1.Research progress of miRNAs regulating TLRs signaling pathway in sepsis

Sepsis refers to the infection caused by systemic inflammatory response syndrome.Sepsis is one of the leading causes of death in critically ill patients in the ICU and its morbidity and mortality are increasing year by year globally.According to statistics,in China,the incidence of sepsis in ICU is 8.68% and the mortality rate can be as high as 44.7%,therefore,severe sepsis in the field of medicine to become one of the problems to be overcome.MicroRNAs (miRNAs) play an important role in the pathogenesis of sepsis through the TLRs / NF-κB signaling pathways,which play an important role in immune regulation.In this review,the progress of miRNAs involved in TLRs signaling pathway in sepsis is reviewed.
2.GluR2 expression in developing rat cochlear nucleus and relationship with development of synapse

Objective To investigate the trend of developmental expression of GluR2,subtype of alpha-amino-3-hydroxy-5-methylisoxazole-4-propionic acid(AMPA) receptor and synaptophysin(SYP) in the rat cochlear nucleus(CN) in different developmental stages,and explore the association of GluR2 expression with the development of synapse. Methods SD rats of 2,3,4,6,8 and 10 weeks old were selected,the expression of GluR2 and SYP in CN was detected with immunofluorescence histochemical method,and the association of them was explored. Results GluR2 expression was observed in all the neurons of CN in each postnatal groups.The expression was relatively weaker in the second and third week,became denser in the fourth week,reached the peak in the sixth week and then sharply decreased to the weakest in the tenth week.The expression of GluR2 was denser at granular cell layer,while weaker at molecular layer and multipolar cell layer in the dorsal CN.SYP expression was detected in all the neurons of CN in each postnatal groups.The expression was weakest in the second week,significantly denser in the fourth week,reached the peak in the sixth week,was then sharply decreased and stably maintained. Conclusion The expressions of GluR2 and SYP in the postnatal rat CN exhit an equally age-dependent tendency.The expression of GluR2 in the CN may be associated with the the maturation and function development of the CN.The different expression and distribution of GluR2 and SYP in the rat CN of different developmental stages may be involved in the development and plasticity of auditory center.

6.The expression levels of three T-type calcium channel receptors in inner ear of C57BL/6J mice with age.

OBJECTIVE:
To explore the expression levels of three T-type calcium channel receptors (α1G; α1H; α1I) in the cochlea and spiral ganglion neurons of C57BL/6J mice with different ages.
METHOD:
Thirty cases of C57BL/6J mice were divided into three groups (6-8 W, 24-26 W, 42-44 W) according to the age. The expressions of three T-type calcium channel receptors were quantified by RT-PCR after hearing thresholds measured by ABR.
RESULT:
Three receptors were detected in the cochlea and spiral ganglion neurons of 6-8 W C57BL/6J mice. The quantitative results showed that the expression levels of α1H and α1I were highest among three receptors in spiral ganglion neurons and in the cochlea respectively. The expression levels of three receptors significantly decreased with age,especially at the age of 4244 W.
CONCLUSION
The expression of T-type calcium channel receptors reduced with age in the inner ear of C57BL/6J mice.

7.Study on pharmacokinetic of vancomycin in normal and endophthalmitis eyes in rabbit

