4. Research progress on Chinese materia medica in treatment of chemotherapy-induced leukopenia
Chinese Traditional and Herbal Drugs 2019;50(20):5088-5095
Leukopenia is one of the serious side effects of chemotherapy in cancer therapy, which affects the treatment progress, prognosis, and outcome of cancer patients. Clinically, it mainly relies on colony-stimulating factor drugs such as granulocyte colony-stimulating factor (G-CSF). However, with the increase of white blood cells number, its function is relatively insufficient. What’s more, the price of G-CSF is high, which is accompanied by certain toxic as well as side effects. So the clinical application is limited. Traditional Chinese medicine has unique advantages and multi-directional effects in preventing and treating chemotherapy side effects and reducing the symptoms of leukopenia. This article reviews the progress of traditional Chinese medicine in the treatment of leukopenia after chemotherapy by literature retrieval, and provides a reference for the treatment of leukopenia with traditional Chinese medicine in clinic.

AIM:To investigate the effect of ovarian carcinoma cells on ? chain expression and the secretion of Tc1/Tc2 type cytokine in CD8+ T cells, and its role in the ovarian carcinoma induced immunosuppression.METHODS:The supernatants of human ovarian carcinoma cell lines of OVCAR3,CAOV3 and SKOV3 and RPMI-1640 were added into CD8+ T cells(groups I,II,III,and control),which were isolated from the peripheral venous blood of healthy persons. The expression of ? chain was analyzed by Western blotting.Thiazolyl blue(MTT)method was used to detect the effects of those cell line supernatants on the growth of CD8+ T cells. The secretion of the Tc1 type cytokine interferon(IFN)-? mRNA and the Tc2 type cytokine interferon(IL)-10 mRNA were detected by RT-PCR.RESULTS:The expression of ? chain was significantly lower in groups I,II,and III in comparison with that in control group. The absorbance at the wavelength 570 nm of CD8+ T cells culture in the group I,II,and III was all significantly lower than that in the control group. The IFN-? expression was significantly lower in groups I,II,and III in comparison with that in control group, while the expression of IL-10 was significantly higher.CONCLUSION:Ovarian carcinoma may suppress CD8+ T cell proliferation and secretion of the Tc1/Tc2 type cytokine through inhibition of ? chain, which may play an important role in the ovarian carcinoma induced immunosuppression.

ObjectiveTo investigate the effects of 1-3-n-Butylphthalide on the blood-brain barrier (BBB) following whole brain irradiation in rats.Methods144 male Sprague Dawley rats were randomly divided into sham-irradiation group, irradiation group,1-3-n-Butylphthalide group, and irradiation plus 1-3-n-Butylphthalide group.Whole-brain irradiation was given as a single-dose of 10 Gy using 4 MV X-ray.The rats were injected intraperitoneally with 1-3-n-Butylphthalide at 0.3 mg/kg,1.0 mg/kg,3.0 mg/kg once per day.The changes of the BBB were assessed by Evans blue (EB) assay. The expression of vascular endothelial growth factor (VEGF) in the brain tissue was determined by immunohistochemistry. The circulating endothelial cells (CECs) isolated from right ventricular blood were counted.MRI was evaluated with the T1-weighted images,T2-weighted images and MRI enhancement images induced by Gd-DTPA.The data were compared among the groups through Student-Newman-Keuls test.ResultsCompared with the sham-irradiation group, the EB content, the expression of VEGF in the brain tissue and the CECs were significantly increased in the irradiation group (2.81∶ 7.82,P =0.002;5.83∶ 10.26,P=0.003;3.16∶6.14,P =0.002).The signal intensity of T1-weighted images was significantly decreased while T2-weighted images and the enhancement rate significantly increased in the irradiation group (P =0.004 -0.018 ).Compared with irradiation group, the EB content, the expression of VEGF and the CECs were decreased significantly in the irradiation plus 1-3-n-Butylphthalide group ( 7.80∶ 3.86,P =0.007 ; 10.83 ∶ 5.26,P =0.008 ;6.36∶ 3.64,P =0.009 ).However, the changes in the MRI were significantly attenuated ( P =0.008-0.026,and 0.006 -0.038,respectively).Conclusions Following whole brain irradiation,1-3-n-Butylphthalide can decrease the permeability of the BBB in rats via decreasing VEGF expression and decreasing the CECs.

OBJECTIVETo investigate the effect of miRNA-101 on the expression of the enhancer of zeste homolog 2 (EXH2) in human androgen-independent prostated cancer LNCaP cells.
METHODSWe divided LNCaP cells into a blank control, a negative control, and a miRNA-l01 transfection group, constructed the vector by transfecting synthetic miRNA-101 mimics into the LNCaP cells, and evaluated the efficiency of transfection by fluorescence microscopy. Then we determined the expression level of EZH2 mRNA by qRT-PCR in the three groups of cells and that of the EZH2 protein in the negative control and transfection groups by Western blot.
RESULTSGreen fluorescence signals were observed in over 70% of the LNCaP cells in the transfection group after 24 hours of transfection. At 72 hours, the expression of miRNA-101 was significantly upregulated in the transfected cells (P < 0.01), that of EZH2 mRNA was remarkably lower in the transfection group (0.01 ± 0.10) than in the blank control (0.95 ± 0.40) and negative control (0.86 ± 0.30) groups (both P < 0.01), and that of the EZH2 protein was increased in the negative control but decreased in the transfection group with the extension of culture time.
CONCLUSIONmiRNA-101, with its inhibitory effect on the expression of EZH2 in LNCaP cells, is a potential biotherapeutic for prostate cancer.
