Objective To study the molecular mechanism of the remission in children with acute lymphocyte leukemia(ALL),as well as the expression of thyroid hormone receptor interacting protein 3(TRIP3) gene in children with ALL and explore the relationship between TRIP3 and ALL.Methods Fasting venous blood 2-4 mL was collected,anticoagulanted with ethylenediamine tetraacetic acid(EDTA),then perpheral blood mononuclear cells was collected by Ficoll density gradient centrifugation,total RNA was extracted by Trizol one step method.Reverse transcription-polymerase chain reaction(RT-PCR) was used to detect TRIP3 expression in peripheral blood lymphocytes in 73 ALL children of different stages and 20 normal children.The relationship between TRIR3 expression in children and ALL release was analyzed.Results 1.Expression of TRIP3 was significantly lower both after initial treatment and during recrudescence than that in normal children(Pa0.05).3.In children with ALL after initial treatment,the remission rate was significantly higher in TRIP3 positive patients than in TRIP3 negative ones(remission rate discern 25.0% vs 84.2%,P
2.Comparison of the Curative Effect of Concurrent Chemoradiotherapy and Sequential Chemoradiotherapy in Non-small Cell Lung Cancer

Objective To analyze the curative effect and toxicity of concurrent chemoradiotherapy and sequential chemoradiotherapy in non-small cell lung cancer patients.Methods 64 patients with unresectable stage Ⅲ non-small cell lung cancer in our hospital between May 2010 and Mar 2013 were randomly assigned into concurrent group and sequential group for this test.Concurrent group patients (32 cases) were given Cisplatin and Gemcitabine on the 1,4,7 week during radiotherapy.Sequential group patients accepted 3 cycles chemotherapy before radiotherapy.Results The response rate was 68.8% in concurrent group,and 43.8% in sequential group.Difference was significant between the response rates of the two groups (P < 0.05).There was significant difference in haematological toxicity,which was 100% in concurrent group and 65.6% in sequential group (P < 0.05).There was no significant difference in rates of radiation pneumonia and radiation esophagitis in the two groups (P > 0.05) Conclusions Concurrent chemoradiotherapy has better curative effect than sequential chemoradiotherapy.Concurrent chemoradiotherapy is a safe and effective treatment for patients with unresectable stage Ⅲ non-small cell lung cancer.
3.Relationship of congenital urogenital abnormalities and chromosomal abnormalities: a report of 200cases

A total of 200 outpatients with congenital urogenital abnormalities were recruited.Peripheral blood from each patient had mixed lymphocyte culture and chromosome karyotype was analyzed.Among them,22 (11%) cases showed abnormal chromosomal karyotypes.The aberrations included abnormal chromosome number (n =13),abnormal chromosome structure (n =8) and sex reversal syndrome (n =1).Chromosomal aberrations are important causative factors of congenital urogenital abnormalities.

·AIM: To determine the effect of amniotic membrane transplantation(AMT) on the disorders of ocular surface. ·METHODS: From May 2002 to July 2007, twenty patients (20 eyes), who had an previous experienced methods of sorts, including contact lens or tarsorrhaphy, underwent AMT. ·RESULTS: The disorders were healed after the first AMT in 10 eyes (50%) within an average of 20 days after surgery and recurred in 1 eye (10%).·CONCLUSION: AMT is helpful in the treatment of the disorders of ocular surface.
6.Proton magnetic resonance spectroscopy of the thalamus and hypothalamus in patients with first-episode depression.

OBJECTIVETo investigate the presence of abnormal metabolism in the thalamus and hypothalamus in patients with first-episode depression.
METHODSThirty drug-naive patients with first-episode depression and 30 age-matched controls were scanned with proton magnetic resonance spectroscopy ((1)H-MRS) for Naa, Cho, Cr and mI.
RESULTSCompared with the control group, the patients showed significantly reduced mI and mI/Cr of the hypothalamus, reduced mI/Cr of the left thalamus, and lowered Cho, ml, and ml/Cr of the right thalamus (P<0.05).
CONCLUSIONPatients with first-episode depression may have myo-inositol and phosphoric acid metabolism disorder in the thalamus and hypothalamus with malfunction of cellular osmotic pressure adjustment mechanism. Abnormal mI/Cr in the thalamus and hypothalamus may represent an important biochemical change in advanced patients with depression.

7.Effect of new Pulian Ointment () in treating psoriasis of blood-heat syndrome: A randomized controlled trial.

OBJECTIVETo objectively assess the clinical efficacy and safety of a new Pulian Ointment (, NPLO) in treating psoriasis of blood-heat syndrome of Chinese medicine.
METHODSA total of 108 patients with psoriasis of blood-heat syndrome were equally assigned, using a randomizing digital table, to the test group treated externally with NPLO and the control group treated with placebo; the medication was done using a singleblinded method twice a day. Meanwhile, all patients received by oral intake a conventional Chinese decoction for clearing heat and cooling blood; the therapeutic course was 4 weeks for both groups. The therapeutic efficacy, changes in the Psoriasis Area and Severity Index (PASI) score and various aspects of the lesion, including scaly eruption, erythema, infiltration, size, score of itching as well as adverse reactions were observed.
RESULTSThe trial was completed in 100 patients, 51 in the test group and 49 in the control group. The remarkably effective rate was 45.10% and the total effective rate was 84.31% in the test group, which were significantly higher than those in the control group, 12.24% and 51.02%, respectively, showing a significant difference between groups (P<0.01). The test group also showed better effects in the improvement of the PASI score of the lesions and scores on erythema, infiltration, size of lesion as well as itching. No adverse event was found in either group.
CONCLUSIONNPLO is a Chinese remedy for the external treatment of psoriasis of the blood-heat syndrome with a reliable therapeutic efficacy and good safety.

