2.Determination of Phenolic Environmental Estrogens in Food Packing Materials by Nonaqueous Capillary Electrophoresis-Chemiluminescence

Nonaqueous capillary electrophoresis-chemiluminescence (NACE) method was developed for the determination of five phenolic environmental estrogens including bisphenol A (BPA),4-nonylphenol(4-NP) ,4-tert-octylphenol(4-tOP),4-tert-butylphenol(4-tBP) and tetrabromobisphenol A(TBBPA) in food packaging material. Target compounds in soaking solution of food packaging material samples were separated by NACE-CL after derivatization with 4-(4,5-diphenyl-lH-imidazol-2-yl) benzoyl chloride ( DIB-Cl) ,reacted with per-oxyoxalate chemiluminescence system,and then detected through magnifying light signal with photomultiplier. With 17β-E_2 as internal standard,qualitative and quantitative analysis of the target compounds were performed by relative migration time and the ratio of relative chemiluminescence intensity,respectively. Several influence factors on separation with NACE,such as the composition and proportion of organic solvent,the concentration of electrolyte,temperature surrounding of the capillary,the concentration of acetic acid and work voltage,were investigated. And the peroxyoxalate chemiluminescence system was optimized. Under these conditions,4-tBP,BPA,4-OP,4-NP and TBBPA were all separated and showed good linearities in the ranges of 0.0095-3.0 mg/L,0.0087-3.0 mg/L,0.0085 -3.0 mg/L,0.011 -3.0 mg/L,0.009-3.0 mg/L,respectively,with correlation coefficients over 0. 9947. The RSDs of relative migration time and relative chemiluminescence intensity ratio were 0.9% -3.0% and 6.5% - 8.6%. Recoveries for the spiked samples ranged from 86.7% to 98. 8%. Five common food packaging material samples were analyzed. The proposed method is simple and sensitive for the quantitative determination of phenolic environmental estrogens in food packaging material samples.
3.Study on drug resistance genes forβ-lactams in multidrug-resistant Acine-tobacter baumannii

Objective To investigate prevalence of resistance genes for β-lactams in clinically isolated multidrug-resistant Acinetobacter baumannii (MDRAB)in a hospital.Methods 22 clinically isolated MDRAB strains were performed antimicrobial susceptibility testing,resistance genes forβ-lactams (TEM,SHV,CTX-M,PER,DHA, IMP ,VIM ,SIM,OXA-23,OXA-24,and OXA-58)in these strains were detected with polymerase chain reaction. Results The resistant rates of 22 isolates of MDRAB to ceftazidime,cefotaxime,cefepime,gentamycin,amikacin, ciprofloxacin,and compound sulfamethoxazole were all 100.00%;to imipenem,meropenem,and cefoperazone/sul-bactam were 50.00%,40.91 %,and 31 .82% respectively,intermediated rates were 31 .82%,36.36%,and 31 .82% respectively.The carriage rates of OXA-23,TEM,IMP ,and PER were 100.00% (n =22),72.73 %(n=16),54.55% (n = 12),and 18.18% (n =4)respectively.Detection results of SHV,CTX-M,DHA,VIM , SIM,OXA-24,and OXA-58 were all negative.Conclusion Carriage of IMP ,TEM,PER,and OXA-23 resistance genes are the major resistance mechanisms of MDRAB to β-lactamase antimicrobial agents in this hospital.
4.Design and medical application of T-shaped shoes with chuck regulator

Objective To design T-shaped shoes with a chuck regulator to facilitate the orthopedics patient in lower limb immobilization,nursing and rehabilitation training.Methods Each of the shoes was composed of the body of low temperature thermoplasticized plate,a regulator,two cross bars controlled by the regulator and a circular silica gel liner at the heel.The stability of triangles kept the limb involved at the middle position,and prevented the lower limb from inward turning,outward turning or dropping.The shape of the triangles was modified by adjusting the regulator to form an individualized fixation posture for each patient.Results The shoes facilitated clinical nursing,enhanced the patient comfort and decreased the complications.Conclusion The shoes gains advantages in wearing,low cost and repeated use,and thus is worthy promoting practically.
5.Hyperthermia Combined with Chemotherapy for Ovarian Cancer:A Meta-analysis

Objective To assess the effectiveness and safety of hyperthermia combined with chemotherapy(HCT)compared with chemotherapy (CT)for ovarian cancer. Methods Related literatures were searched by two independent investigators from the following electronic databases:PubMed,Cochrane Library,EMbase,VIP,WanFang Data,CNKI and CBM.Then the meta?analysis was performed by using RevMan 5.3 software. Results A total of 10 RCTs involving 546 patients were included. The results of Meta?analysis showed that the HCT group was superior to the CT group in the effective rate,tumor control rate,effective rate of CA125,ascites control rate and PD,with significant differences(P<0.05). There was no significant difference in the improvement rate of life quality and the incidences of myelosuppression,severe myelosuppression,severe nausea and vomiting,severe constipation and diarrhea,liver and renal damage(P>0.05). Conclusion Compared with CT,HCT can significantly increase short?term curative effect,ameliorate the quality of life,and it does not increase the incidence of adverse reactions. HCT is worthy of clinical applica?tion.
6.Low Nanomolar Cucurbitacin-I Induces G2 / M Phase Arrest and Apoptosis in Gastric Cancer Cells and the Underlying Mechanism

