AIM: To investigate the effectiveness of dosing technology and excipient application on solid drug's moisture-proof property and to provide a method improving the solid drug's moisture-proof property. METHODS: Based on the analysis of the moisture absorbing process and the absorbing mechanics,a new concept "active moisture absorbance site"(AMAS) was defined and mathematic model was constituted. Two constants as: initial AMAS concentration C_0,moisture resistance coemcient K,were proposed to indicate the drugs moisture-proof ability. The CI WU JIA extracts and SHUANG HUANG LIAN extracts were used to conduct experiments, which were designed to investigate the improvement of the drug's moisture resistance ability when varying the AMAS's concentration and/or its space distribution. RESULTS: Reducing the amount of moisture absorbance excipients in each dosage of solid drug can lower the drug's moisture absorbance ability, and applying a thin film coating material outside the drug can also slow the drug's moisture absorbance rate. CONCLUSION: The proposed mathematic model is useful for finding a way to improve the drug's moisture resistance;experimental data shows that the drug's moisture-proof property would be improved by reducing the amount of moisture absorbance excipient in each dosage and improving the moisture distribution resistance through a certain dosing technology, such as film coating technology.

China Oncology 2016;(1):97-101
Background and purpose:This study investigated the value of cervical ultrasound and TSH-sup-pressed thyroglobulin in the diagnosis of recurrence or metastasis of differentiated thyroid carcinoma (DTC).Methods:This study analyzed the data on 196 thyroid carcinoma patients who underwent neck dissection after clinically sus-pecious recurrence of DTC in Peking Union Medical College Hospital from Aug. 2010 to Dec. 2014. Among the 196 patients, 62 patients sonographically suspecious recurrence after total thyroidectomy and/or radioactive iodine ablation therapy were enrolled in this study. The ultrasonic features of lymph node involvement were retrospectively analyzed. The value of the ultrasonic features in the diagnosis of lymph node involvement and a TSH-suppressed serum Tg level in the diagnosis of recurrence or metastasis of DTC was also evaluated. Results:Of the 62 patients, 59 were pathologi-cally conifrmed with lymph node involvement, 1 case with local recurrence and 2 cases without recurrence or metasta-sis. There were 121 ultrasonographically suspected lymph nodes, conifrmed by pathology, 92 were metastatic ltsions, 25 were non-metastatic, 3 were ifbrous tissue and 1 was striated muscle. The positive predictive rate of cyst, calciifcation or hyper-echogenicity in cortex was 100% in the diagnosis of lymph node involvement. There was a signiifcant difference in disordered vascularity and cyst in cortex between metastatic and non-metastatic lymph nodes. Forty-nine patients were positive for Tg, whereas 13 were negative. The accuracy, sensitivity and speciifcity of TSH-suppressed serum Tg in the diagnosis of recurrence or metastasis of DTC were 82.3%, 81.7% and 100%.Conclusion:The disordered vascularity and cyst in cortex of the lymph node are highly speciifc indexes in diagnosing lymph node involvement. TSH-suppressed serum Tg level has high diagnostic value for detection of recurrence or metastasis of DTC. Cervical ultrasound can identify recurrent or metastatic lesions in both Tg-positive and Tg-negative patients.

Objective To study the regulatory effect of interferons(IFNs) on retinoic acid-inducible gene-Ⅰ(RIG-Ⅰ) and the roles of RIG-Ⅰ in IFNs signaling pathway. Methods RIG-Ⅰ expression before and after IFNs treatment in mouse embryonic fibroblasts(MEFs) were analyzed with Northern blotting and semi-quantitative RT-PCR assay.MEFs isolated from wild-type and RIG-Ⅰ-/-mice were used to test growth inhibition and antiviral activity of IFNs with MTT assay and cytopathic effect inhibition assay. Results Both IFN-? and IFN-? could induce RIG-Ⅰ expression in MEFs.Treated with 100 U/mL IFN-?,growth inhibition and antiviral activity of MEFs from wild-type mice were more significant than those from RIG-Ⅰ-/-mice.With the absence of RIG-Ⅰ,the antiviral protective role IFN-? plays was significantly weaker than the wild type. Conclusion RIG-Ⅰ gene is a novel mediator of interferon effects on cells. It may participate in the inflammation responses mediated by IFNs through modulating cytokines production.
