Objective To establish the animal model with stress ulcer(SU) and probe the relationship between the gastric mucosal injury and lasting stress time,the dynamic structural changes of pH of gastric juice.Methods Twenty-four SD young rats were randomly divided into 4 groups:control group,45 min(groupⅠ),90 min(groupⅡ),3 h(groupⅢ)groups under water immersion restraint stress(WRS).The change of gastric mucosal ulcer index(UI),pH of gastric juice were observed.Results Acute gastric mucosal damage was induced by WRS,with the WRS time prolonged,UI increased gradually and pH of gastric juice remarkably decreased in experimental rats.UI was positively rela-ted with stress time(r=0.957 P
5.Promotion of proliferation and migration of Müller cells by RPE cells in a co-culture system

AIM: To investigate the role of retinal pigment epithelium (RPE) in the growth of Müller cells using a co-culture system in vitro . METHODS: Müller cells were cocultured with RPE cells under both normoxic and hypoxic conditions in Transwell chamber culture system. Müller cell proliferation was evaluated by MTT assay. The number of cells which migrate through micropores and stay on the outer bottom side of insert systems were observed and counted. RESULTS: The activities of proliferation and migration of Müller cells when cocultured with RPE cells were significantly higher than those of the Müller cells when cultured alone at all time points under both normoxic and hypoxic conditions. However, for both the coculture and control groups, there is no significant difference between the measurements at 3 and 6 hours. CONCLUSION: Evidence suggests that RPE, when co-cultured with Müller cells, can stimulate migration and proliferation of Müller cells under both hypoxic and normoxic conditions in a time-dependent manner; how-ever, there is no evidence to support the synergetic interaction of RPE and Müller cells co-cultured under hypoxic conditions.
6.Analysis of 35 Reports of Adverse Drug Reactions Induced by Shuxuetong Injection

OBJECTIVE:To analyze the rules and characteristics of adverse drug reactions(ADR)induced by Shuxuetong in-jection,and provide reference for clinical rational drug use. METHODS:ADR cases induced by Shuxuetong injection in Beijing collected by national ADR monitoring center in 2012 were retrieved,and 35 cases of the ADR were statistically analyzed. RE-SULTS:ADR induced by Shuxuetong injection had relatively high incidence in elder over 60 years old(68.57%);ADR might oc-curred after 5 min with off-label use and it mainly occured after 10 min-2 h(40.00%);clinical manifestations were mainly systemic damage and nervous system damage,and all patients were returned to normal after symptomatic treatment except one with unclear outcome. CONCLUSIONS:ADR induced by Shuxuetong injection is related to age and dosage. Clinical medication should be strict-ly in accordance with the instructions,strengthen medication monitoring and reduce incidence of ADR to ensure medication safety.
7.Effects of Müller cells on growth of RPE cells in a co-culture system in hypoxia condition

Background More and more evidences suggest that the interaction of retinal progression of retinal diseases.Objective Present study was to investigate the primarily cultured and digested using explant culture method and trypsas acidic protein(GFAP) staining,S-100 staining and cytokine-18 staining co-cultured under the normoxia and hypoxia using transwell chamber,and the proliferation and migration of cultured RPE cells were examined using MTT and compared with only RPE cells cultured group at 3, 6, 24 and 48 hours.Results Over 90% of primarily cultured RPE cells showed the positive response for S-100.The proliferation and migration of RPE cells were significantly increased in hypoxia condition compared with normoxia condition (P<0.01).In hypoxia group,amount of proliferation and migration of RPE cells in co-culture group were higher than the only RPE cells cultured group(P<0.01).Conclusion Hypoxia appear to aggravate the proliferation and migration of RPE cells under the hypoxia status.
9.Perforin expression on peripheral lymphocytes of patients with systemic lupus erythematosus and the effect of glucocorticosteroid on its expression

Objective To investigate the quantity of perforin (PF) expressed on peripheral lymphocytes of active systemic lupus erythematosus (SLE) patients and its distribution in lymphocytes subsets. The effect of glucocorticosteroid on PF expression is also explored. Methods Phenotype analysis of peripheral lymphocytes was carried out by flow cytometry. SLE patients with SLEDAI≥10 (n=16) and normal controls (n=12) were enrolled into this study. The PF expressed on the iymphocytes of SLE patients with disease flares (n=10) Was studied before and after glucocorticosteroid treatment. Results SLE patients with active disease were characterized by higher proportions of PF-positive lymphocytes and higher perforin density. PF and CD8 double positive T cell proportions was evidently increased. The proportions of PF-positive lymphocytes was correlated to SLEDAI (r=0.673). After a short period of glucocorticosteroid therapy, a reduced expression of PF was observed. Conclusion There is an increased expression of perforin in peripheral lymphocytes from SLE patients with active disease. CD8 positive T cell subsets expand significantly in those patients. In addition, the frequency of PF-positive lymphocytes is correlated with SLEDAI. Finally, glucocorticosteroid inhibits the expression of perforin.
