Objective To investigate the pathogenesis of age-related cortex and nuclear cataract.Methods The levels of transforming growth factor-?_2 (TGF-?_2)mRNA,proliferating cell nuclear antigen (PCNA),ratio of Bcl-2/Bax,fibronection (FN),vimentin and the density of lens epithelial cells (LECs) were measured and compared among age-related nuclear cataract,cortex cataract and normal LECs.Results The ratio of TGF-?_2 mRNA expression/?-actin expression,the levels of PCNA protein,Bax and Bcl-2 in age-related nuclear cataract(0.16?0.02,16.01,15.97, 16.97)and in cortex cataract (0.27?0.01,1.98,16.93,1.44) were significantly different from those in normal LECs(0.15+0.02,2.43,16.84,6.12),all P

The research status is reviewed on taxonomic position, ecology, biological active matter and artificial cultivation of Shiraia bambusicola. Following problems are presented: whether there are new species and host specificity of Shiraia bambusicola. Since the artificial cultivation of stroma of Shiraia bambusicola has not been success, Shiraia bambusicola is in situation of emerge of itself and perish of itself so that it is not beneficial to resource protection and continual utilization of this medicinal fungus. It is pointed out that molecular biology of Shiraia bambusicola should be further researched and technology of artificial cultivation of Shiraia bambusicola should be explored in order to exploit and usage of Shiraia bambusicola.
4.Rapid detection of chromosomal aneuploidies by multiplex ligation-dependent probe amplification

Objective To test whether multiplex ligation-dependent probe amplification(MLPA)could be used for the prenatal detection of the most common aneuploidies of chromosomes 13,18,21,X,and Y.Methods 34 cases including 22 blood samples(12 with trisomy 21,1 with monosomy X,one male witll extra Y and 8 healthy persons),4 cord blood samples with Down syndrome and 8 amniotic fluid samples ( 1 with trisomy 21 and 7 normal fetuses)were recruited into this study.All samples were confirmed by karvotype analysis. DNA was extracted from blood and amniotic lysate was incubated with proteinase K.MLPA was used to determine the relative copy numbers.Results The resuhs were available within 48 h and were concordant with karyotype analysis in all but one case of amniotic fluid that was suggested to be triploid sample 69,XXY by MLPA or contaminated by maternal blood.This sample actually was found containing a number of red blood cells after centfifugation in test. In total,the concordance rate with clinical characteristics was 97.1%.The Ratio values of 13,18,21,X in normal samples were approaching 1.0 except chromosome Y having slightly higher variation in relative copy number.The difference of ratio means between the normal and trisomy 21 samples was statistically significant by one-way ANOVA(F=298.906.P=0.000).Conclusion Computer assisted MLPA with high sensitivity is a rapid,simple,automatic and reliable method for detection of common chromosomal aneuploidies.

7.The effects of Nd:YAG laser-aided debracket on the bonding strength of brackets and the temperature of pulp cavity.

OBJECTIVETo study the best parameter of impulse Nd:YAG Laser-aided debracket.
METHODSTo measure the bonding strength of ceramic bracket and pulp cavity temperature after different laser energy. First group was not irradiated by laser, second group was irradiated by laser of power 5 W and irradiating duration 2 s, third group was power 3 W and duration 3 s, and fourth group was power 2 W and duration 5 s.
RESULTSThere were significant differences of bonding strength among the four groups. The bonding strength of ceramic bracket was significantly reduced by laser irradiating. There were significant differences of the pulp cavity temperature increasing among different laser power irradiating groups. The lowest temperature increasing (3.65 degrees C) group was the group of laser power 3 W and irradiating duration 3 s.
CONCLUSIONThe bonding strength of ceramic brackets was statistically reduced after laser irradiation and the best parameter of impulse Nd:YAG Laser-aided debracket is laser power 3 W and irradiating duration 3 s.

