2.Quality standard of Xinfang Biyan Capsule

AIM: To establish a standard for quality control of Xinfang Biyan Capsule(Radix Saponikovae, Radix Angelicae Dahuricae, Radix Scutellariae, etc.). METHODS: TLC and HPLC were used. RESULTS: Flos Magnoliae, Radix Saponikovae, Flos Chrysanthemi, Radix Bupleuri in the capsule could be identified by TLC, and baicalin content in the capsule could be measured by RP-HPLC with a linear relatioship at a range of 0.243 ?g- 2.43 ?g(r= 0.999 8 ). The average recovery was 98.15% and RSD was 0.15% . CONCLUSION: The methods are accurate and can be used for the quality control of Xinfang Biyan Capsules.
3. Study on the predictors of the menopausal symptoms related quality of life in patients with breast cancer undergoing chemotherapy

Objective • To survey the menopausal symptoms related quality of life in patients with breast cancer undergoing chemotherapy and its determinants, which provides theoretical basis on intervention. Methods • In this cross-sectional study, a questionnaire survey was conducted on 484 breast cancer patients undergoing chemotherapy in 9 hospitals nationwide recruited by convenience sampling approach, from July 2016 to December 2017. Descriptive statistics, ANOVA, Spearman correlation and multiple linear regression analyses were preformed to explore the risk factors of the menopausal symptoms related quality of life. Results • The mean score of the Menopause Rating Scale (MRS) was 9.580±6.174 with 75.8% larger than 4, which indicated poor quality of life. One-Way ANOVA results showed the effect of marital status, monthly income, and the change of menstrual status on the MRS score were statistically significant. Spearman correlation results showed that the MRS score was positive correlated with anxiety and depression score and chemotherapy symptoms score, but negative correlated with social support score. Three of the nine variables, chemotherapy symptoms, anxiety, and information and emotional support, were selected by the multiple linear stepwise regression analysis, and could explain 43.0% of total variance (P<0.01). Conclusion • Most breast cancer patients undergoing chemotherapy have poor menopausal symptoms related quality of life. The main determinants were chemotherapy symptoms, anxiety, and information and emotional support. Informational and emotional support should be actively provided and mental health interventions should be strengthened, which could effectively improve their quality of life.

Objective To evaluate the diagnostic value of fluorescent quantitative polymerase chain reaction(PCR) for Mycoplasma pneumoniae (MP) in children with MP pneumonia(MPP).Methods From Jun.2008 to Jan.2009,153 cases hospitalized with pneumonia were enrolled,and 30 cases without respiratory infection were enrolled as control group.Their respiratory secretion (including nasopharyngeal secretion,sputum,bronchialalveolar lavage fluid or pharyngeal swab) samples were collected for fluorescent quantitative PCR for MP.And their single or paired serums were collected for specific MP antibody detection.Results There were 123 cases confirmed with MPP by serology,among whom 114 cases were MP PCR positive.The quantitation of MP DNA was among 1.20?106-3.66?1010 gene copys/L. There were 30 cases with pneumonia negative with MP by the paired serum serology,among whom 2 cases were MP PCR positive,and the quantitation of MP DNA was (1.08-3.02)?107gene copys/L.All cases of control group were MP PCR negative.During the first and second weeks of the MPP onset,the sensitivity of MP-IgM from the first single blood samples were 66.7% and 83.9%,respectively.While the sensitivity and specificity of MP PCR were 92.7% and 93.3%,respectively.From the third week of the disease onset,the sensitivity of MP-IgM from the first single blood samples increased to 90.9%-100%.The clinical manifestations of MPP were nonspecific.Conclusions PCR is superior to serology for early diagnosis on MP infection.Combination of the 2 methods may be helpful to early and accurate diagnosis on MP infection.

In the present study, the specifically knockdown models of P-gp or MRP2 were constructed by using a series of chemically synthesized small interfering RNA (siRNA) in vitro. The expression of P-gp and MRP2 was measured by real-time PCR and Western blot, and the function was evaluated by applying P-gp and MRP2 substrate, rhodamine and methotrexate. The results showed that MRP2 siRNA-3 or P-gp siRNA-2 significantly decreased the mRNA expression of MRP2 or P-gp, the inhibition ratio was 68% or 84%; MRP2 siRNA-3 or P-gp siRNA-2 at a dose of 80 nmol x L(-1) significantly reduced the protein expression of MRP2 or P-gp at 48 h after treatment, the inhibition ratio was 62% or 70%. Meanwhile, other transporters were not influenced by siRNA. When pretreatment with MRP2 siRNA-3 or P-gp siRNA-2, the efflux of methotrexate or rhodamine decreased significantly and the intra-cellular concentration increased. The results suggested that chemically synthesized siRNA could significantly inhibit the expression and function of MRP2 and P-gp, and the model of RNAi in vitro could be used to evaluate the role of efflux transporters in transportation of drugs.

