2.Antiarrhythmic effect of TJ0711.

To study the antiarrhythmic effect of the newly developed alpha/beta-blocker TJ0711, a variety of animal models of arrhythmia were induced by CaCl2, ouabain and ischemia/reperfusion. Glass microelectrode technique was used to observe action potentials of right ventricular papillary muscle of guinea pig. The onset time of arrhythmia induced by CaCl2 was significantly prolonged by TJ0711 at 0.75, 1.5 and 3 mg x kg(-1) doses. TJ0711 (1.5 and 3 mg x kg(-1)) can significantly shorten the ventricular tachycardia (VT) and ventricular fibrillation (VF) duration, the incidence of VF and mortality were significantly reduced. On ischemia-reperfusion-induced arrhythmic model, TJ0711 (0.25, 0.5, 1 and 2 mg x kg(-1)) can significantly reduce the ventricular premature contraction (PVC), VT, VF incidence, mortality, arrhythmia score with a dose-dependent manner. At the same time, rats serum lactate dehydrogenase (LDH) and creatine kinase (CK) activities decreased significantly by TJ0711 (1 and 2 mg x kg(-1)). Ouabain could cause arrhythmia in guinea pigs, when TJ0711 (0.375, 0.75, 1.5 and 3 mg x kg(-1)) was given, the doses of ouabain inducing a variety of arrhythmia PVC, VT, VF, cardiac arrest (CA) were significantly increased with a dose-dependent manner. In the TJ0711 0.1-30 micromol x L(-1) concentration range, guinea pig right ventricular papillary muscle action potential RP (rest potential), APA (action potential amplitude) and V(max) (maximum velocity of depolarization) were not significantly affected. APD20, APD50 and APD90 had a shortening trend but no statistical difference with the increase of TJ0711 concentration. TJ0711 has antiarrhythmic effect on the sympathetic nerve excitement and myocardial cell high calcium animal arrhythmia model. Myocardial action potential zero phase conduction velocity and resting membrane potential were not inhibited by TJ0711. APD20, APD50 and APD90 were shortened by TJ0711 at high concentration. Its antiarrhythmic action mechanism may be besides the action of blocking beta1 receptor, may also have a strong selective blocking action on alpha1 receptor and reducing intracellular calcium concentration.

3.The correlation between nucleolin and biological behavior of tumor

5.Clinical study on the pulsatility index for Chinese CABG patients

6.Bentall procedure with composite tissue valve conduit

Objective To introduce the clinical experience of Bentall procedure with composite tissue valve conduit. Methods Five patients were diagnosed as severe aortic regurgitation with dilatation of Valsava sinus and sinotubular junction and ascending aorta. Mean age of patients was (55.8?9.4) years (range 47~67). After median sternotomy , cardiopulmonary bypass was established using a single two-stage venous cannula. Temperature was brought to 28℃. After the ascending aorta was occluded, an aortotomy was made and aortic valve was excised. Aorta was removed from sinuses of Valsalva except for generous buttons around coronary ostia. After sizing the aortic annulus, an appropriate-size tissue valve and woven dacron conduit (3 mm greater in diameter than the tissue valve) were chosen to build a composite conduit. It was attached to annulus of aortic valve with pledget-reinforced horizontal mattress stitches to replace the aortic valve and ascending aorta as an en bloc procedure (Bentall procedure). In 2 cases, Coronary artery bypass grafting was performed with greater saphenous vein anastomosed distally to left anterior descending artery and proximately to conduit. Results All cases recovered uneventfully and they were discharged without any postoperative complications. Echocardiography was repeated before discharge. Mean aortic pressure gradient was (18.3?7.6) mmHg. Left ventricular dimension was significantly reduced [(58.8?6.3)mm vs. (43.7?3.1)mm]after operations(P0.05). Follow-up was 2 to 8 months. Conclusion With composite tissue valve conduit, Bentall procedure to replace the aortic valve and ascending aorta as an en bloc procedure is feasible and safe for the elderly patients. A long-term follow-up for the function and structure of tissue valve is needed.
7.Isolation and culture of neural crest stem cells of embryonic mice in vitro

Objective To establish a satisfactory method of isolating,culturing and determining gut neural crest stem cells,which may provide theoretical basis for clinical application.Methods The guts of embryonic mice removed and dissociated were plated into serum-free DMEM/F12 medium.The mitogen-free DMEM/F12 medium supplementd with 10% fetal bovine serum was used to induce differentiation of GNCSCs.Neurospheres and their derivations were determined with immunocytochemical and immunofluorescent staning.Results Neurospheres were generated in the simplified serum-free medium.The staining results showed that enteric neurospheres were GNCSCs and could differentiate into neurons,glial cells and smooth muscle cells by serum-induction.Conclusion GNCSCs have the capacity of self-renewal and proliferation,and give rise to neurons,glial cells and myofibroblasts.
8.Changes of Hormones in the Process of Releasing Dormancy in Thunberg Fritillary(Fritillaria thunbergii)Seeds

By means of HPLC,the changes of endogenous hormones in the process of releasing dormancy in Fritillaria thunbergii Miq. seeds were determined. The results indicated that the content of GA3 had a process of increase while the content of ABA was reduced in general. The content of IAA showed a downward -plateau-upward-downward curve in the process of releasing dormancy. When the dormancy of the seeds were released attemperatures 8~10℃ and 3~5℃,changing pattern of each of the three endogenous hormones were similar,but the duration spanning each change were different.
9.Expressions of Transforming Growth Factor-?1 and Connective Tissue Growth Factor in Viral Myocarditis and Intervention Effect of Valsarta on Them

Objective To explore the expressions of transforming growth factor(TGF)-?1 and connective tissue growth factor(CTGF) in viral myocarditis(VM) and the intervention effect of valsarta on them.Methods Fifty-four male Balb/c mice were randomly divided into 3 groups:control group,treatment group and model group.Model group and treatment group established VM model by intraperitoned injecting 0.1 ml CVB3.They were killed on days 7,14 and 28,respectively.Histological cross sections of heart were stained with hematoxylin-eosin and myocardial histopathplogic scores were counted under optical microscope.The expressions of typeⅠand Ⅲ myocardial collagen,TGF-?1 and CTGF were examined by immunohistochemical method and analyzed by using SPSS 11.5 software.Results Compared with control group,histopathplogic scores,the expressions of type Ⅰand Ⅲ myocardial collagen,TGF-?1 and CTGF increased markedly in model group (Pa
10.EXPERIENCE OF EARLY POSTOPERATIVE TREATMENT AFTER CORONARY ARTERY BYPASS GRAFTING

65mmHg. Sufficient blood products might be indicated when there was excessive oozing. Anticoagulant was not necessary. Two patients died of perioperative myocardial infarction and one of allergic shock to protamine. An incidence of postoperative complications as low as 2 2% (5/225) was obtained. Five patients were found to have various postoperative complications, which were completely cured. All the rest patients were uneventful, and they were discharged after (3 0?2 6) days of stay in the ICU and (8 2?1 5) days after the operation. Therefore, it is our belief that comprehensive and careful postoperative management is essential to achieve excellent results.