8.Relationship between expression of GYPC and TRIP3 genes and prognosis of acute lymphoblastic leukemia in children.

OBJECTIVETo study the relationship of GYPC and TRIP3 gene expression and the prognosis of acute lymphoblastic leukemia (ALL) in children in order to explore the molecular biological mechanisms of recurrence and remission of ALL.
METHODSThirty-eight newly diagnosed ALL children were enrolled. Of the 38 patients, 31 achieved complete remission (CR) and 12 relapsed. Semi-quantitative RT-PCR was employed to measure blood GYPC and TRIP3 gene expression. Twenty blood samples from normal children were used as controls.
RESULTSBlood GYPC expression in newly diagnosed ALL children was significantly higher than that in the control group (p<0.01) and the CR group (p<0.01). The expression of GYPC gene in the CR group was similar to that in the control group. Other than the control group (p<0.01) and the CR group (p<0.01), the GYPC expression of the relapse group was significantly higher than that in the newly diagnosed group (p<0.01). The CR group showed lower GYPC gene expression than the nonjremission group before treatment (p<0.05). Blood expression of TRIP3 gene in the newly diagnosed and the relapse groups was significantly lower than that in the control group (p<0.05). The CR group had increased TRIP3 gene expression compared with the control group (p<0.01) as well as the newly diagnosed and the relapse groups (p<0.01). Of the 38 newly diagnosed ALL children, the patients with positive TRIP3 expression showed higher remission rate than those with negative TRIP3 (p<0.05). The TRIP3 gene expression before treatment in patients who achieved CR was higher than that in non-remission patients (p<0.05).
CONCLUSIONSA high GYPC gene expression is associated with an unfavorable outcome, in contrast, a high TRIP3 gene expression is associated with a favorable outcome in childhood ALL.

9.Clinical significance of insulin-like growth factor-1 receptor in platinum-based chemotherapy for non-small cell lung cancer.

UNLABELLEDOBJECTIVE; To explore the influence of the expression of insulin-like growth factor-1 receptor(IGF-1R)in end-stage non-small cell lung cancer.
METHODThe expression of IGF-1R was detected in 39 paraffin-embedded chemotherapy-naive non-small cell lung cancer tumor samples with immunohistochemical method,and the relationship between the outcomes of platinum-based chemotherapy and IGF-1R expression was analyzed.
RESULTSIGF-1R expression was detected in 21 cases (53.8%). The IGF-1R expression status shown no correlation with tumor pathological status,tumor differentiation status,history of smoking,as well as smoking index. Better outcomes of platinum-based chemotherapy were observed in patients with negative IGF-1R expression.
CONCLUSIONIGF-1R expression may be used to predict the effectiveness of the first-line platinum-based chemotherapy for end-stage non-small cell lung cancer.

10.Association of synovial cyclic citrullinated peptide expression with Th17/Treg imbalance and synovitis in rheumatoid arthritis patients

Objective To assess the association of synovial cyclic citrullinated peptide(CCP)expression with T helper 17(Th17) cells/Regulatory T cells (Treg) imbalance,the histological and clinical features of synovitis in rheumatoid arthritis (RA).Methods CCP expression in synovial specimens from 39 patients with RA and 35 controls was detected by immunohistochemistry assay(IH) using 6×His tagged anti-CCP single chain fragment V (ScFv) antibodies,which were generated by pHEN2 phagemid recombinant antibodies display system.The frequencies of Th17/Treg cells in the peripheral blood mononuclear cells (PBMCs) were determined by flow cytometry (FCM).Th17/Treg cells associated cytokines were analyzed by enzyme-linked immunosorbent assay (ELISA).The histological scores and clinical features of synovitis were included in the study.Chi-square test and ANOVA were used for statistical analysis.Results ① The prevalences of synovial C CP expression were significantly different between RA group and the control(76.9% and 11.4% respectively,X2=31.9,P<0.01).② The frequencies of Th17 cells,Th17/Treg ratio,Th17 cells associated cytokines as IL-6,IL-17a,IL-23,TNF-α,and the Treg cells associated cytokines TGF-31,the serum and synovial fluid anti-CCP antibodies in the RA patients with synovial CCP positive expression were significantly higher in RA patients with CCP positive than those with CCP negative.Disease activity score DAS28 index and histological features quantified variations of the synovial biopsy specimens (synoviocyte hyperplasia,focal aggregates of lymphocytes,and diffuse infiltrat(e)s of lymphocytes) in RA were higher in synovial CCP positive expression patients than in the negative.Conclusion Synovial CCP expression is strongly associated with the Th17/Treg imbalance and synovitis,which may play a crucial role in the pathogenesis of RA.
