the fingerprint of 12 batches of YHZT from 10 manufacturers. The common peaks were confirmed by mass spectrometric analysis. Similarity analysis (SA), hierarchical cluster analysis (HCA) and principal component analysis (PCA) were applied to analyze the fingerprint chromatography. The results showed that there were 15 common peaks in which 12 components were from Corydalis Rhizoma and 3 from Radix Angelicae Dahuricae. The similar degrees of 12 batches of YHZT tablet were between 0.498 and 0.999. Based on the values, they could fall into three groups. The result of HCA showed that 12 batches of samples could be divided into 3 classes. The results of PCA indicated that the first three principal components (PCs) could represent the 15 common peaks. According to the scores of 3 PCs, 12 batches of samples could be divided into three categories. The classification result was mainly affected by the components of Corydalis Rhizoma. The classification results of three methods were basically the same. Based on the combination of three methods, 12 samples can be divided into three grades in the aspect of quality. It was concluded that the quality differences of YHZT tablet from different manufacturers were obvious. The combining application of three methods can be used in the evaluation on fingerprint of samples from different sources and influence factor analysis. It can be used as an effective method for quality evaluation.
9.Optimization of Ultrasonic Extraction of Asperosaponin VI from Dipsaci Radix by Response Surface Methodology

This study was aimed to optimize extraction technology of Asperosaponin VI from Dipsaci Radix by re-sponse surface methodology (RSM). A single-factor test and Box-Behnken design (BBD) were employed to optimize three extraction variables, including extraction time (A), ethanol concentration (B), and the ratio of ethanol to materi-al (C). The transfer rate of Asperosaponin VI was used as the evaluation index. The results showed that the optimized conditions were A of 33.13 min, B of 51.58 %, C of 23.39 mL·g-1. Under these conditions, the transfer rate was 88.39 ± 0.212% (n = 3), which was well matched with the predicted transfer rate. It was concluded that RSM can be used for extraction optimization of ultrasonic extraction process for Asperosaponin VI. This method had certain prac-tical values.
