2.Gene Expression of the Micrococcus luteus Fibrinolytic Enzyme (MLFE) in Bacillus subtilis WB600

MLFE (Micrococcus luteus fibrinolytic enzyme) is a fibrinolytic enzyme produced by Micrococcus luteus ML909 strain. The promoter and signal peptide-coding sequence of ?-amylase gene from Bacillus amyloliquefaciens DC-4 was cloned and fused to the sequence coding for mature peptide of MLFE (Gen-Bank: EU232121), forming the fusion gene called amymlfe. This hybrid gene was inserted into the Escherichia coli-Bacillus subtilis shuttle plasmid vector pSUGV4 and expression plasmid pSU-AmyMLFE was constructed. After transformation with B. subtilis WB600, transformant WB600/pSU-AmyMLFE was obtained and produced clear hydrolyzed zones on fibrin plates. The fibrinolytic activity in supernatants of WB600/pSU-AmyMLFE fermented for 24 hours was tested and found to be 238 UKU/mL. The results of SDS-PAGE analysis showed that there was indeed recombinant protein in supernatants. The Western blotting showed that the molecular weight of the expressed protein was the same as expected. These results indicate that the gene, amymlfe, is successfully expressed in B. subtilis WB600.

MicroRNAs (miRNAs) are a class of highly conserved non-coding small RNA molecules.They regulate gene expression by inhibiting transcription or translation.Ischemic stroke is one of the major diseases of resulting in death and disability worldwide.The final outcome of its pathological process is neuronal death.Neuronal apoptosis is mainly seen in ischemic penumbra,and saving the neurons in penumbra are the key to the treatment of ischemic stroke.This article reviews the roles of miRNAs in the neuronal apoptosis during cerebral ischemia.
4.Neovascularization profile in rat allogenic penetrating keratoplasty

AIM: To observe the neovascularization process with no intervention in rat allogenic penetrating keratoplasty. METHODS: Allogenic penetrating keratoplasties were successfully performed in 34 female SPF SD rats with no intervention after operations. Corneal neovascularization(CNV) process was noted on day 4, 7, 15 and 30 with operating microscope. The vascular area surface was calculated using the formula C/12×3.14×[r2-(r-I)2].RESULTS: CNV was noted in 29 out of 34 rats (85%). Firstly, the new vessels distributed around the cornea like a brush then gradually extended towards the center. The vessels were distorted and massive with branched tails, they continued growing to reticulated veins in peak time then gradually atrophied. The average neovascularization area (SE) on day 4, 7, 15 and 30 was 11.8±3.5mm2, 18.5±4.0mm2,14.4±4.3mm2 and 6.0±1.8mm2 respectively and 12.7±1.9mm2 in total. The average percentage that new vessels accounting the whole cornea area(SE) was 30.8%±8.7%, 65.3%±12.8%, 59.4%±14.5%,36.2%±10.9% and 48.7%±6.4% in total.CONCLUSION: In rat allogenic penetrating keratoplasties without intervention, CNV presented on day 4 and reached the maximum area on day 7. Then the vessels gradually atrophied, about 50% of the maximum area still remained on day 30.
5.Effect of exogenous PGC -1α on VEGF expression in human retinal vascular endothelial cells

AlM: To investigate the effect of exogenous peroxisome-proliferator-activated receptor-γcoactivator-1α ( PGC-1α) on vascular endothelial growth factor ( VEGF) expression in human retinal vascular endothelial cells ( HRVEC) .METHODS:Recombinant PGC-1α protein was added to HRVEC, and no recombinant PGC-1α protein was added to HRVEC as control group. After 24h of incubation, two groups of cells were then placed into a normoxic ( 20%O2 ) or hypoxic ( 1% O2 ) environment for another 16h. The expression of VEGF mRNA and protein were detected by real - time PCR, ELlSA and immunofluorescence cytochemistry.RESULTS: VEGF mRNA and protein levels in the cells were significantly increased by recombinant PGC - 1αprotein both under normoxia and hypoxia conditions as compared with control groups (P<0. 01).CONCLUSlON: PGC-1α can upregulate the expression of VEGF in HRVEC under normoxia and hypoxia conditions.
6.Risk factors of electric bicycle road-traffic injuries: a case-control study

