1.The Influence of Immune RNA on Endotoxin-Induced Uveitis

Objective: Our aim was to investigate whether the immune RNA (iRNA) can inhibit the endotoxin-induced uveitis (EIU). Methods: The EIU was induced by injecting endotoxin of E.Coli into rats hypodermically. iRNA was abstracted from the livers and spleens of the rabbits which had been injected by E.Coli six times. The iRNA was introduced intraperitoneally into the rats both at half an hour and 3 hours after the injection of endotoxin. The effect of iRNA was evaluated by protein determination and cell count in aqueous houmer(AH). Results: The appearance of EIU in the iRNA treated rats was delayed, about 3 hours later than the rats received only endotoxin injection. The AH protein decreased by 58.34% and cellular infiltration decreased by 27.62% in iRNA treated group at the 24 hours. Conclusion: The iRNA may partly prevent the development of EIU and significantly reduce the severity of EIU.
2.Expression of silence information regulator 1 (Sirt1) in B cells in peripheral venous blood of subacute cutaneous lupus erythematosus

Objective To investigate alterations in epigenetic regulation in B cells in subacute cutaneous lupus erythematosus (SCLE).Methods Peripheral blood mononuclear cells (PBMC) were isolated from the peripheral venous blood of SCLE patients and healthy donors with density gradient centrifugation.B cells were isolated with CD19 micro beads and protocols provided by the manufacturer.Total RNAs were isolated with RNA kits.The expressions of silence information regulator 1 (Sirt1) mRNA were investigated with quantitative real time polymerase chain reaction (PCR).Results The levels of Sirt1 mRNA were significantly increased in SCLE B cells relative to healthy controls (P < 0.01).Conclusions Histone modifications appear abnormal in B cells in SCLE.
3.Risk factors of pulmonary non-tuberculous mycobacterial infection in 489 patients with sputum acid-fast bacilli positive and/or mycobacteria culture positive

Objective To analyze the risk factors of pulmonary non-tuberculous mycobacterial (NTM) infection in sputum acid-fast bacilli positive and/or mycobacteria culture positive patients.Methods One hundred and sixty-three patients with pulmonary NTM infection were recruited from Jan 2006 to Jun 2011 in Shanghai Pulmonary Hospital and 326 patients with sputum positive pulmonary tuberculosis who were selected by random systemic sampling method in the same period were recruited as control.The data were retrospectively analyzed.The related factors were compared between groups by chi-square test.The risk factors of pulmonary NTM infection were analyzed by binary Logistic regression model.Results There were statistically significant differences of age,history of smoking,chronic obstructive pulmonary disease (COPD),bronchiectasis,thin walled cavities focus,purified protein derivative (PPD) test between patients with pulmonary NTM infection and patients with pulmonary tuberculosis (all P<0.05).Univariate analysis showed that age [45-60year (OR=2.637,95%CI:1.631-4.264; P<0.001); >60 year (OR=4.194,95%CI:2.581-6,813 ; P<0.001)],history of smoking [10-20 year (OR=1.842,95%CI:1.0843.070; P=0.024),>20 year (OR=2.040,95%CI:1.167-3.567; P=0.012)],COPD (OR=2.698,95%CI:1.588-4.583; P<0.001),bronchiectasis (OR=3.566,95%CI:2.343-5.427;P<0.001),thin walled cavities focus (OR=2.592,95%CI:1.581-4.250; P<0.001) and a weak-positive reaction of PPD test (OR=2.389,95%CI:1.276-4.472; P=0.006) were all risk factors of pulmonary NTM infection.Multivariate analysis showed that age>60 year (OR=3.961,95%CI:2.183-7.189 ; P<0.001),bronchiectasis (OR =3.880,95 % CI:2.342-6.487 ; P<0.001),thin walled cavities focus (OR=2.898,95%CI:1.567-5.360; P<0.001),COPD (OR=2.503,95% CI:1.289-4.857; P=0.007),age45-60 year (OR=2.452,95%CI:1.391 4.325; P=0.002)anda weak-positive reaction of PPD test (OR=2.295,95%CI..1.132-4.652; P=0.021) were independent risk factors of pulmonary NTM infection.Conclusion In sputum acid-fast bacilli positive and/or mycobacteria culture positive patients,age≥ 45 year,COPD,bronchiectasis,thin walled cavities focus and a weak-positive reaction of PPD test are risk factors of pulmonary NTM infection.The clinicians should pay close attention to the results of species identification.
4.Research on Pretreatment Method of Styrene Type Macroporous Resin and Its Application on Separation and Purification of Traditional Chinese Medicine