8.Medication rule for treatment of functional dyspepsia: an analysis of traditional Chinese medicine literature based on China National Knowledge Internet.

By retrieving the clinical research literature of treatment functional dyspepsia by traditional Chinese medicine (TCM) from January 2004 to December 2014 based on China National Knowledge Internet (CNKI), we would establish a TCM decoction database for treating functional dyspepsia in this study. One hundred and sixty-four literature were included, involving 159 prescriptions, 377 medicines, in a total of 1 990 herbs. These herbs can be divided into 18 categories according to the effectiveness; and qi-regulating herbs, blood circulation herbs, and antipyretic herbs ranked top three ones according to the frequency of usage of the herbs, whose medicine usage frequency accounted for 51.81%. Usage frequency of 16 herbs was over 30, and Atractylodes, Radix, Poriaranked top three according to the usage frequency. Medicinal properties were divided into 9 kinds according to the frequency statistics, and the top three were warm, flat, and cold. Taste frequency statistics were classifiedinto 9 kinds, and the top three were acrid, sweet, and bitter. In frequency statistics of the meridian tropism of herbs, it was classifiedinto 11 kinds, and the top three were spleen, stomach, lung. The analysis can provide a reference for treatment and study of TCM of functional dyspepsia.

9.Study on mechanism for anti-hyperlipidemia efficacy of rhubarb through assistant analysis systems for acting mechanisms of traditional Chinese medicine.

Rhubarb is a traditional Chinese medicine (TCM), wildly used in treating the disease of hyperlipidemia. However, its components are complicated, so that it is still difficult to clear the specific roles of its various components in blood lipids regulation in. So we decide to systematically study the anti- hyperlipidemia mechanism of rhubarb. We integrated multiple databases, based on entity grammar systems model, constructed molecular interaction network between the chemical constituents of rhubarb and hyperlipidemia. The network includes 231 nodes and 638 edges. Thus we infer the interactions of active targets and disease targets to clarify the anti-hyperlipidemia mechanism. And find that rhubarb can promote excretion of cholesterol; inhibit clotting factors and improve blood circulation; inhibit the release of inflammatory cytokines and maintain fat metabolism balance; inhibit cholesterol and triglyceride synthesis; and other ways to achieve lipid-lowering effect. Thus this study provides reference for novel drug development and component compatibility, and also gives a new way for the systematically study of acting mechanism of traditional Chinese medicine.

10.Cyclic stress promotes the proliferation of rabbit condyle chondrocytes

BACKGROUND:The condyle is one of important areas of lower mandible growth, and has growth remodeling capacity lifetime. The in vivo functional research fails to obtain satisfactory outcome due to the complexity of physiological environment, nondirectiveness of stimulating factor transduction, and difficulty to control experimental conditions. Further studies wil explore the effect of stress on condyle chondrocytes in vitro.
OBJECTIVE:To observe the effect of cyclic stress on the proliferation of rabbit condyle chondrocytes cultured in vitro.
METHODS:Rabbit condyle chondrocytes were separated and cultured in vitro. Passage 3 chondrocytes were subject to cyclic tensile stress (10%surface elongation, 6 cycles/min) for 1, 6, 12, 24 hours. While those cells without stress served as the control group. The cellcycle changes were detected with flow cytometry analysis, and the cellproliferation were analyzed by MTT assay.
RESULTS AND CONCLUSION:The flow cytometry analysis showed that S-phase promoting factor exhibited significant differences at 6 and 12 hours compared with control group, and got the maximum value at 24 hours (P<0.01). MTT assay results showed that the cells proliferation at 6 and 12 hours exhibited significant differences compared with the control group, and got the maximum value at 24 hours (P<0.01). The cyclic stress can obviously promote the proliferation of condyle chondrocytes, and the stimulative effect can be sustained 24 hours.
