Academic Journal of Second Military Medical University 2010;31(3):264-268
Objective To evaluate the effects of Annexin A1 on migratory and invasive ability of colon cancer cells. Methods Eukaryotic expression vector pcDNA3. 1-Annexin A1 was constructed and analyzed by restriction analysis and sequencing. The recombinant plasmid pcDNA3. 1-Annexin A1 was transfected into SW480 cells by liposome-mediated gene transferred method. The stable transfectants were obtained after screening with G418. Real-time PCR and Western blotting analysis were used to examine the expression of Annexin A1 mRNA and protein in SW480 cells before and after transfection. Wound-healing experiment and Transwell invasion assays were used to study the effects of Annexin A1 on the migratory and invasive ability of SW480 cells. Results The recombinant plasmid pcDNA3. 1-Annexin A1 was successfully constructed. The results of real-time PCR and Western blotting analysis showed that the Annexin A1 expression was significantly higher in cells transfected with pcDNA3. 1-Annexin A1 than in un-transfected cells or those transfected with empty vectors (P<0. 01), while there was no significant difference between the last two groups (P>0.05). The migratory rate of SW480 cells in pcDNA3. 1- Annexin A1 group was significantly higher than those in un-transfected cells or transfected with empty vectors ([0. 415 ± 0. 002) vs [0. 267± 0. 003] and [0. 271 ±0. 002], P<0.05), and there was no significant difference between the latter two groups. The migrating SW480 cells in pcDNA3. 1-Annexin A1 group was significantly more than those in the other two groups ([ 221. 75± 12. 07] vs [162. 80± 12. 07] and [164. 25±9. 50], P<0. 05). Conclusion Overexpression of Annexin A1 can increase the migratory and invasive ability of SW480 cell line.
2. Cancer stem cells of multiple myeloma: Recent progress
Academic Journal of Second Military Medical University 2010;30(8):953-955
Cancer stem cells (CSCs) are a small proportion of tumor cells with the property of stem cells in the tumor tissue; they are capable of self-renewal, multi-lineage differentiation and serve as the source of tumor cells and tumor tissues. The discovery of multiple myeloma CSCs and the study on its relationship with side population cells and niche provide a new interpretation on the pathogenesis of multiple myeloma.

Objective:To investigate the application of leukogenic medicine in cancer patients and standardize the use of WBC increasing medicine in a hospital. Methods:A retrospective analysis of 579 patient records from January 2014 to December in a hospital diagnosed as malignant tumor and issued WBC increasing medicine . The kinds of drugs, timing of administration, dosage, frequency of administration, the use of medication, drug combination and the choice of solvent were analysed by collecting the patients’ essential information, chemotherapy, myelosuppression etc. Results: The l white blood drugs mainly oral preparations; There were 414 rational drug use medication orders, accounting for 71. 50 percent;irrational drug use medical record 165,accounting for 28. 50% . The dosage and duration of treatment in line with requirements, and unreasonable cases mainly suitable varieties and timing of administration based,42. 42% and 27. 88% ,respectively;followed by repeated dosing and combination therapy is not suitable,13. 33% 1and 2. 73,respectively;a lower incidence of solvent choice unsuitable,is 3. 64% . Conclusion:The use of WBC increasing medicine need to be standardized in cancer patients in the hospital. Clinicians need to strengthen studying the knowledge of the application of WBC increasing medicine and use WBC increasing medicine rationally.

Objective To investigate the changes of protein kinase C(PKC) activity in peripheral blood T lymphocytes of the children with idiopathic thrombocytopenic purpura (ITP) and the relationships between PKC activity and T lymphocytes activation and thrombocyte decrease. Methods Sterilized peripheral blood were collected from ITP children (n=35) and healthy children (n=30).T lymphocytes were isolated and purified by the T cell segregation enrichment column. The total PKC activity was detected by non-radioactive assay.FasL,the T cell activated marker, was determined by flow cytometer.Platelet count was performed by hematocytometer.Result Compared with healthy children, total PKC activity in ITP children was significantly enhanced (0.97?0.21 nmol/min.ml vs 0.60? 0.13 nmol/min.ml,?s,P

Chinese Journal of Medical Ultrasound (Electronic Edition) 2012;09(6):521-524
Objective To summarize the sonographic characteristics of Peutz-Jeghers syndrome in children and investigate the diagnostic value of ultrasound. Methods Retrospective analysis of sonographic characteristics of 19 cases with pathologically-confirmed Peutz-Jeghers syndrome were conducted.ResultsExcept for one child, other 18 patients all had more than one polyp with the maximum of sixteen. The polyp with the smallest size of 0.8 cm×0.5 cm×0.8 cm was found in left colic flexure, while the largest one was found in superior rectal with a measurement of 7.6 cm×2.7 cm×3.6 cm.The polyps showed radiatiform echotexture. And hypoechoic and hyperechoic components were present alternately.Blood flow was easily identified within these polyps by color Doppler sonography.Intussusceptions were observed in 11/19 patients.Among them,10 cases were located at small intestine. The sonographic finding was a concentric structure with a polyp at the top. Conclusions The sonographic appearence of Peutz-Jeghers syndrome is characteristic. Before surgery, ultrasonographic examination could achieve a accurate diagnosis for Peutz-Jeghers syndrome.

Objective To investigate the exprestion of apoptotic signal proteins(FADD,Fas,FasL and NF-?B) in peripheral blood T lymphocytes in idiopathic thrombocytopenic purpura (ITP) children and its correlation with clinical outcome. Methods Collecting aseptic peripheral blood from ITP children (n=35) and healthy children (n=30), T lymphocytes were isolated and purified by the T cell Segregation Enrichment Column, Fas,FasL and T cell apoptosic ratio were detected by FCM. Immunohistochemical staining was used to detect the level of NF-?B and FADD.Results The expression rates of Fas,FADD in ITP children decreased, but the expression rates of FasL,NF-?B increased. The differences between ITP children and heathy children had statistics significance(P

Objective:To observe the clinical efficacy of point application with Da Huang (Rhizoma Rhei Cruda) powder at Shenque (CV 8) plus moxa-salt hot compress on the umbilicus for preventing gastrointestinal adverse reactions after chemotherapy for non-Hodgkin lymphoma (NHL).Methods:A total of 60 cases with NHL under chemotherapy were divided into two groups by hospitalization order, with 30 cases in each group. The control group was treated with routine nursing and the observation group was additionally given point application with Da Huang (Rhizoma Rhei Cruda) powder plus moxa-salt hot compress on the umbilicus,to compare the effect in preventing gastrointestinal adverse reactions after chemotherapy between the two groups. Results:The occurrence rates of constipation, nausea, vomiting and poor appetite on the second day and fifth day after chemotherapy were obviously lower in the observation group than those in the control group, with statistically significant differences between the two groups (all P<0.05).Conclusion:The point application with Da Huang (Rhizoma Rhei Cruda) powder at Shenque (CV 8) plus maxa-salt hot compress on the umbilicus can produce more significant efficacy in preventing the gastrointestinal adverse reactions after chemotherapy for NHL than routine nursing. Moreover,it is simple and easy to be used and popularized.