Background Vancomycin has been increasingly recommended for the management of endophthalmitis,but few research report has been published about the pharmacokinetics of intravitreal vancomycin up to now.It is necessary to have an exact method to measure the concentration of vancomyein in animal eyes after intravitreal injection.Objective This study was to observe and compare the phamacokinetical process of vancomycin in serum,vitreous and aqueous humor between normal and infected rabbit eyes.Methods Seventy-two healthy adult rabbits were randomly divided into normal group and infected group and 36 rabbits for each.The animal models of endophthalmitis were established by intravitreal inoculation of 2000 CFU/ml staphylococcus aureus in the right eyes of rabbits in the infected group.Once endophthalmitis developed,0.1 ml vancomycin ( 10 g/L) solution was injected into the vitreous of every rabbit.The peripheral blood,vitreous and aqueous humor samples were respectively collected in 4 rabbits for each group at 0.5,2,4,6,12,24,48,72 and 84 hours after injection for detection of vancomycin concentration by high performance liquid chromatography(HPLC-UV).3p97 software was used to create fit parameters of pharmacokinetics.This experiment followed the Regulations for the Administration of Affair Concerning Experimental Animals by State Science and Technology Commission (Version 1988).Results The accuracy of HPLC fitted the detecting request of biological specimen.The concentration-time data of vancomycin in normal rabbit aqueous humor and vitreous was subject to two-compartment model.The pharmacokinetic parameters were separately as following:Cmax was 50.16 mg/L and 751.42 mg/L,t1/2was 51.04 hours and 53.21 hours.The concentration-time data of vancomycin in infected rabbit aqueous humor and vitreous was subject to one-compartment model.The pharmacokinetic parameters were separately as following:Cmaxwas 24.94 mg/L and 687.66 mg/L,t1/2was 11.42 hours and 12.91 hours.The concentration of vancomycin in serum was much lower and almost undectable.The concentration of vancomycin in vitreous was gradually reduced as the prolong of time after injection in both normal group and infected group,but a obvious decline after increased level was scen in aqueous humor.Compared with normal group,the concentrations of vancomycin in both vitreous and aqueous humor were reduced at various time points(P<0.05,P<0.01 ).Conclusions HPLC is simple,highly sensitive and specific for the pharmacokinetic analysis of vancomycon.These results indicate that pharmacokinetic parameters of vancomycin alter in pathological condition,which is helpful for us to establish the better treatment guidelines for endophthalmitis.

9.Enhancers on the transmembrane transport of chlorogenic acid.

To investigate the influence of the difference enhancers on the transport mechanism of chlorogenic acid (CGA) across Caco-2 cells model, a RP-HPLC method was adopted to detect the concentrations of CGA. At the concentrations of 20 to 80 microg x mL(-1), the difference of absorption rate constants (K(a)) was not statistically significant. At the concentrations of 40 and 20 microg x mL(-1), the ratios of apparent permeability coefficients (P(app)) of the apical to basolateral and the basolateral to apical were 1.14 and 1.18, respectively. With the effect of enhancers K(a) and P(app) increased, the absorption half-life (T1/2) decreased. CGA passed through the Caco-2 cell membrane mainly by passive transport. It showed that monocarboxylic acid transporter (MCT) could be involved in the across membrane transport process of CGA. Borneol had no effect on the cell membrane transport processes. The order of increasing absorption of CGA caused by the enhancers was sodium lauryl sulphate > sodium taurocholate > carbomer.

10.Features of anovulatory infertility patients of gan-yin deficiency syndrome: a primary exploration.

OBJECTIVETo explore the laws of anovulatory infertility patients of Gan-yin deficiency syndrome (GYDS), and to analyze the correlation between GYDS and partial sex hormones and metabolic parameters.
METHODSRecruited were 103 anovulatory infertility patients, including 48 of GYDS and 55 of non-GYDS. At the same time, 20 healthy pluripara at the child-bearing period were recruited as the control group. The body mass index (BMI) and waist to hip ratio (WHR) were measured. Serum levels of luteinizing hormone (LH), follicle stimulating hormone (FSH), testosterone (T), prolactin (PRL), estradiol (E2), sex hormone binding globulin (SHBG), fasting blood glucose (FBG), fasting insulin (FINS), total cholesterol (TC), triglyceride (TG), low density lipoprotein-cholesterol (LDL-C), high density lipoprotein-cholesterol (HDL-C), and leptin were detected. The inter-group difference of the above indices was compared between the two groups.
RESULTSLevels of BMI, FINS, HOMA-IRI, leptin, TC, TG, LDL-C, T, PRL, and LH were higher in the GYDS group and the non-GDS group than in the control group (P < 0.01, P < 0.05), while SHBG was lower in the GYDS group and the non-GYDS group than in the control group (P < 0.01). Only the PRL level was higher in the GYDS group than in the non-GYDS group (P < 0.01).
CONCLUSIONSGan-yin deficiency is a predominant manifestation in anovulatory infertility patients. Partial disorder of some sex hormones and metabolic derangement might be common pathological factors for anovulatory infertility, while increased PRL levels was dominant in GYDS.