Objective:
To investigate the risk factors of road traffic injuries caused by electric bicycles, so as to provide insights into the prevention and control of road traffic injuries.
Methods:
A case-control study was performed (cases to controls ratio of 1∶2). The riders of electric bicycles with road-traffic injuries in Songjiang District, Shanghai Municipality during the period between June 2020 and May 2021 were included in the case group, and age-, gender- and occupation-matched riders of electric bicycles without road-traffic injuries in the same study area during the same study period served as controls. Participants' demographic data, living habits, health status, awareness of safe riding and riding behaviors were collected using questionnaire surveys, and the risk factors of road-traffic injuries caused by electric bicycles were identified using a multivariable conditional logistic regression model.
Results:
There were 167 participants in the case group, including 122 men (73.05%) and with an age of 38 (21) years, and 334 participants in the control group, including 244 men (73.05%) and with an age of 32 (18) years. Multivariable conditional logistic regression analysis identified sleep duration of less than 8 h (OR=1.760, 95%CI: 1.111-2.786), occasional/frequent anxiety (OR=5.140, 95%CI: 1.067-24.750), unawareness of riding speed limit of 25 km/h (OR=2.352, 95%CI: 1.460-3.788), having been warned or punished for violating traffic rules (OR=2.246, 95%CI: 1.243-4.060), running a red light (OR=1.725, 95%CI: 1.043-2.852), riding speed of ≥ 30 km/h (sometimes, OR=2.409, 95%CI: 1.475-3.933; often, OR=4.711, 95%CI: 2.153-10.309) as risk factors of electric bicycle road-traffic injuries.
Conclusions
Electric bicycle road-traffic injuries is associated with sleep duration, anxiety, awareness of traffic rules, and unsafe riding behaviors. Traffic safety education and law enforcement is required to be reinforced among riders of electric bicycles.
7.Study on working concentration of PDL in primary culture of rat hippocampal neuron

Objective To explore working of concentration of PDL used in primary culture of rat hippocampal neurons.Methods 3 pregnant wistar rats were executed by cervical dislocation, the embryos were taken out and the hippocampal tissue was dissected quickly.Then the tissue was digested by trypsin and planted into dishes with proper concentration(300000/3.5cm vessle) which were coated by different PDL solution in different concentration (0.01,0.05,0.1,0.25,0.5,0.75,1,2 mg/mL).The state of the cultured neurons was observed to determine the most suitable concentration of PDL solution in coating dishes.Neurons activity was observed by CCK-8 Kit.Results Most neurons had adhered in 4 h.Protrusion of neurons began to grow in 1 d.The connection between neurons appeared in 4 d.Neurons matured and the network-connection was set up in 7 d.The best working concentration of PDL is between 0.25 and 0.75 mg/mL, neurons grew well and activity was optimum during this concentration.Conclusion The working concentration of PDL is important for the hippocampal neuron culture and this work is worth being applied.

9.Clinical Observation of Nimesulide Combined with Ibuprofen in the Treatment of Pain

OBJECTIVE:To observe the short-term efficacy and safety of nimesulide combined with ibuprofen in the treatment of pain. METHODS:84 patients with pain were randomly divided into observation group and control group. Control group was oral-ly given 0.3 g Ibuprofen sustained-release capsule,twice a day. Observation group was additionally given 100 mg Nimesulide cap-sule,twice a day. The treatment course for both groups was 7 d. Clinical efficacy,visual analogue (VAS) score,quality of life (QOL) score,coagulation indexes (prothrombin time,thrombin time,fibrinogen,activated partial thromboplastin time) before and after treatment,and incidence of adverse reactions in 2 groups were observed. RESULTS:The total effective rate in observa-tion group was significantly higher than control group,the differences were statistically significant(P<0.05). Before treatment, there were no significant differences in the VAS score and QOL score between 2 groups(P>0.05). After treatment,VAS scores in 2 groups were significantly lower than before,and observation group was lower than control group,QOL scores were significantly higher than before,and observation group was higher than control group,the differences were statistically significant(P<0.05). And there were no significant differences in the coagulation indexes and incidence of adverse reactions between 2 groups(P>0.05). CONCLUSIONS:The short-term efficacy of ibuprofen combined with nimesulide is superior to ibuprofen alone in the treatment of pain,with similar short-term safety.
10.The comparison of two methods of detecting pepsinogen

Objective To investigate colloidal gold immune chromatography (GICA ) and enzyme linked immunosorbent assay (ELISA) detection pepsinogen (PG) consistency of results ,so as to provide experimental evidence for the use of colloidal gold im‐munochromatography assay PG and briefly analysis of its clinical utility .Methods Totally 40 cases in our hospital digested samples and medical centers 60 healthy population sample in Endoscopy Center by endoscopy diagnosed with inflammation of the stomach disease ,respectively ,with colloidal gold immune chromatography and enzyme‐linked immunosorbent assay PG ,colloidal gold immu‐noassay chromatography method to be evaluated ,as the reference ELISA method .The results were compared to the method and bias analysis .Results GICA method with a correlation coefficient ELISA test results were greater than 0 .95 ,indicating a good correla‐tion between the two methods and there is no overall bias;sensitivity and specificity of the two methods were (0 .775 ,0 .85) and (0 .782 ,0 .823) .Conclusion Both GICA and ELISA can be used for screening or diagnosis of stomach illnesses .