This article was aimed to study pretreatment method of styrene type macroporous resin and its application in the industrial production. The organic residues and heavy metals of styrene type macroporous resin were detected respectively by gas chromatography and atomic absorption spectrophotometry before and after pretreatment to optimize the pretreatment process. The results showed that contents of organic residues and heavy metals of styrene type macroporous resin with the preferred method of pretreatment are in line with relevant regulations. It was concluded that this pretreatment method was simple and feasible, which is suitable to be applied in the industrial production.
5.Cerebrospinal fluid adenosine deaminase activity for the diagnosis of tuberculous meningitis

Objective To evaluate cerebrospinal fluid adenosine deaminase(CSF-ADA)activity in the diagnosis of tuberculous meningitis(TMB), and to observe its dynamic changes. Methods A total of 160 patients were included and were divided into two groups: 76 cases of TBM and 84 cases of non-TBM.Among the cases of non-TBM, there were 36 cases of bacterial meningitis, 30 cases of viral meningitis and 18 cases of cryptocoocal meningitis. All the patients were measured with their CSF-ADA activity by Enzymecoupled assay(Trinder method)and 47 patients of TBM were measured again after 2 weeks' and 6 weeks'antitubercular therapy. Results were expressed as(-x)± s. Mann-Whitney U test and paired-samples t test were used. Results CSF-ADA activity in TBM group was(12.9 ±6.4)U/L, while that in the non-TBM group was(6.0 ± 4.1)U/L, the difference was of statistical significance(U = 7.860, P < 0.05). With the cutoff value of 9 U/L, the sensitivity and specificity to differentiate TBM from non-TBM was 84.21% and 83. 33%, respectively. CSF-ADA activity decreased in TBM patients after antitubercular treatment.Conclusions CSF-ADA activity can be an effective laboratory marker for early differential diagnosis of TMB with the cut-off value of 9 U/L. Dynamic changes of CSF-ADA activity may be a indicator for the effect of antitubercular treatment.
6.Acoustic Characteristics Study on Tone/i/with ContinuousIy VariabIe Pitch and Loundness Made by NormaI AduIts

Objective By analyzing the graph features of fundamental frequency at different rates of/i/with continuously variable pitch and loundness made by normal adults,we could explore that the rate and gender have some impact on the graph to lay the foundation of regulating the clinical maximum counting ability.Methods Nor-mal adults of 51 (25 males,26 females were included).After each deep breathing,the subjects respectively took sustained speech/i/ with continuously variable pitch and lounolness twice at different rates of fast,medium and slow speed.Some parameters were measured byDoctor Speechat different rates of the tone/i/:Total time(T), Mean time standard deviation (MTSD),Mean wave crest (MWC),Mean wave trough (MWT),Mean frequency range (MFR),Mean frequency range standard deviation (MFRSD),comparing the speed having some effect on those above parameters with gender.ResuIts ①With the increasing rate of speech,the tone/i/of T (P<0.001), MFR (P<0.01)and MTSD (P<0.001)were significantly decreased,MWT significantly increased (P<0.001), While the MWC and MFRSD did not change significantly (P>0.05 ).②The tone /i/ of T (P>0.05 )were not effected by gender,female's MWT (P<0.001),MWC (P<0.001),MFR (P<0.001),MFRSD (P<0.01)were obviously higher than male's,while female's MTSD were significantly larger than male's (P<0.01).ConcIusion We should consider the impact of rate of speech and gender in clinical tests and values of MCA.The maximum counting capacity can be respectively tested at three different rates of speech for each subject,but only are the tone/i/of fun-damental frequency graphs with MWT,MWC,MFR,and MFRSD in normal range with corresponding speed and gender,can we get the value or can the value be valid.
7.Optimization of the Inclusion Process of Essential Oil in Xiaoyu Anshen Capsules