8.Mechanism of hepatitis B virus infection of trophoblast cells and hepatitis B virus intrauterine infection

The HBsAg IHC staining positive cells could be observed in 6 placental tissues and 3 fetus' liver tissues,and HBcAg was also positive in 1 case of fetus' liver tissue.After co-incubating the tropho- blastic cells and HBV DNA positive serum in vitro,HBsAg expression and HBV DNA could be detected.Apoptosis of HBV-infected trophoblastic cells increased,which was demonstrated by in vivo and in vitro experiments and the apoptosis of placental cells was correlated with the cord blood HBV DNA level.The results of in vitro experiments showed that the apoptosis of trophoblastic cells increased with the elongation of infection time.After 6 months,1 of 12 newborns was positive for HBsAg,HBeAg and anti-HBc,6 was positive for anti-HBs.Conclusions The mechanism of HBV intra-uterine infection may be that HBV breaches the placental barrier and infects the fetus.The localization and replication of HBV in fetal tissues and organs are probably the important factors of chronic HBV infections in neonates.The apoptosis of trophoblastic cells may be the protective mecha- nism for the placental barrier to block the HBV intra-uterine transmission.
9.NAC decreases insulin resistance induced by FFA in rats

Objective To investigate the changes of peripheral insulin resistance after lipid infusion and the effect of N-acetylcystein(NAC) intervention.Methods Thirty-seven normal male SD rats,eight weeks old,were randomly divided into three groups,FFA group,NS group and NAC group(using into NAC 300 mg/(kg?d) two weeks before infusion).Catheters were implanted into right atrium via the jugular vein and left carotid artery.A technique for a 48-h infusion in unrestrained rats was used for triglyceride and heparin or saline infusion.The infusion period started on day 2 after surgery.48-h after infusion,we determined free fat acid(FFA),nitrotyrosine,malonaldehyde(MDA),reduced glutathione hormone(GSH) level in plasma.The glucose infusion rat(GIR) was measured by hyperinsulinemia euglycemic clamp to evaluated the perpherial insulin resistance.The expressions of IRS-1,IRS-2 gene in muscle were detected by real time PCR.Results(1)The FFA,nitrotyrosine and MDA con-centrations in FFA group were higher than that in NS group,but GSH level in plasma was lower.NAC intervention could reverse these effects.(2)GIR was decreased significantly in FFA group as compared with NS group[(8.34?1.8)mg/(min?kg)] vs[(13.56?1.7)mg/(min?kg)],(P
10.Impact of excessive blood-lead levels on T cell subsets in lead exposed workers.

The aim of this study was to explore the impact of lead poisoning on immunological function of T cell subsets in lead-exposed workers through analysis of clinical cases so as to provide the basis for prevention and treatment of occupational diseases. 70 cases who had excessive level of lead in blood and urine and need treatment by driving out lead were collected as observation group through detection of a large blood and urine samples from 2007 to 2010, meanwhile 60 normal healthy people without lead exposure were enrolled in control group. The blood and urine-lead level and T cell subsets in these two groups were detected and analyzed by JP-1A polarograph and flow cytometry respectively. The results showed that among 70 workers in observation group 47.14% had lower CD3(+)CD4(+) level, 71.43% had lower CD4(+)/CD8(+) ratio, the ratio-inversed cases reached to 41.42%, while 13.33% people had lower CD3(+)CD4(+) level, 25% had CD4(+)/CD8(+) ratio below normal level in control group. The blood-lead concentration and CD4(+)/CD8(+) ratio in observation group were (4.63 ± 1.27) µmol/L and 1.20 ± 0.50 respectively; the blood-lead concentration and CD4(+)/CD8(+) ratio in control group were (0.72 ± 0.35) µmol/L and 1.75 ± 0.62, respectively. It is concluded that the obvious increase of cases with cellular immune dysfunction are observed in occupational workers with excessive blood lead, indicating that the lead can suppress human cellular immune function. Moreover along with increase of blood lead concentration of these workers, more people has abnormal T cell subsets, which shows that the blood lead concentration is associated with immune function of T-cells.