Background:Upon inhibition of STAT3 signaling pathway,cucurbitacin-I elicits anticancer effect in various malignancies. However,the anticancer effect and underlying mechanism of cucurbitacin-I in gastric cancer is still elusive. Aims:To explore the effect of low nanomolar cucurbitacin-I on cell proliferation,cell cycle and apoptosis in human gastric cancer cells and the underlying mechanism in vitro. Methods:Human gastric adenocarcinoma cell lines AGS and HGC-27 were treated with cucurbitacin-I at low nanomolar concentration. The anti-proliferative effect of cucurbitacin-I was detected by CCK-8 assay and colony formation assay. Flow cytometry was used to assess the cell cycle and apoptosis. Expressions of cell cycle-related proteins,as well as activation of related pathways such as caspase-3 / PARP apoptotic pathway,STAT3, GADD45α and JNK/ p38 MAPK signaling pathways were determined by Western blotting. Results:Cucurbitacin-I markedly inhibited the growth of gastric cancer cells at low nanomolar concentration by inducing G2 / M phase arrest and apoptosis via a STAT3-independent manner. Furthermore,it was revealed that the anticancer effect of cucurbitacin-I was associated with up-regulation of GADD45α,activation of JNK/ p38 MAPK signaling pathway and the subsequent apoptotic events. Conclusions:The present study provides new insights into the mechanism of anticancer effect of cucurbitacin-I, supporting cucurbitacin-I as an attractive therapeutic drug in gastric cancer.
7.Biological Characteristics of Paecilomyces cicadae LB Strain

The effect of carbon source,nitrogen source,temperature,moisture,pH and light on the fungal growth,conidia production and conidia germination of Paecilomyces cicadae LB were studied.For the fun-gal growth and conidia production,the optimum carbon sources were soluble starch and glucose,while the optimum nitrogen source was peptone.For the fungal growth and spore germination of P.cicadae,the opti-mum temperature was 25?C~27?C,for the conidia production,the optimum temperature was 25?C.Conidia geminated at RH 90%~100%,but did not below 90%.When RH reaches to 100% or the conidia were in wa-ter,the germination rate was the highest.The range of pH for the fungal growth,conidia production and co-nidial germination was 4~10,while the optimum pH for the fungal growth was 6 and 6~7 for the conidia production and conidial germination.The light treatment significantly influenced fungal conidia production.The lethal temperature for the fungal conidia was 55?C remaining 10 minutes.The present results suggest the isolate LB can adapt to nutrition and environment more widely,and has greater potential as biological control factor against of pest.

9.The regulating mechanism of inducible nitric oxide synthase in the hepatic injury of obstructive jaundice

Objective To explore the regulating mechanism of inducible nitric oxide synthase(iNOS) in hepatic injury of obstructive jaundice (OJ) in vivo and in vitro experiments. Methods (1) Rat hepatocytes were isolated by in situ collagenase perfusion and primary culture. Hepatocytes were pretreated with various concentrations of iNOS inhibitor SMT for 20 min. After pretreatment, 50?M GCDC was added for an additional 24hr. Cells were next detected by FCM and TUNEL.(2) Experimental obstructive jaundice (BDL) was induced by double ligation of the bile duct in rats. After BDL for 3d、7d、14d、and 21d, the apoptotic status in liver of all rats were determined with TUNEL, and iNOS protein in liver of OJ was ditermined with immunohistochemistry method. Results (1) SMT decreased GCDC-induced apoptosis in a concentration-dependent manner. (2) The apoptotic rate of liver was related to length of time of OJ. Apoptosis index (AI) was highest from rats with 14d bile duct ligation. The stronger the iNOS expression, the higher was the number of apoptotic cells that was found in OJ. Conclusions iNOS is involved in the regulation and the occurrence and progression of hepatic injury of obstructive jaundice.
10.Clinical features of Crohn's disease at different location

Objective To study the clinical features of Crohn's disease(CD)at different location,I.e.small intestine.colon and ileocolon,to facilitate the knowledge of the disease.Methods Data of 103 patients,hospitalized to our department from 2000 to January 2008,were retrospectively analyzed,including general status,clinical manifestations,laboratory findings and pathological changes.Results The cohort included 70 males and 33 females,with the age when the diagnosis was confirmed ranging from 12 to 70,with a peak at 20-29 yr.The location of the main lesion was at small bowel(L1)in 39(38%),at colon (L2)in 16(16%)and at ileum-colon(L3)48(46%).There was no significant difference between each group,regarding the age of onset or the when the diagnosis was confirmed.The incidence of intestinal obstruction was higher in L1 patients than that in L3(P<0.05).In group L1,12(30.8%)were diagnosed by capsule endoscopy,and 17(44%)were confirmed by colonoscopy.Diagnostic rates of L2 and L3 with reference to clinical manifestations.combined with colonoscopy and pathology were 87.5%and 83.3%,respectivelv.In this cohort,there were 23(22.3%)mild cases,58(56.3%)moderate cases and 22 (21.4%)severe cases,according to simplified CD activity index(CDAI).The rate of severe case in L3 was 59.0%(13/48),which was significantly higher than that in L1(P<0.05).Conclusion In patients with CD,L1 is characterized by delayed diagnosis and need of emergent surgery,while L3 is featured with extensive involvement.severe complications and systemic manifestations.Severe case is common in 13 patient,capsule endoscopy and Colonoscopy are important in early detection of the disease to decrease operation rate and delay the time of first operation.