OBJECTIVE:To optimize the inclusion process of the volatile oil of Pogostemon cablin and Acorus tatarinowii in Xiaoyu anshen capsule.METHODS:The inclusion process was carried out by saturated water solution method.And the inclusion process was optimized by orthogonal experiment with the inclusion ratio and yield of the volatile oil as indexes taking the ratio of volatile oil to ?-CD,the temperature and the time of inclusion as factors.The physical phase of the inclusion complex was detected by differential scanning calorimetry(DSC).RESULTS:The optimum inclusion process was established as follows:the ratio of oil to ?-CD=1∶8;the ratio of water to ?-CD=10∶1;the inclusion time was 1h and the inclusion temperature was 50 ℃.The inclusion rate was as high as 90%.The DSC revealed that a new physical phase has been formed for the inclusion complex.CONCLUSION:The intermolecular inclusion complex with good stability and high yield of volatile oil could be obtained from the optimized inclusion process.

Plant transgenic system secures a safe,economical and reliable supply of recombinant proteins.Plant oilbody expression system simplifies the downstream purification steps and reduces capital investment based on the nature of oleosin including high expression and easy extraction.The structures and characteristics of seed oil body and oleosin were reviewed.And the research progress and industry of the seed oil body expression system,as a new bioreactor,to produce valuable recombinant proteins were discussed.The benefits and questions of the oil body expression system were also set forth.New medicine haFGF based on the oilbody system is being developed,and its biological activity is being analyzed.As a new resource for medicine protein,oilbody expression system will be perfected and applied broadly.
9.Anti-depressant effect and mechanism of supercritical CO2 extract from Compound Chaigui Fang.

The tail suspension test (TST), forced swimming test (FST) and chronic unpredictable mild stress (CUMS) model were used to evaluate the anti-depressant effect of supercritical CO2 extract from Compound Chaigui Fang (FFCGF). A nuclear magnetic resonance (NMR)-based metabonomics combined with multivariate statistical analysis was performed to explore the mechanism of FFCGF. Rats were conducted by CUMS procedure for 28 days and drugs were administrated at the same time. The body weight, sucrose preference, crossings and rearings in open-field tests were evaluated and the urine was collected simultaneously. The metabonomic profiles of rats' urine were analyzed by NMR and potential biomarkers were searched by multivariate statistical analysis. The results showed that administration of FFCGF significantly decreasing the immobility time in FST and TST and improving rats' body weight, sucrose preference, crossings and rearings in CUMS, which were indication that the anti-depressant effect of FFCGF was abvious. Significant differences in the metabolic profile of the CUMS treated group and the control group were observed, which were consistent with the results of behavioral tests. Decreased levels of acetic acid, succinic acid, 2-oxidation glutaric acid and citric acid and increased glycine and pyruvic acid in urine were significantly affected by the CUMS procedure and the 6 biomarkers were reversed evidently after administration of FFCGF. These changes were suggestion that the anti-depressant mechanism of FFCGF was associated with energy metabolism, lipid metabolism and amino acid metabolism.

To investigate the therapeutic effect of artesunate on mouse cytomegalovirus pneumonia, the BALB/c-nu mice were infected with murine cytomegalovirus-green fluorescent protein (MCMV-GFP) by nose dropping method. The experimental protocol was approved by the Medical Laboratory Animal Ethics Committee of Guangzhou Medical University. The BALB/c-nu mice were randomly divided into five groups: control group, MCMV pneumonia group, and artesunate (60, 120, and 240 mg·kg-1) groups. The survival rate, weights, and virus loads in lungs among the groups were observed. The degree of histopathologic changes in lungs was assessed directly by hematoxylin-eosin (HE) assay.
